A technique is described in which areas of large Spurr epoxy thick sections are directly popped off of glass slides by inverted Beem capsules filled with polymerized plastic. This technique allows the survey of large regions of tissue with the maintenance of tissue relationships and the selection of specific areas for electron microscopic studies. The brittle hair fibres from a case of osteogenesis imperfecta were studied by scanning and transmission electron microscopy. Normal hair fibres are extremely difficult to infect with T. mentagrophytes. As a tentative assay for biochemical defects of hair fibres, experimental dermatophytic infection of the hair with T. mentagrophytes was performed. The massive growth induced suggested that the experimental fungal infection may indeed indicate a biochemical defect in the hairs from our case of osteogenesis imperfecta.

Two siblings with junctional epidermolysis bullosa are described: both survived beyond parturition. They were treated with the usual therapeutic doses of phenytoin, dapsone, prednisolone, and zinc supplements without effect. Investigation of the skin of one of the patients showed that his fibroblasts, collagen synthesis and collagenase levels were normal. A histochemical study was effected, by light and electron microscopy, on the activity of arylsulphatases A and B in the submandibular and sublingual glands of male and female rabbits, distinguishing, among the latter, between the physiological estrus and anestrus condition. The results obtained demonstrated that the enzymatic activity is more intense in the sublingual gland and that the estrus condition causes an increase in this activity. No correlation was found between the topochemical distribution of the soluble arylsulphatases and that of sulphates.

The authors' own clinical observations have shown that a pressure-lowering effect in secondary glaucomas resistant to therapy can be obtained by photocoagulation of the pars plicata of the ciliary body using an argon laser. Small-intestinal biopsy specimens from five children with gluten-sensitive enteropathy (GSE) were studied by scanning electron microscopy (SEM) before dietary treatment and sequentially after removal of gluten from the the diet. Liposarcoma cells in pleural fluid from two patients with metastatic pleomorphic liposarcoma are described. The major diagnostic feature of such specimens, perceivable by light microscopy, is the presence of solitary, pleomorphic giant cells with cytoplasmic vacuoles. In some cells, transmission electron microscopy revealed numerous cytoplasmic lipidic droplets. With scanning electron microscopy, the cellular surfaces were markedly pleomorphic, with ruffles, blebs and long, thin processes.

Transfixing keratoplasty was performed in a patient seven years after a lens extraction, following the appearance of corneal edema with hypertony and proliferation of a retrocorneal veil. Electron-microscope studies have been carried out on the undescended testicle since 1974. Material examined comprised 25 sections of undescended testicles from boys under five. The studies demonstrated that the ultrasctucture of undescended testes shows pathological changes even in the second year of life, and that these affect all the cellular structures, the seminiferous epithelium, supporting elements, seminal tubules and interstitial tissues. It is therefore concluded that operative treatment of retentio testis should be completed during the second year of life.

Scanning electron microscopic studies were performed on 25 deciduous and permanent teeth from members of 7 kindreds with dominant nonlethal osteogenesis imperfects (OI). Two families had normal teeth on clinical and radiological examination; five families had blue or brown opalescent teeth with specific radiologic findings. The freezing velocity, the most important parameter for the quality of cryofixation of biological objects, was measured in frog liver specimens. One case of herpes zoster keratitis studied after corneal graft is described. Light and electron microscopy investigations show severe pathological changes under the coneal epithelium: Bowman's layer disruption, superficial stromal alterations, sub-epithelial phagocyte proliferation with lipid and mucopolysaccharid vacuolar deposits, abnormal vascularization of the posterior stroma. The authors stress on this interesting ultrastructural study because of very rare cases of the same disease reported in the literature.

A technique is described whereby inclusion-bearing cells identified by light microscopy in stained smears of urinary sediment were reprocessed for examination in the electron microscope. The corneal surface of rabbits and vervet monkeys was studied by means of scanning electron microscopy after the influence of the cationic surfactant, benzalkonium chloride. The concentration of the drug was 0.02% or 0.01% and the exposure time was 2 or 4 min. The corneas treated with the drug had characteristic changes with small holes or more extensive lesions of the superficial cell membranes, and loss of microvilli or microplicae. The damage was considered to be due to the lytic effect of benzalkonium chloride upon the plasma membranes.

Light and electron microscopic examination of portio biopsies from eleven patients with trichomoniasis vaginalis revealed in four patients clusters of cells of T. vaginalis (T. vag.) which were attached to the vaginal mucosa. The minimum gap between adjacent trichomonad cells was the size of gap junctions (2 nm).
