Exertional heat stroke (HS) is a hyperthermic crisis triggered by an excessive accumulation of Ca2+ in skeletal muscle fibers. We demonstrated that exercise leads to the formation of calcium entry units (CEUs), which are intracellular junctions that reduce muscle fatigue by promoting the recovery of extracellular Ca2+ via store-operated Ca2+ entry (SOCE). Here, we tested the hypothesis that exercise-induced assembly of CEUs may increase the risk of HS when physical activity is performed in adverse environmental conditions (high temperature and humidity). Adult mice were: (a) first, divided int…

KEY POINTS: Knee-extensors demonstrate greater fatigue resistance in females compared to males during single-limb and whole-body exercise. For single-limb exercise, the intensity-duration relationship is different between sexes, with females sustaining a greater relative intensity of exercise. This study established the power-duration relationship during cycling, then assessed fatigability during critical power-matched exercise within the heavy and severe intensity domains. When critical power and the curvature constant were expressed relative to maximal ramp test power, no sex difference was

BACKGROUND: A common and disruptive symptom of multiple sclerosis is difficulty in walking. Deficits in ankle proprioception and in plantarflexor muscle function may contribute to these mobility issues. In this study, ankle proprioceptive ability and plantarflexor performance of people with multiple sclerosis (PwMS) were compared to healthy controls to determine whether multiple sclerosis causes impairments in these systems.
METHODS: PwMS (n = 30, median EDSS 4.0, IQR 2) were compared to age- and sex-matched healthy controls (n = 30) across tests of ankle proprioception and plantarflexor musc

BACKGROUND: Respiratory muscle weakness causes inadequate respiration, ineffective coughing, impaired exercise capacity, and autonomic dysfunction. Inspiratory muscle strength training (IMST) may improve these functions; however, its effect in Parkinson's disease (PD) remains unknown.
OBJECTIVE: To investigate the effects of IMST on upper and lower extremity exercise capacity, muscle oxygenation, autonomic function, cough strength, fatigue, and dyspnea in patients with PD.
METHODS: A randomized, controlled, triple-blind study. Twenty patients with PD received IMST at 50% of maximal inspirato

It is important to determine the enabling mechanisms that underlie locomotor performance to explain the evolutionary patterns and ecological success of animals. Our aim was to determine the extent to which calcium (Ca(2+)) handling dynamics modulate the contractile properties of isolated skeletal muscle, and whether the effects of changing Ca(2+) handling dynamics in skeletal muscle are paralleled by changes in whole-animal sprint and sustained swimming performance. Carp (Cyprinus carpio) increased swimming speed by concomitant increases in tail-beat amplitude and frequency. Reducing Ca(2+) re

The effects of chronic hypoxia (CH) on respiratory muscle performance have hardly been investigated, despite clinical relevance. Results from recent studies are indicative of unique adaptive strategies in hypoxic diaphragm. Respiratory muscle tolerance of acute severe hypoxic stress was examined in normoxic and CH diaphragm in the presence and absence of a nitric oxide (NO) synthase inhibitor. We tested the hypothesis that improved tolerance of severe hypoxic stress in CH diaphragm is NO-dependent. Wistar rats were exposed to normoxia (sea-level, n = 6) or CH (ambient pressure = 380 mmHg, n =

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is characterized by disabling fatigue of at least 6 months, in addition to symptoms such as muscle pain and muscle weakness. There is no treatment provides long-term benefits to most patients. Recently, clinical research suggested the involvement of pyruvate dehydrogenase (PDH) in ME/CFS. PDH is a crucial enzyme in the mitochondria matrix that links glycolysis to the tricarboxylic acid cycle and oxidative phosphorylation. However, it is little known whether PDH could be a therapeutic target. The purpose of this study was to establish

OBJECTIVES: Conduction block (CB) from focal neuropathy is often associated with weakness and fatigue in affected muscles. Ulnar neuropathy at the elbow (UNE) provides an excellent model to examine the relationships between electrophysiologically defined CB and quantitative measurement of weakness and fatigue.
METHODS: Eight healthy control subjects (47 ± 14 years) and nine patients (53 ± 3 years) with clinical and electrophysiological features of UNE with CB were studied. All underwent bilateral, ulnar motor nerve conduction studies recording from the first dorsal interosseous muscle as well

This study aimed to systematically review fatigue responses following basketball match-play and during congested match schedules, considering performance, physiological, athlete-reported, and sleep-related outcomes. Relevant articles published until 23 January 2024 were searched using Scopus, Web of Science, and SPORTDiscus. After screening, 44 studies were included in the systematic review. The main findings indicate that, in most cases, vertical jumping and linear sprinting showed significant decrements at match-end (<1 hour post-match) compared to pre-match [small-to-very large effect sizes

Adults with late-onset Pompe disease (aLOPD) are characterized by muscular contractile tissue deterioration. However, their neuromuscular performances are poorly known. We aimed to compare maximal muscle strength, activation, explosive strength and neuromuscular fatigue between aLOPD and controls. We studied 20 aLOPD and 20 matched controls. Isometric maximum voluntary contraction (MVC) torque was obtained for the hip, knee and ankle muscles. The voluntary activation level (VAL) during knee extensor MVC was assessed using interpolated twitch technique. Explosive strength was evaluated for knee

Prolonged intensive care unit (ICU) hospitalization due to acute respiratory distress syndrome results in reduced muscle strength and increased fatigability. This study investigated the central and peripheral neuromuscular mechanisms underlying these impairments in sixty COVID-19 patients up to 12 months post-ICU discharge. Muscle strength (maximal voluntary contraction; MVC), functional exercise capacity (six-minute walk test; 6MWT), and perceived fatigue (Fatigue Severity Scale; FSS) were assessed at each follow-up. High-Density surface electromyography was collected from the Tibialis anteri

Activity-induced muscle pain increases release of interleukin-1β (IL-1β) in muscle macrophages and the development of pain is prevented by blockade of IL-1β. Brain derived neurotrophic factor (BDNF) is released from sensory neurons in response to IL-1β and mediates both inflammatory and neuropathic pain. Thus, we hypothesized that metabolites released during fatiguing muscle contractions activate macrophages to release IL-1β, which subsequently activate sensory neurons to secrete BDNF. To test this hypothesis, we used an animal model of activity-induced pain induced by repeated intramuscular a

Fatigue is an increasingly prominent global health issue. Periplaneta americana glycoprotein (PAG) possesses multiple pharmacological activities including antioxidation, regulation of gut microbiota (GM) and immunity, which are highly consistent with the targets of anti-fatigue treatment. This study aimed to investigate the efficacy and mechanisms of PAG in anti-fatigue. Initially, the antioxidant activity of PAG was evaluated in vitro. Subsequently, a fatigue mouse model was established to evaluate its anti-fatigue effects in vivo. Finally, Spearman correlation analysis was conducted to explo

The influence of inducible heat stress proteins on protecting contracting skeletal muscle against fatigue-induced injury was investigated. A line of transgenic mice overexpressing the inducible form of the 72-kDa heat shock protein (HSP72) in skeletal muscles was used. We examined the relationship between muscle contractility and levels of the constitutive (HSC73) and inducible (HSP72) forms of the 72-kDa heat shock protein in intact, mouse extensor digitorum longus (EDL), soleus (SOL), and the diaphragm (DPH). In all transgenic muscles, HSP72 was expressed at higher levels compared with trans

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is characterized by various disabling symptoms including exercise intolerance and is diagnosed in the absence of a specific cause, making its clinical management challenging. A better understanding of the molecular mechanism underlying this apparent bioenergetic deficiency state may reveal insights for developing targeted treatment strategies. We report that overexpression of Wiskott-Aldrich Syndrome Protein Family Member 3 (WASF3), here identified in a 38-y-old woman suffering from long-standing fatigue and exercise intolerance, can
