Aldh3a1-mediated detoxification of reactive aldehydes contributes to distinct muscle responses to amyotrophic lateral sclerosis progression.

Different muscles exhibit varied susceptibility to degeneration in Amyotrophic Lateral Sclerosis (ALS), a fatal neuromuscular disorder. Extraocular muscles (EOMs) are particularly resistant to ALS progression, and exploring the underlying molecular nature may offer significant therapeutic value. Reactive aldehyde 4-hydroxynonenal (HNE) is implicated in ALS pathogenesis, and Aldh3a1 is an inactivation-resistant intracellular aldehyde dehydrogenase that detoxifies 4-HNE to protect eyes against UV-induced oxidative stress. We detected prominently higher levels of Aldh3a1 in mouse EOMs compared to

Muscular dystrophy contrasted with denervation: different mechanisms underlying spontaneous fibrillations.

The data reviewed in this paper indicate that spontaneous fibrillations do not involve a cholinergic mechanism since non-depolarizing anticholinesterase drugs such as Mestinon fail to increase spontaneous fibrillations in denervated muscle. Fibrillation potentials are related to the changes in electrical properties of the membrane of denervated muscle fibres which lead to the appearance of spontaneous subthreshold depolarizations, sometimes triggering a propagated potential. Fibrillations seem to appear in cycles and this may depend on the depression of spontaneous depolarization by muscle act

The role of partial denervation of the puborectalis in idiopathic faecal incontinence.

Recent studies have suggested that patients with idiopathic faecal incontinence have neuropathic damage to the external and sphincter and pelvic floor musculature, though no direct assessment of puborectalis function has been made in intact man. Obstetric trauma and chronic straining at stool are thought to be possible causes for damage to the puborectalis nerve supply. In this study the motor unit potential duration in the external sphincter and puborectalis was measured by conventional electromyography in incontinent patients and two groups of continent patients who strained at stool. Our re

Intracellular calcium in canine muscle biopsies.

Intracellular staining for calcium was studied in muscle biopsies from 15 dogs by the alizarin red S (ARS) stain. Rare positive fibres were present in normal muscle and in denervation atrophy. The percentage of positive fibres was slightly increased in polymyositis, dermatomyositis and canine temporal/masseter myositis and markedly increased in progressive muscular dystrophy. Calcium-positive fibres were usually so-called large-dark (hypercontracted) fibres or necrotic fibres, although there was occasional staining of normal and atrophied fibres. These results indicate the probable involvement

[Fasciculation potentials in ALS-significance, and relationship with clinical features].

The Awaji criteria proposed in 2008 re-evaluated electrophysiological findings and emphasized the diagnostic significance of fasciculation potentials (FPs) in amyotrophic lateral sclerosis (ALS). FPs were regarded to be equivalent to denervation potentials (fib-psw) in terms of clinical significance. The implication is that FPs are closely related to the progressive denervation process of muscles in ALS. The characteristic features of FP in ALS are complexity and instability. Complex FPs (CFPs) are thought to originate in distal axonal sprouts, associated with the reinnervation process. These

Targeted lung denervation in sheep: durability of denervation and long-term histologic effects on bronchial wall and peribronchial structures.

BACKGROUND: Targeted lung denervation (TLD), a novel bronchoscopic procedure which attenuates pulmonary nerve input to the lung to reduce the clinical consequences of neural hyperactivity, may be an important emerging treatment for COPD. While procedural safety and impact on clinical outcomes have recently been reported, the mechanism of action has not been reported. We explored the long-term pathologic and histopathologic effects in a sheep model of ablation of bronchial branches of the vagus nerve using a novel dual-cooled radiofrequency ablation catheter. METHODS: Nineteen sheep underwent

Electronic pacing of incapacitated head and neck structures.

Paralysis in the head and neck can affect any motor or mixed cranial nerves and the cervical roots. Most conspicuous deficits, however, involve the larynx and the face. The capacity for denervated striated muscle to undergo reinnervation, and the presence of remaining sources of information, have allowed coordinated rehabilitation of incapacitated cervical neuromuscular systems. The object of further related research should focus on the long-term efficacy of the reinnervated muscle machinery and the potential complexities of electronic integration.

[Hypertrophy of the calf secondary to S1 radiculopathy].

Muscle hypertrophy following denervation is an uncommon phenomenon. Calf hypertrophy in man with S1 radiculopathy is here reported. Electrophysiological examination of hypertrophic muscles revealed a neurogenic pattern with spontaneous bizarre repetitive discharges. Posterior tibial nerve pharmacological block failed to abolish these discharges. During the following three years the hypertrophy decreased while the spontaneous repetitive discharges persisted unchanged. Relationship between radiculopathy, 'neurogenic' hypertrophy and bizarre repetitive discharges is discussed.

[The ultrasound assessment of neuro-muscular apparatus by the radial nerve injuries].

The use of ultrasound (US) scanning to assess the muscular function during the reinnervation is a new concept in medicine. The US signs of muscle denervation were thoroughly described in the article. The US muscle monitoring was performed by the complete and partial radial nerve injury in various follow-up periods. Absolute and relative indications for surgery were determined. The comparative characteristics of structural muscular changes, together with the US test for the assessment of the treatment efficacy and nerval and muscular recovery were suggested.

[Recent advances in the mechanism of insulin-like growth factor-1-mediated exercise-induced improvement of skeletal muscle atrophy].

Skeletal muscle atrophy is characterized by a reduction in both the size and quantity of skeletal muscle fibers, resulting in impaired muscle strength and function. It mainly includes disuse muscle atrophy, aging muscle atrophy, denervated muscle atrophy and muscle atrophy caused by disease etc. As a cost-effective way, exercise has been widely used in the prevention and treatment of skeletal muscle atrophy, but its mechanism for improving skeletal muscle atrophy remains unclear. Recent studies have indicated that insulin-like growth factor 1 (IGF-1) plays an important role in improving muscle

Timing of proteasome inhibition as a pharmacologic strategy for prevention of muscle contractures in neonatal brachial plexus injury.

Neonatal brachial plexus injury (NBPI) causes disabling and incurable contractures, or limb stiffness, which result from proteasome-mediated protein degradation impairing the longitudinal growth of neonatally denervated muscles. We recently showed in a mouse model that the 20S proteasome inhibitor, bortezomib, prevents contractures after NBPI. Given that contractures uniquely follow neonatal denervation, the current study tests the hypothesis that proteasome inhibition during a finite window of neonatal development can prevent long-term contracture development. Following neonatal forelimb dene

Partial wrist denervation versus patient education and self-managed exercise therapy in patients with wrist osteoarthritis: study protocol for a randomized controlled trial.

BACKGROUND: Wrist osteoarthritis (OA) is mainly posttraumatic, affects younger individuals than other types of OA, and can lead to significant disability. The first-line treatment is patient education and exercise, but surgery may be considered if symptoms persist. A mini-invasive surgical approach for wrist OA is partial wrist denervation, which, in theory, alleviates pain by resecting sensory nerve branches while preserving movement. Although denervation has shown mixed results, studies suggest improvements in pain and function. However, the long-term efficacy remains uncertain, with a notab

Myopathy with thick filament (myosin) loss following prolonged paralysis with vecuronium during steroid treatment.

A 20-year-old female hospitalized with status asthmaticus was treated with bronchodilators, antibiotics, and high-dose corticosteroids, and was paralyzed with vecuronium for 10 days to facilitate mechanical ventilation. When this was discontinued, she was found to have a flaccid quadriplegia with areflexia and 4-fold elevation in serum creatine kinase. A muscle biopsy showed extensive loss of thick (myosin) myofilaments, sometimes in core-like distribution, with relative preservation of thin (actin) filaments and Z-discs. Muscle strength returned to normal after 2 months. The pathological lesi

Preliminary report on experimental free autografts of skeletal muscle to the myocardium.

In three greyhounds, free grafts of pronator teres muscle from the foreleg were applied to the left ventricular myocardium, and showed successful gross and microscopic survival of the graft six months later. The free grafts of skeletal muscle were transplanted as complete muscle bellies two weeks after preliminary denervation by median nerve neurectomy, and at the time of suture to the myocardium, the central end of the divided left phrenic nerve was implanted into the graft. The animals were normally active and preserved normal electrocardiograms after grafting. The use of such grafts in clin

[Aplasia of the thenar eminence: a partial and distal form of radial ray dysplasia. 6 cases].

Six cases of thenar muscle aplasia are reported. Aplasia of the abductor pollicis brevis and opponens pollicis muscles was noted in all cases. The flexor pollicis brevis muscle was inconstantly involved and the abductor pollicis muscle was normal. The aplasia was unilateral in five cases and bilateral in one. Bone dysplasia of the scaphoid, first metacarpal and thumb was associated with muscle aplasia. Arterial dysplasia was also observed in four cases. In all cases electrophysiological examination showed that the thenar branch of the median nerve was not involved. Electromyography showed no s

Distinct muscarinic acetylcholine receptor subtypes contribute to stability and growth, but not compensatory plasticity, of neuromuscular synapses.

Muscarinic acetylcholine receptors (mAChRs) modulate synaptic function, but whether they influence synaptic structure remains unknown. At neuromuscular junctions (NMJs), mAChRs have been implicated in compensatory sprouting of axon terminals in paralyzed or denervated muscles. Here we used pharmacological and genetic inhibition and localization studies of mAChR subtypes at mouse NMJs to demonstrate their roles in synaptic stability and growth but not in compensatory sprouting. M(2) mAChRs were present solely in motor neurons, whereas M(1), M(3), and M(5) mAChRs were associated with Schwann cel
