The perception of damage-associated molecular patterns and phytocytokines by pattern recognition receptors activates danger-triggered immuni … eption transiently enhances these associations. NIK1, NIK3 and NIK6 undergo robust ligand-dependent phosphorylation that requires PEPR signalling, whereas PEPR1 and BAK1 can each phosphorylate NIKs. NIKs are dispensable for Pep2-induced PEPR-SERK association but are required for full phosphorylation output from the receptor complex, for efficient recruitment of B …

Ammonium (NH 4 +) uptake and transport in plants are mediated by ammonium transporters (AMTs), which are tightly regulated by phosphorylation to prevent excess ammonium influx and toxic accumulation. However, the mechanism of phosphorylation-mediated inactivation is unique and remains unclear. Here, we report Cryo-EM structures of the rice ( Oryza sativa) ammonium transporter OsAMT1;1 in its wild-type and phosphomimetic variant T452D states. OsAMT1;1 forms a stable homotrimer, with conserved motifs affirming a shared transport mechanism within the AMT/MEP/Rh superfamily. Our structural analysi…

Calpain-3 (CAPN3) is a Ca2+-dependent cysteine protease predominantly expressed in skeletal muscles. Mutations in CAPN3 are associated with limb girdle muscular dystrophy R1 (LGMDR1), an autosomal recessive muscular dystrophy. CAPN3 is uniquely activated by rapid autolytic processing; however, the mechanisms regulating this activation are poorly understood. Here we show that phosphorylation regulates CAPN3 autolytic processing in living cells. We found that ouabain, a cardiac glycoside, activated ERK, which induced the phosphorylation of CAPN3 at S19 located within its N-terminal region. This …

The regulation of replication fork (RF) speed is essential for faithful genome duplication and the maintenance of genome stability, and its … nditions. We then discuss how anticancer agents targeting DNA replication modulate RF speed through phosphorylation-dependent signaling pathways. Finally, we examine emerging roles of PARylation and UFMylation as rapid, replisome-centered regulatory mechanisms that fine-tune fork progression and stability, and we highlight their clinical implications, including a …

BACKGROUND: Palmitoyl transferases ZDHHCs and its mediated protein palmitoylation in several malignant tumors have been reported. However, t … t Cys577. More specifically, ZDHHC9 mediated STAT1 conversion of Cys577 S-palmitoylation and Tyr701 phosphorylation via the JAK1-STAT1 signaling pathway. Notably, palmitoylation of STAT1 at Cys577 is a key mediator of ZDHHC9-mediated GC progression, relying on nuclear pSTAT1-Tyr701 and its transcription of downstream genes.
CONCLUSIONS: Taken together, ZDHHC9 pr …

At the molecular level, ULK1 catalyzed Bcl-2 associated X protein (Bax) phosphorylation at Ser184, creating a recognition motif for Parkin-mediated ubiquitination and proteasomal targeting. Targeting mitochondrial oxidative phosphorylation (OxPhos) has become a recurring strategy in the treatment of cancer, particularly in acute myeloid leukemia (AML). Early reports suggested that leukemic blasts, and especially leukemia stem cells, depend disproportionately on mitochondrial respiration, implying a therapeutic window for systemic inhibition of the electron transport system (ETS) and OxPhos. Yet, the clinical record of broad OxPhos inhibition has been disappointing. In the present review, we argue that the pivotal question is not whether mitochondria matter for cancer, but whether…

Dynamin-related protein 1 (Drp1) is an abundant platelet protein best known for its function in mitochondrial fission. However, little is known about how Drp1 is controlled during platelet activation. While evaluating signaling pathways leading to phosphorylation of Drp1 in platelets, we identified a phosphorylation network wherein activation of G protein-coupled receptors or immunoreceptor tyrosine-based activation motif (ITAM)/hemITAM receptors resulted in phosphorylation of Ser616-Drp1 via p38. These signaling mechanisms were reinforced by ADP- and thromboxane A2 (TxA2)-mediated amplificati…

Increased OXPHOS boosts ROS production, which further promotes NRF2 nuclear translocation, activating the transcription of DNA repair protein UBE2T. This enhanced DNA damage repair ability enables prostate cancer cells with high AKR1C3 exp …

The centriole duplication cycle must be tightly controlled and coordinated with the chromosome cycle. Aberrations in centriole biogenesis can cause developmental disorders, ciliopathies and cancer, yet the molecular determinants controlling centriole numbers and the link between the two cycles remain poorly characterized. Here, we demonstrate that McIdas, previously implicated in cell cycle regulation and multiciliogenesis, plays a critical role in maintaining proper centriole numbers. McIdas localizes to centrioles, where it exhibits dynamic localization throughout the cell cycle, dependent u

Metabolic reprogramming is a hallmark of cancer, enabling tumor cells to meet their increased biosynthetic and energetic demands. Although cells possess the capacity for de novo serine biosynthesis, most transformed cancer cells preferentially rely on exogenous serine uptake to sustain their growth, yet the regulatory mechanisms driving this metabolic dependency remain poorly understood. Here, we uncover a mechanism by which Polo-like kinase 1 (PLK1), frequently overexpressed in prostate cancer, orchestrates a metabolic shift in serine and sphingolipid metabolism through phosphorylation of pho…

In silico analysis of different ABCB-type proteins revealed structural similarities within the linker domain, including the presence of [R/K-X-S/T] and [R/K-X-X-S/T] motifs, which are potential phosphorylation sites for kinases of the AGCVIII superfamily. Based on these findings, a phosphorylation-based regulatory mechanism is proposed for ABCB alkaloid transporters. Plants avoid self-toxicity by compartmentalizing secondary metabolites in specialized cells. Even when this mechanism keeps them away from sensitive metabolic processes, it also could restrict them from reaching the different loca…

BACKGROUND: Although mitochondrial dysfunction is an established hallmark of ventricular remodeling, the molecular mechanisms governing this process remain incompletely characterized. This study systematically investigates the regulatory role of Src homology 2 domain-containing protein tyrosine phosphatase 2 (SHP2) in AMPK-mediated mitochondrial dysfunction and its pathological consequences in ventricular remodeling.
METHODS: Ventricular remodeling was induced by angiotensin II (Ang II) or isoproterenol (ISO) in vivo and in vitro, with assessment of hypertrophy and fibrosis using echocardiogr

Protein kinases integrate cellular signals through complex phosphorylation cascades, yet resolving how chemical perturbations trigger and modulate these cascades in therapeutic targets remains a major challenge. Here, we dissect adenosine 5'-monophosphate-activated protein kinase (AMPK) proteoforms during activation through controlled biochemical reactions with a hybrid mass spectrometry (MS) approach integrating bottom-up MS for site-specific kinetics with top-down proteoform characterization. We reveal that AMPK phosphorylation proceeds through hierarchical cascades rather than binary switch…

Parkinson's disease (PD) is characterized by the progressive accumulation and prion-like spread of pathological α-synuclein (α-syn). Increasing evidence suggests that α-syn pathology may originate in the gastrointestinal tract before spreading to the central nervous system; however, the intestinal mechanisms that initiate pathological α-syn phosphorylation and intercellular transfer remain poorly understood. Here, we investigated the role of Toll-like receptor 2 (TLR2) in regulating α-syn phosphorylation using intestinal epithelial cell model. We found that pSer129-positive α-syn immunoreactiv…
