Prevalence of intestinal parasites in private-household cats in Japan.

The present study is the first national investigation of intestinal parasites in private-household cats in Japan. A total of 942 faecal samples were collected from private-household cats. Giardia species was assessed using an enzyme-linked immunosorbent assay kit and other intestinal parasites were identified microscopically. The overall prevalence of intestinal parasites was 10.1%; two protozoan parasites (Giardia species and Cystoisospora species) and five helminths (Toxocara cati, Toxascaris leonina, Ancylostoma tubaeforme, Taenia species and Spirometra erinacei) were detected. The total pr…

Gastrointestinal parasites in goats from Monte Castelo, Santa Catarina, Brazil.

This study was carried out with the aim of estimating the degree of gastrointestinal helminth infection in goats on the Northern Plateau of Santa Catarina. Twelve young females and 11 adult females were used. Every 28 days, feces samples were taken to quantify the nematode eggs per gram of feces (EPG). Larval culturing was performed on a pool of positive samples from the same group. The fecal egg counts (FECs) ranged from zero to 10,400 EPG in the young group and zero to 7,600 EPG in the adult group. The mean FECs were between 583.3 and 4441.7 in the young group and between 418.2 and 2181.8 in

Separation of parasites from human blood using deterministic lateral displacement.

We present the use of a simple microfluidic technique to separate living parasites from human blood. Parasitic trypanosomatids cause a range of human and animal diseases. African trypanosomes, responsible for human African trypanosomiasis (sleeping sickness), live free in the blood and other tissue fluids. Diagnosis relies on detection and due to their often low numbers against an overwhelming background of predominantly red blood cells it is crucial to separate the parasites from the blood. By modifying the method of deterministic lateral displacement, confining parasites and red blood cells …

Immunity to visceral leishmaniasis using genetically defined live-attenuated parasites.

Leishmaniasis is a protozoan parasitic disease endemic to the tropical and subtropical regions of the world, with three major clinical forms … infection may be a prerequisite for immunological memory. Thus, genetically altered live-attenuated parasites with controlled infectivity could achieve such memory. In this paper, we discuss development and characteristics of genetically altered, live-attenuated Leishmania donovani parasites and their possible use as vaccine candidates against VL. In additi …

Spatial niche competition among helminth parasites in horse's large intestine.

The spatial distribution of large intestinal helminth parasites of 50 horses was studied. Both adult and mucosal larval stages were examined and counted within the ventral colon, dorsal colon and caecum. Thirty-three species of adult parasites were identified and their distribution in the three large intestinal regions reported, together with the localization of cyathostome encysted larvae and of empty mucosal cysts. In order to highlight interspecific interactions, both Spearman pairwise correlation (among all parasites) and multivariable negative binomial regression (among the most abundant …

Overexpression of histone H2A modulates drug susceptibility in Leishmania parasites.

Resistance to antimonials has emerged as a major hurdle to the treatment and control of visceral leishmaniasis (VL), also know as kala-azar (KA), the disease caused by Leishmania donovani, in India where >60% of KA patients are unresponsive to sodium antimony gluconate (SAG) treatment. Determinants of resistance in laboratory strains are partly known, however the mechanism operating in field isolates is not well understood. In microarray-based expression profiling with RNA isolated from field isolates of drug-resistant and -sensitive L. donovani parasites, genes encoding histone 1 (H1), histon…

Parasites in motion: flagellum-driven cell motility in African trypanosomes.

Motility of the sleeping sickness parasite, Trypanosoma brucei, impacts disease transmission and pathogenesis. Trypanosome motility is driven by a flagellum that harbors a canonical 9+2 axoneme, together with trypanosome-specific elaborations. Trypanosome flagellum biology and motility have been the object of intense research over the last two years. These studies have led to the discovery of a novel form of motility, termed social motility, and provided revision of long-standing models for cell propulsion. Recent work has also uncovered novel structural features and motor proteins associated

Laboratory diagnosis of infections due to blood and tissue parasites.

Microscopy remains the cornerstone of the laboratory diagnosis of infections due to blood and tissue parasites. Examination of thick and thin peripheral blood smears stained with Giemsa or other appropriate stains is used for detection and identification of species of Plasmodium, Babesia, Trypanosoma, Brugia, Mansonella, and Wuchereria. Even in the hands of well-trained technologists, diagnosis may be hampered by the sparseness of organisms on the slide and by the subjective nature of differentiating similar-appearing organisms. Microscopy and/or culture of ulcer, bone marrow, tissue aspirate,…

Competition promotes the evolution of host generalists in obligate parasites.

Ecological theory traditionally predicts that interspecific competition selects for an increase in ecological specialization. Specialization, in turn, is often thought to be an evolutionary 'dead end,' with specialist lineages unlikely to evolve into generalist lineages. In host-parasite systems, this specialization can take the form of host specificity, with more specialized parasites using fewer hosts. We tested the hypothesis that specialists are evolutionarily more derived, and whether competition favours specialization, using the ectoparasitic feather lice of doves. Phylogenetic analyses …

Platelets kill intraerythrocytic malarial parasites and mediate survival to infection.

Platelets play a critical role in the pathogenesis of malarial infections by encouraging the sequestration of infected red blood cells within the cerebral vasculature. But platelets also have well-established roles in innate protection against microbial infections. We found that purified human platelets killed Plasmodium falciparum parasites cultured in red blood cells. Inhibition of platelet function by aspirin and other platelet inhibitors abrogated the lethal effect human platelets exert on P. falciparum parasites. Likewise, platelet-deficient and aspirin-treated mice were more susceptible …

Uninfected mosquito bites confer protection against infection with malaria parasites.

Despite decades of research and multiple initiatives, malaria continues to be one of the world's most debilitating infectious diseases. New insights for malaria control and vaccine development will be essential to thwart the staggering worldwide impact of this disease (A. Bjorkman and A. Bhattarai, Acta Trop. 94:163-169, 2005); ultimately successful vaccine strategies will undoubtedly be multifactorial, incorporating multiple antigens and targeting diverse aspects of the malaria parasites' biology (M. F. Good et al., Immunol. Rev. 201:254-267, 2004). Using a murine model of malaria infection, …
