Isoflurane and surgery aggravate APOE4-dependent lipid dysregulation and neural dysfunction, leading to neurological impairment in male mice.

PURPOSE: Perioperative exposure to the volatile anesthetic isoflurane (ISO) has been associated with cognitive and olfactory deficits and may increase the risk of Alzheimer's disease (AD). Apolipoprotein E4 (APOE4), the strongest genetic risk factor for AD, contributes to disease pathogenesis through disrupted lipid homeostasis. However, whether and how isoflurane interacts with APOE genotype to influence neurological vulnerability remains unclear. METHODS: Young adult, presymptomatic humanized APOE4 and APOE3 knock-in mice underwent laparotomy under 2 h of isoflurane anesthesia. Microglia an

Magnetic Resonance Imaging Confirmed Olfactory Bulb Reduction in Long COVID-19: Literature Review and Case Series.

An altered sense of smell and taste was recognized as one of the most characteristic symptoms of coronavirus infection disease (COVID-19). Despite most patients experiencing a complete functional resolution, there is a 21.3% prevalence of persistent alteration at 12 months after infection. To date, magnetic resonance imaging (MRI) findings in these patients have been variable and not clearly defined. We aimed to clarify radiological alterations of olfactory pathways in patients with long COVID-19 characterized by olfactory dysfunction. A comprehensive review of the English literature was perfo…

Olfaction Is Related to Motor Function in Older Adults.

BACKGROUND: Among older adults, both olfaction and motor function predict future cognitive decline and dementia, suggesting potential shared causal pathways. However, it is not known whether olfactory and motor function are independently related in late life. METHODS: We assessed cross-sectional associations of olfaction with motor and cognitive function, using concurrent data on olfactory function, mobility, balance, fine motor function, manual dexterity, and cognition in 163 Baltimore Longitudinal Study of Aging participants aged 60 and older without common neurological diseases (n = 114 wi

The human brain at stage 16, including the initial evagination of the neurohypophysis.

Thirty-nine sectioned embryos of stage 16 were studied. Up to this stage the amygdaloid body is derived entirely from the medial eminence, which was purely diencephalic in stage 14, but now extends also to the telencephalon. The area of the future olfactory bulb is indicated by the presence of olfactory fibres entering the brain wall; the future olfactory tubercle is characterized by cellular islands. The presence of the hippocampal thickening and various histological features make it possible to outline the main, future cortical areas already at this early stage: archi-, paleo-, and neopalliu

Effects of an ambient odor on brain activations during episodic retrieval of objects.

This study investigated the brain areas involved in episodic memory retrieval of pictures depending on the characteristics of the contextual environment during encoding (i.e. presence or absence of an odor). In the first stage, subjects were presented with a series of 32 pictures. Half of the pictures were presented while the subjects smelled an odor (vanillin). No particular odor was associated with the presentation of the other half of the pictures. Two weeks later, a retrieval task was performed in which the same pictures were presented during an fMRI session but without any odor associatio

Solitary olfactory groove schwannoma.

Intracranial schwannomas that do not involve major cranial nerves in the posterior fossa are uncommon, especially if they are not associated with neurofibromatosis type II (NF-2). Subfrontal olfactory groove schwannomas are extremely rare. We report a cystic schwannoma arising from the olfactory groove in a 38-year-old Caucasian male who presented with headache, vomiting and visual impairment. We briefly review the pertinent literature, stress the distinctive clinical and neuroradiological findings and discuss the surgical planning and theories about the pathogenesis of schwannomas in this unu

Trigeminal event-related potentials in patients with olfactory dysfunction.

OBJECTIVE: There are mutual interactions between the olfactory and trigeminal systems. The purpose of this study was (1) to assess trigeminal sensitivity using chemosensory event-related potentials (CSERPs) in patients with olfactory dysfunction and (2) to evaluate whether trigeminal ERPs were different with regard to the presence or absence of olfactory ERPs. MATERIALS AND METHODS: Prospective study of 60 patients who presented with olfactory dysfunction (postinfectious olfactory loss: n = 19, posttraumatic olfactory loss: n = 28, and olfactory loss due to idiopathic, or toxic causes: n = 13

Noradrenergic neuromodulation in the olfactory bulb modulates odor habituation and spontaneous discrimination.

Noradrenergic projections from the locus coeruleus (LC) project to the olfactory bulb (OB), a cortical structure implicated in odor learning and perceptual differentiation among similar odorants. The authors tested the role of OB noradrenaline (NA) in short-term olfactory memory using an animal model of LC degeneration coupled with intrabulbar infusions of NA. Specifically, the authors lesioned cortical noradrenergic fibers in mice with the noradrenergic neurotoxin N-Ethyl-N-(2-chloroethyl)-2-bromobenzylamine hydrochloride (DSP4) and measured the effects on an olfactory habituation/spontaneous

Differential Modulation of Cadmium and Cobalt on Conspecific Alarm and Predator-Induced Neophobic Responses in Zebrafish (Danio rerio).

The olfactory system is pivotal in mediating conspecific alarm responses and predator avoidance behaviors in zebrafish (Danio rerio), with its impairment posing significant survival risks. This study examined the differential impacts of two environmentally relevant heavy metals, cadmium (Cd) and cobalt (Co), on olfactory function and predator-induced behavioral responses. Adult zebrafish were exposed to sublethal concentrations of Cd (4 days) and Co (4 h), followed by behavioral assays involving exposure to conspecific alarm substance (CAS) and predator-derived chemical cues. Cd exposure signi

Olfactory function evaluated by SPECT.

Few articles on neuroimaging techniques in the study of central and peripheral olfactory pathways are present in the literature. By Single Photon Emission Computed Tomography (SPECT), cortical perfusion increment after sensorial stimulation can be evaluated objectively. In the present research, 10 healthy adults underwent SPECT by CER.TO.96 cerebral tomograph, before and after olfactory stimulation with lavender-water. A variable degree of cortical activation was detected in all patients. Gyrus rectus (+24.5%), orbito-frontal cortex (right +26.6%, left +25.6%), and superior temporal (right +9.…

Role of LncRNA in Trauma Susceptibility and Resilience to Post-Traumatic Stress Disorder (PTSD): A Pilot Study in the African American Population.

PURPOSE: Childhood adversities are associated with the development of post-traumatic stress disorder (PTSD). However, not all individuals exposed to severe trauma develop psychopathology, underscoring the need for a better understanding of the molecular pathophysiology underlying vulnerability to PTSD. Evidence suggests that the peripheral olfactory system, which is accessible in the nose, regulates the structure and function of olfactory regions relevant to stress biology. Long non-coding RNAs (lncRNAs) control transcriptional regulation via epigenetic mechanisms, and since these elements are

Differential projections of the densocellular and intermediate parts of the hyperpallium in the pigeon (Columba livia).

The visual Wulst in birds shows a four-layered structure: apical part of the hyperpallium (HA), interstitial part of HA (IHA), intercalated part of hyperpallium (HI), and densocellular part of hyperpallium (HD). HD also connects with the hippocampus and olfactory system. Because HD is subjacent to HI, the two have been treated as one structure in many studies, and the fiber connections of HD have been examined by afferents and efferents originating outside HD. However, to clarify the difference between these two layers, they need to be treated separately. In the present study, the fiber connec

[Morphologic features of the chemosensory, visual and vestibular pathways of Helix lucorum].

The structure of chemosensory, visual and statocyst pathways of the snail Helix lucorum has been studied by means of five histological methods. The chemoreceptors of the optic tentacle bulb, small and large neurons of tentacle ganglion and bipolar cells of olfactory nerve send their processes to the CNS of the mollusc. In the CNS these processes divide into many clusters of branches. All protocerebral cells and the cluster of neurons of the commissural part of metacerebrum send their processes within the olfactory nerve towards the periphery. The axons of eye cells terminate in the enlargement

Ultrastructural and cytochemical identification of apoptotic cell death accompanying development of the fetal rat olfactory nerve layer.

It has been previously shown that the embryonic olfactory nerve contains, in addition to glial ensheathing cells, a large population of differentiated neurons that migrate from the developing olfactory epithelium, in close association with the olfactory axon fascicles. The purpose of our study was to verify the hypothesis according to which a process of physiological cell death might be involved in the progressive disappearance of these migrating neurons that has been reported during late embryonic stages in several immunocytochemical studies. To do so, we have investigated the development of

Parameter characteristics in intranasal drug delivery: A key to targeting medications to the olfactory airspace.

BACKGROUND: The nose is a viable pathway for topical drug delivery to the olfactory cleft for treatment of obstructive smell loss and nose-to-brain drug delivery. This study investigates how variations in nasal vestibule morphology influence intranasal spray drug transport to the olfactory cleft and olfactory roof/bulb regions. METHODS: The unilateral nasal vestibule morphology in three healthy subjects with healthy normal nasal anatomy was classified as Elongated (Subject DN001), Notched (Subject DN002), and Standard (Subject DN003). Computational fluid and particle dynamics modelling were u

Diet-Induced Obesity Induces Transcriptomic Changes in Neuroimmunometabolic-Related Genes in the Striatum and Olfactory Bulb.

The incidence of obesity has markedly increased globally over the last several decades and is believed to be associated with the easier availability of energy-dense foods, including high-fat foods. The reinforcing hedonic properties of high-fat foods, including olfactory cues, activate reward centers in the brain, motivating eating behavior. Thus, there is a growing interest in the understanding of the genetic changes that occur in the brain that are associated with obesity and eating behavior. This growing interest has paralleled advances in genomic methods that enable transcriptomic-wide ana

Rapid classical conditioning of odor response in a physiological model for olfactory research, the tiger salamander.

In recent years there have been a number of important advances in the understanding of cellular mechanisms related to olfactory function. Hypotheses regarding the complex relationships among odorant structure, physiological activity and behavioral outcome generated by these findings, however, remain largely untested due to a paucity of psychophysical data on stimulus discrimination in the same experimental species. Comparisons between behavioral and physiological responses are essential for elucidating the critical aspects of stimulus coding in sensory systems. We have developed a method for g

Venous saving in olfactory meningioma's surgery.

Bifrontal craniotomy and interhemispherical approach, usually used to remove olfactory groove meningioma, does not allow a view of posterior pole, which is very important in the surgical management of this neoplasm, and, provides for an anatomical sacrifice of frontal bridging veins, not without complications as a result of venous infarction. From 1975 the pterional approach was performed on 20 patients with olfactory meningioma, with good results in 90% of cases. This approach presents several advantages over bifrontal craniotomy and, in order to minimise surgical damage, it avoids planned sa

An Integrated Care Strategy for the Follow-up of Patients With COVID-19-Associated Chemosensory Dysfunction.

The proportion of patients with residual olfactory and gustatory dysfunction after COVID-19 is increasing, and practical health care strategies need to be developed to manage this novel situation in otolaryngology services worldwide. Starting from our experience in a large Italian hospital, we estimated that >1500 people will complain of some form of olfactory and gustatory dysfunction in the future months in our region. We want to share our logistical and clinical integrated pathway that is aimed to screen and refer each patient to the most appropriate level of care in order to optimize resou

The alarmin-ILC2 axis as a candidate mechanism for persistent olfactory dysfunction in allergic rhinitis.

BACKGROUND: Allergic rhinitis (AR) is an underrecognized contributor to olfactory dysfunction (OD). Although smell loss in AR is commonly attributed to conductive obstruction, some patients exhibit persistent or disproportionate hyposmia despite improvement in nasal congestion, suggesting that additional sensorineural mechanisms may exist. HYPOTHESIS: We hypothesize that, in a subset of patients with persistent, moderate-to-severe, type 2-high AR-associated OD without macroscopic olfactory cleft obstruction, allergen-induced epithelial stress in the olfactory cleft may activate an alarmin-ILC
