Pituitary adenylate cyclase-activating polypeptide controls stimulus-transcription coupling in the hypothalamic-pituitary-adrenal axis to mediate sustained hormone secretion during stress.

External and internal stimuli that threaten homeostasis trigger coordinated stress responses through activation of specialised neuroendocrine circuits. In mammals, the hypothalamic-pituitary-adrenal (HPA) axis mediates responses to stressors such as restraint, ultimately enhancing adrenocortical hormone secretion. Pituitary adenylate cyclase-activating polypeptide (PACAP) has been implicated in central control of the HPA axis, and we have recently shown PACAP-dependent expression of corticotropin-releasing hormone (CRH) and secretion of corticosterone in response to restraint. We now provide a

Hypothalamic-amygdala gating of caregiving-like prosocial behavior in mice.

Distress sharing within an ingroup instinctively motivates caregiving-like prosocial behavior in social animals and humans, yet how aversive social stimuli paradoxically trigger such behavior remains unclear. Here, we show that stress-state matching in observer mice elicits caregiving-like behaviors (allogrooming and injury-targeted allolicking) toward cagemates in pain, resulting in synchronized social buffering. Importantly, we identify that the excitation of vasopressin receptor 1a-expressing (V1aR-expressing) neurons in the central medial amygdala (CeM) underlies both emotional-state match

Targeting cardiomyocyte-derived extracellular vesicle miR-574-5p protects against cognitive impairment induced by ischemia-reperfusion.

BACKGROUND: Acute myocardial ischemia/reperfusion (MI/R) increases risk for cognitive decline, yet the underlying mechanisms mediating heart-brain communication remain poorly understood. Small extracellular vesicles (sEVs) have emerged as novel long-range signaling mediators and may play a critical role. METHODS: MI/R was induced by transient ligation of the left anterior descending artery. Cognitive performance was assessed using multiple behavioral tests. sEVs derived from cardiomyocytes were labeled to track their distribution and cellular uptake in the brain. Heart and brain tissues were

PVN oxytocin - VTA neurocircuit modulate the emergence of general anesthesia induced by isoflurane in mice.

AIMS: The paraventricular nucleus of the hypothalamus (PVN) plays a pivotal role in regulating consciousness transition, in which oxytocin neurons (OXT) are involved. The ventral tegmental area (VTA) is one of the important projection regions of the PVNOXT, closely related to the transition of anesthesia status. However, whether oxytocin neurons in the PVN and PVNOXT-VTA pathway contribute to isoflurane-induced anesthetic state transitions is unknown. METHODS: Immunostaining and fiber photometry were used to verify the neuronal activity during the isoflurane anesthesia. Chemogenetic or optogen

Vagal control of the brain-esophagus axis ameliorates stress-induced esophageal motility dysfunction in male mice.

Mental stress serves as a major contributor to a spectrum of esophageal motility disorders, including distal esophageal spasm (DES), a condition marked by premature contractile activity of the lower esophageal sphincter (LES). These spasms are often exacerbated by psychiatric conditions such as distress, anxiety, and depression. This study investigates the impact of mental stress on LES contractile function in mice. Chronic unpredictable mild stress (CUMS) was observed to reduce food intake, prolong esophageal emptying time, and increase LESP, along with elevated levels of inflammatory factors

Paraventricular oxytocin neurons attenuate post-ischemic brain injury by suppressing microglia-mediated neuroinflammation.

BACKGROUND: Neuroinflammation is caused by the overactivation of microglia, contributing to secondary brain injury in ischemic stroke. Oxytocin (OXT), a neuropeptide synthesized by neurons in the paraventricular nucleus (PVN) of the hypothalamus, has demonstrated potential in mitigating inflammatory responses across various pathological conditions. However, research on the role of PVNOXT neurons in ischemic stroke is limited, and the modulatory effect of these neurons on neuroinflammation remains unclear. METHODS: Transient middle cerebral artery occlusion (tMCAO) was performed in mice. OXT l

Salt-inducible kinase 1 and CREB-regulated transcription co-activator 1 in the paraventricular nucleus participate in the antidepressant-like mechanism of (2R, 6R)-hydroxynorketamine and (2S, 6S)-hydroxynorketamine in mice.

BACKGROUND: Previous studies have demonstrated that ketamine's metabolic conversion to (2S, 6S; 2R, 6R)-HNK is indispensable for its fast-onset and long-lasting antidepressant-like actions, and moreover, both (2R, 6R)-HNK and (2S, 6S)-HNK exert N-methyl-D-aspartate receptor (NMDAR)-independent antidepressant-like activities. Up to date, many pharmacological targets other than NMDAR have been reported for (2R, 6R)-HNK and (2S, 6S)-HNK. We have previously found that salt-inducible kinase 1 (SIK1) and cAMP response element binding protein (CREB)-regulated transcription co-activator 1 (CRTC1) in t

Psilocybin induces sex- and context-specific recruitment of the stress axis.

Following decades of prohibition, psychedelic drugs have reemerged as promising therapeutics for stress-related conditions, including depression and post-traumatic stress disorder. Still, their impact on stress-related brain regions and the hypothalamic-pituitary-adrenal (HPA) axis remains unclear. This work explores the acute effects of psilocybin on the primary regulators of the HPA axis: corticotropin-releasing hormone neurons in the paraventricular nucleus of the hypothalamus (CRHPVN). Here, using blood plasma measurements and in vivo single-fiber photometry, we demonstrate that psilocybin

Acute Optogenetic Activation of the Subfornical Organ Produces Sympathetically Mediated Increases in Blood Pressure.

INTRODUCTION: The subfornical organ (SFO) is a vital blood pressure-controlling region that elicits blood pressure changes likely via an excitatory (or glutamatergic) projection to the paraventricular nucleus of the hypothalamus (PVN). However, the role of this SFO-PVN pathway in blood pressure control has been poorly defined in the literature. As such, the present study aimed to examine the functional connectivity between the SFO neurons and the PVN and how they intersect to control blood pressure. METHODS: In Lewis rats (n = 10), glutamatergic SFO neurons (SFOglut) were transduced with chan

Regulation of stress susceptibility by chromatin-binding protein PHF6 in the pituitary intermediate lobe.

The pituitary intermediate lobe (IL) is a surprisingly understudied pituitary region. Here, we find that chromatin-binding protein PHF6 is enriched in the pituitary IL and plays a crucial role in regulating stress susceptibility in mice. Conditional knockout of Phf6 in PHF6-positive IL cells (ILPHF6 cells) significantly reduces acute stress-induced anxiety-like behaviors and chronic stress-induced depression-like behaviors by impairing cellular activation and the release of stress-related hormones. Mechanistically, conditional knockout of Phf6 in the pituitary IL cells downregulates the calciu

Hormonal mechanisms in the paraventricular nuclei associated with hyperalgesia in Parkinson's disease model rats.

Pain is a major non-motor symptom of Parkinson's disease (PD). The relationship between hyperalgesia and neuropeptides originating from paraventricular nucleus (PVN) in 6-hydroxydopamine (6-OHDA) rats has already been investigated for oxytocin (OXT), but not yet for arginine vasopressin (AVP) and corticotropin-releasing hormone (CRH). The present study aimed to investigate the alterations in these neuropeptides following nociceptive stimulation in PD model rats and to examine the mechanisms of hyperalgesia. Dopaminergic nigrostriatal lesions were induced by injecting 6-OHDA into the medial for

Aging leads to sex-dependent effects on pair bonding and increased number of oxytocin-producing neurons in monogamous prairie voles.

Pair bonds powerfully modulate health, which becomes particularly important when facing the detrimental effects of aging. To examine the impact of aging on relationship formation and response to loss, we examined behavior in naive 6-, 12-, and 18-month male and female prairie voles, a monogamous species that forms mating-based pair bonds. We found that older males (18-months) bonded quicker than younger voles, while similarly aged female voles increased partner directed affiliative behaviors. Supporting sex differences in bonding behaviors, we found that males were more likely to sample both p

Regulation of Thyroid Hormone Levels by Hypothalamic Thyrotropin-Releasing Hormone Neurons.

Background: Thyrotropin-releasing hormone (TRH) neurons in the paraventricular nucleus of the hypothalamus (PVN) have been identified as direct regulators of thyrotropin (TSH) and thyroid hormone (TH) levels. They play a significant role in context of negative feedback by TH at the level of TRH gene expression and during fasting when TH levels fall due, in part, to suppression of TRH gene expression. Methods: To test these functions directly for the first time, we used a chemogenetic approach and activated PVN TRH neurons in both fed and fasted mice. Next, to demonstrate the signals that regul

Afferent innervation of the ischemic kidney contributes to renal dysfunction in renovascular hypertensive rats.

The ablation of renal nerves, by destroying both the sympathetic and afferent fibers, has been shown to be effective in lowering blood pressure in resistant hypertensive patients. However, experimental studies have reported that the removal of sympathetic fibers may lead to side effects, such as the impairment of compensatory cardiorenal responses during a hemodynamic challenge. In the present study, we evaluated the effects of the selective removal of renal afferent fibers on arterial hypertension, renal sympathetic nerve activity, and renal changes in a model of renovascular hypertension. The Paraventricular Nucleus of the Hypothalamus: Development, Function, and Human Diseases.

The paraventricular nucleus of the hypothalamus (PVH), located in the ventral diencephalon adjacent to the third ventricle, is a highly conserved brain region present in species from zebrafish to humans. The PVH is composed of three main types of neurons, magnocellular, parvocellular, and long-projecting neurons, which play imperative roles in the regulation of energy balance and various endocrinological activities. In this review, we focus mainly on recent findings about the early development of the hypothalamus and the PVH, the functions of the PVH in the modulation of energy homeostasis and

Inhibition by lipopolysaccharide of naloxone-induced luteinising hormone secretion is accompanied by increases in corticotropin-releasing factor immunoreactivity in hypothalamic paraventricular neurones in female rats.

We have recently reported that lipopolysaccharide (LPS), a bacterial endotoxin, inhibits steroid-induced as well as naloxone-induced luteinising hormone (LH) secretion in ovariectomised oestrogen-primed rats. In the present study, we examined whether corticotropin-releasing factor (CRF) may be involved in the LPS-induced inhibition of LH secretion. Unanaesthetised rats were treated with an intravenous (i.v.) injection of LPS (10 microg) or saline, followed by an i.v. injection of naloxone (20 mg/kg). After sequential blood samples were collected for determination of serum LH concentrations, th

Pineal and habenula calcification in schizophrenia.

Animal data indicate that melatonin secretion is stimulated by the paraventricular nucleus (PVN) of the hypothalamus and that lesions of the PVN mimic the endocrine effects of pinealectomy. Since the PVN lies adjacent to the third ventricle, I propose that periventricular damage, which is found in schizophrenia and may account for the third ventricular dilatation seen on computed tomographic (CT), may disrupt PVN-pineal interactions and ultimately enhance the process of pineal calcification (PC). To investigate this hypothesis, I conducted CT study on the relationship of PC size to third ventr

Paraventricular nucleus region controls pituitary-adrenal function in Brattleboro rats.

To study the role of the paraventricular nucleus and of neurohypophysial hormones in the control of ACTH secretion, the paraventricular nuclei (PV) of Brattleboro diabetes insipidus rats (DI) were lesioned (L) with a knife; sham-lesioned DI (S) served as controls. Four days later, the rats were stressed by ether inhalation, and blood samples were taken during and 30-40 min after stress for the determination of corticosterone. The median eminence (ME) and neural lobe (NL) were homogenized in 50 microliters of 0.1 N HCl and frozen pending bioassay of corticotropin-releasing factor (CRF). PV lesi

The hypothalamus: anatomy and functions.

The neuroanatomy of the human hypothalamus is reviewed with special interest focused on its neuroendocrine role. The magnocellular neurons in the supraoptic and paraventricular nuclei are the site of synthesis of the nonapeptides antidiuretic hormone and oxytocin and their carriers, the neurophysins. They are in close relation with the posterior lobe of the pituitary which contains their axonal neurosecretory endings. The parvocellular neurons are scattered around the third ventricle, from the preoptic area towards the infundibulum. They control the adenohypophysis by the releasing hormones fo

G protein-coupled receptor 75 mediates depression-like behaviors and HPA axis hyperactivity through an excitatory arcuate-paraventricular nucleus circuit.

Metabolic dysregulation is a core pathological feature of and a major risk factor for depression. In this study, the role of G protein-coupled receptor 75 (GPR75), a brain-enriched metabolic sensor, in the pathogenesis of depression was explored and its potential role as a therapeutic target was evaluated. In chronic social defeat stress (CSDS) models, GPR75 expression was specifically upregulated in the arcuate nucleus glutamatergic (ARCVglut2) neurons of susceptible mice, a finding corroborated by analyses of postmortem brains from patients with depression. Conditional (GPR75-cKO) or local (
