Potential roles of metalloprotease mediated ectodomain cleavage in signaling by the endothelial receptor tyrosine kinase Tie-1.

The orphan receptor tyrosine kinase Tie-1 is expressed predominantly in endothelial cells. Expression of this receptor is increased in physiologic angiogenesis and pathologic situations including tumor growth and arteriovenous malformations. Tie-1 is essential for vascular development where it acts in later stages of angiogenesis to suppress endothelial activation and stabilize the newly formed vessel. Stimulation of protein kinase C in endothelial cells results in endoproteolytic cleavage of Tie-1, releasing the extracellular ligand-binding domain of the receptor. We show that this is mediate

Modelling disc degeneration-induced neurovascular invasion on a chip for chronic low back pain therapeutic testing.

Intervertebral disc (IVD) degeneration is the most common musculoskeletal disorder worldwide. Yet, the underlying pathological processes remain largely unclear, primarily due to the lack of suitable in vitro models that accurately replicate the complex interactions of vascular and neural growth, inflammatory cytokine secretion, and immune cell infiltration that contribute to disc degeneration. To address this gap, we developed an affordable microfluidic IVD model using a 5-axis computer numerical control (CNC) micromilling machine with a multi-chamber culture design featuring a central disc zo

Endothelial FGF13 impairs angiogenesis by accelerating oxygen-independent HIF-1α degradation.

Angiogenesis is a tightly regulated process essential for both development and disease, yet the role of fibroblast growth factor 13 (FGF13) in vascular remodeling has remained unclear. Here, we identify FGF13 as a key negative regulator of physiological and pathological angiogenesis. Using endothelial cell-specific knockout and overexpression mouse models, combined with retinal vascularization assays, hindlimb ischemia and myocardial infarction models, and in vitro endothelial cell functional assays, we demonstrate that loss of FGF13 enhances, whereas its overexpression impairs, neovasculariza

[Limbal stem cell deficiency management. A review].

Limbal stem cell deficiency is predominantly caused by severe eye burns resulting in a decreased or a complete ablation of the regenerative potential of these stem cells. The inability to reconstruct the corneal epithelium further leads conjunctivalization of the gimbal-epithelial barrier. These abnormalities collectively result in the progressive opacification of the cornea responsible for blindness that is driven by chronic corneal ulceration and neovascularization. The underlying pathology of the cornea affects the homeostasis of the neighboring conjunctiva, eyelids, and tear film. Therefor

Differential Roles of Angiogenesis in the Induction of Fibrogenesis and the Resolution of Fibrosis in Liver.

Liver fibrosis is a wound healing process that includes inflammation, deposition of extracellular matrix molecules, and pathological neovascularization. Angiogenesis, which is defined by the formation of new blood vessels from pre-existing vessels, is a complex and dynamic process under both physiological and pathological conditions. Although whether angiogenesis can induce or occur in parallel with the progression of hepatic fibrosis has not yet been determined, intrahepatic sinusoidal formation and remodeling are key features of liver fibrosis. Some recent evidence has suggested that experim

Macrophage plasticity and function in cancer and pregnancy.

As the soil of life, the composition and shaping process of the immune microenvironment of the uterus is worth exploring. Macrophages, indispensable constituents of the innate immune system, are essential mediators of inflammation and tissue remodeling as well. Recent insights into the heterogeneity of macrophage subpopulations have renewed interest in their functional diversity in both physiological and pathological settings. Macrophages display remarkable plasticity and switch from one phenotype to another. Intrinsic plasticity enables tissue macrophages to perform a variety of functions in

Overview of the Use of Optical Coherence Tomography Angiography in Neovascular Age-Related Macular Degeneration.

The aim of this review is to present and discuss the use of optical coherence tomography angiography (OCTA) in age-related macular degeneration (AMD). OCTA is a non-invasive imaging procedure that gives a detailed indirect view of physiological and pathological vessels in the retina and choroid membrane. Compared with dye-based imaging, OCTA provides a segmented presentation of the individual vascular layers and plexuses, thus enabling previously unattainable differentiation and classification of pathological vascular changes within or underneath the retina. In particular, OCTA facilitates ear

The role of long noncoding RNAs in ocular angiogenesis and vascular oculopathy.

Long noncoding RNAs (lncRNAs) are RNA transcripts over 200 nucleotides in length that do not code for proteins. Initially considered a genomic mystery, an increasing number of lncRNAs have been shown to have vital roles in physiological and pathological conditions by regulating gene expression through diverse mechanisms depending on their subcellular localization. Dysregulated angiogenesis is responsible for various vascular oculopathies, including diabetic retinopathy, retinopathy of prematurity, age-related macular degeneration, and corneal neovascularization. While anti-VEGF treatment is av

A novel peptide specifically binding to VEGF receptor suppresses angiogenesis in vitro and in vivo.

Vascular endothelial growth factor (VEGF), one of the most important angiogenic factors, plays an essential role in both physiological and pathological angiogenesis through binding to VEGF receptors (VEGFRs). Here we report a novel peptide designated HRHTKQRHTALH (peptide HRH), which was isolated from the Ph.D. -12 phage display library using VEGFR-Fc fusion protein as the bait. This peptide was found to dose-dependently inhibit the proliferation of human umbilical vein endothelial cells stimulated by VEGF. The anti-angiogenesis effect of the HRH peptide was further confirmed in vivo using the

Alternative functions of the complement protein C1q at embryo implantation site.

Complement component C1q is one of the recognition molecules of the complement system which can serve several functions unrelated to complement activation. This molecule is produced at foeto-maternal interface by macrophages as wells as by decidual endothelial cells and invading trophoblast. Foetal trophoblast cells migrating through the decidua in the early stages of pregnancy synthesize and express C1q on their surface, which is actively involved in promoting trophoblast endovascular and interstitial invasion of the decidua. These functions are mediated by two cell surface receptors, gC1qR a

Effects of Animal Venoms and Toxins on Hallmarks of Cancer.

Animal venoms are a cocktail of proteins and peptides, targeting vital physiological processes. Venoms have evolved to assist in the capture and digestion of prey. Key venom components often include neurotoxins, myotoxins, cardiotoxins, hematoxins and catalytic enzymes. The pharmacological activities of venom components have been investigated as a source of potential therapeutic agents. Interestingly, a number of animal toxins display profound anticancer effects. These include toxins purified from snake, bee and scorpion venoms effecting cancer cell proliferation, migration, invasion, apoptoti

Vascular endothelial growth factor expression in the retinal pigment epithelium is essential for choriocapillaris development and visual function.

The choroid in the eye provides vascular support for the retinal pigment epithelium (RPE) and the photoreceptors. Vascular endothelial growth factor (VEGF) derived from the RPE has been implicated in the physiological regulation of the choroidal vasculature, and overexpression of VEGF in this epithelium has been considered an important factor in the pathogenesis of choroidal neovascularization in age-related macular degeneration. Here, we demonstrate that RPE-derived VEGF is essential for choriocapillaris development. Conditional inactivation of VEGF expression in the RPE (in VEGFrpe-/- mice)

One-step coaxial bioprinting of aligned and vascularized 3D skeletal muscle constructs.

Skeletal muscle constitutes approximately 40%-50% of total body weight and plays a vital role in human physiology; however, its regenerative capacity is insufficient in cases of volumetric muscle loss, and effective therapeutic options remain limited. To address this challenge, tissue engineering approaches have focused on constructing functional muscle grafts, with increasing attention on vascularized muscle tissues due to their enhanced integration, regeneration, and reduced fibrosis after transplantation. Nevertheless, most existing strategies rely on complex, multi-step fabrication process

Clinical and histopathological effects of topical pentoxifylline on autonomisation processes in rat dorsal randomised skin flaps.

BACKGROUND: Staged pedicled skin flaps that require pedicle division are frequently employed in reconstructive surgery, rely on their pedicle for vascular supply following transfer until neovascularisation occurs at the recipient site, a phenomenon referred to as autonomisation (pedicle-independent flap viability following neovascularisation). This study sought to investigate the application of topical pentoxifylline to expedite the autonomisation process. METHODS: Caudal-based skin flaps were surgically created in the dorsal region of 60 Wistar rats. On the fifth day, the bases of the flaps

Administration of nebivolol after subcutaneous ovarian autotransplantation improves neoangiogenesis, follicular survival, and growth in mice.

CONTEXT: As ovarian transplantation is performed without blood vessel anastomosis, its main limitation is ischemia. Nitric oxide (NO) has pro-angiogenic and antiapoptotic effects, and nebivolol, a selective β1-blocker with NO-inducing activity, may be a potential drug to improve graft survival. AIM: To evaluate the effect of nebivolol treatment on follicular growth and angiogenesis in murine autotransplanted ovaries. METHODS: Animals were divided into the following two experiments: ovaries were transplanted either subcutaneously or into the renal capsule, and animals were treated with nebivo

Investigating trophoblast invasion and angiogenesis expression changes in a caloric deficient mouse model of fetal growth restriction.

Fetal growth restriction (FGR) is a severe pregnancy complication often caused by placental insufficiency. Proper trophoblast invasion is essential for placental development and function, ensuring adequate nutrient and oxygen supply to the developing fetus. Dysregulation impairs placental perfusion, leading to FGR. This study uses a calorie-restricted mouse model to investigate genes/molecular mechanisms regulating trophoblast invasion across gestational timepoints. Pregnant mice received either a standard or 50 % calorie-restricted diet from E8.5. Placentas and invasion sites were analyzed at

Microenvironment-engineered hydrogels drive osteo-angiogenic coupling to accelerate osteoporotic bone regeneration.

Osteoporotic bone defects repair remians significant clinical challenges, characterized by impaired osteogenic differentiation and dysregulated angiogenesis within a microenvironment of oxidative stress and chronic inflammation. Modulating this pathological microenvironment to create favorable conditions is a pivotal strategy for treating osteoporotic bone defects. In this study, we developed a multifunctional composite hydrogel (SMm) through photo-crosslinking, incorporating Metformin (Met) and magnesium oxide nanoparticles (MgO), to promote osteoporotic bone regeneration. The SMm hydrogel de

Extracellular BRICK1 drives heart repair after myocardial infarction in mice.

Tissue repair after myocardial infarction entails a vigorous angiogenic response that mitigates scarring and worsening of heart function. Angiogenesis in the infarct wound is guided by incompletely defined myeloid cell-endothelial cell interactions. Here, we identify the 75-amino acid microprotein BRICK1 (BRK1) as an indispensable driver of postinfarction angiogenesis in a mouse model of reperfused myocardial infarction. We show that BRK1 is preferentially expressed by myeloid cells and translocates to the extracellular space after myocardial infarction in mice and humans. As a subunit of the

The Role of Endothelial Dysfunction in Fracture Healing: Mechanisms and Potential Effects on Skeletal Repair.

Fracture healing depends on coordinated osteogenesis and restoration of the vascular microenvironment. Endothelial cells support skeletal repair through angiogenesis, tissue perfusion and angiocrine signaling that regulates osteoprogenitor recruitment and differentiation. This narrative review examines endothelial dysfunction (ED) as a potential systemic contributor to impaired fracture healing by integrating evidence from vascular biology, experimental models and clinical studies. ED is characterized by reduced nitric oxide (NO) bioavailability, oxidative stress, inflammation and impaired vas

Coronary endothelial cells undergo venous-enriched disrupted maturation following myocardial infarction.

The coronary microvascular plexus responds to cardiac ischemia through angiogenic neovascularization in the ischemic and border zones to sustain the demand for oxygen and nutrients. Although new blood vessels form, the process of endothelial maturation and extent to which the post-injured microvascular plexus resembles the pre-injured state is unclear. Developmental processes suggest that neovascular endothelial cells must specify toward arterial or venous fates to generate a continuum of heterogenous identities across the microvascular plexus. Therefore, we investigated whether coronary endot
