The m6A Reader MhYTP2 Regulates MdERF54 mRNA Stability and Contributes to Hypoxia Tolerance in Apple (Malus domestica).

Hypoxia is one of the main challenges in apple (Malus domestica) cultivation. However, breeding hypoxia-tolerant cultivars demands a thorough understanding of the responses of apple trees to low oxygen supply. Studies have indicated that N6-methyladenosine (m6A) reader regulates plant stress response by binding to their corresponding mRNA targets with m6A modification. The present study investigated the function and mechanism of apple m6A reader MhYTP2 under hypoxia stress. Here, we found that the overexpression of MhYTP2 improved hypoxia resistance in apple. Previous RNA immunoprecipitation s

CaWRKY20 Negatively Regulates Plant Resistance to Colletotrichum scovillei in Pepper.

Chili anthracnose, a fungal disease caused by Colletotrichum scovillei, is among the most devastating diseases affecting pepper (Capsicum annuum L.). Although WRKY transcription factors play important roles in plant immunity, it is unknown how WRKY gene family members contribute to pepper plant resistance to C. scovillei. Here, CaWRKY20 was found to negatively regulate pepper resistance to C. scovillei, which was demonstrated by virus-induced gene silencing and transient overexpression in pepper. Moreover, overexpression of CaWRKY20 enhanced susceptibility to C. scovillei in tomato. Additional

Grain yield and correlated traits of bread wheat lines: Implications for yield improvement.

Global wheat yields are suffering due to differences in regional climatic conditions and soil fertility. Plant breeders are continuously working to improve the yield per unit area of wheat crop through selecting superior lines as parents. The screening and field evaluation of available lines allow the selection of superior ones and subsequently improved varieties. Therefore, heritable distinctions among 33 bread wheat lines for yield and related attributes were assessed under field conditions. The experiment included thirty lines and three check varieties. Data relating to different plant char

Leaf transpiration plays a role in phosphorus acquisition among a large set of chickpea genotypes.

Low availability of inorganic phosphorus (P) is considered a major constraint for crop productivity worldwide. A unique set of 266 chickpea (Cicer arietinum L.) genotypes, originating from 29 countries and with diverse genetic background, were used to study P-use efficiency. Plants were grown in pots containing sterilized river sand supplied with P at a rate of 10 μg P g-1 soil as FePO4 , a poorly soluble form of P. The results showed large genotypic variation in plant growth, shoot P content, physiological P-use efficiency, and P-utilization efficiency in response to low P supply. Further inv

Genome-wide association study using specific-locus amplified fragment sequencing identifies new genes influencing nitrogen use efficiency in rice landraces.

Nitrogen is essential for crop production. It is a critical macronutrient for plant growth and development. However, excessive application of nitrogen fertilizer is not only a waste of resources but also pollutes the environment. An effective approach to solving this problem is to breed rice varieties with high nitrogen use efficiency (NUE). In this study, we performed a genome-wide association study (GWAS) on 419 rice landraces using 208,993 single nucleotide polymorphisms (SNPs). With the mixed linear model (MLM) in the Tassel software, we identified 834 SNPs associated with root surface are

Adult-plant resistance to leaf scald and net form net blotch in food barley genotypes at a hot spot location in Ethiopia.

Globally, the fungal pathogens Rhynchosporium graminicola and Pyrenophora teres f. teres produce foliar diseases that significantly reduce barley yield. These diseases are known as leaf scald and net form net blotch, respectively. One hundred food barley genotypes in reaction to the diseases were assessed in Ethiopia's natural environment. Since Ethiopia is a secondary center of genetic diversity in barley and consequently its pathogens, this assessment is certainly of interest in identifying new sources of resistance and using the identified genotypes in breeding. In addition, effect of the d

Genome sequence of Jatropha curcas L., a non-edible biodiesel plant, provides a resource to improve seed-related traits.

Jatropha curcas (physic nut), a non-edible oilseed crop, represents one of the most promising alternative energy sources due to its high seed oil content, rapid growth and adaptability to various environments. We report ~339 Mbp draft whole genome sequence of J. curcas var. Chai Nat using both the PacBio and Illumina sequencing platforms. We identified and categorized differentially expressed genes related to biosynthesis of lipid and toxic compound among four stages of seed development. Triacylglycerol (TAG), the major component of seed storage oil, is mainly synthesized by phospholipid:diacy

Transcriptome profiling of Gossypium barbadense inoculated with Verticillium dahliae provides a resource for cotton improvement.

BACKGROUND: Verticillium wilt, caused by the fungal pathogen Verticillium dahliae, is the most severe disease in cotton (Gossypium spp.), causing great lint losses worldwide. Disease management could be achieved in the field if genetically improved, resistant plants were used. However, the interaction between V. dahliae and cotton is a complicated process, and its molecular mechanism remains obscure. To understand better the defense response to this pathogen as a means for obtaining more tolerant cultivars, we monitored the transcriptome profiles of roots from resistant plants of G. barbadense

Prevalence of Toxoplasma gondii in Galapagos birds: Inference of risk factors associated with diet.

Toxoplasma gondii is a zoonotic intracellular parasite of particular concern in the conservation of wildlife due to its ability to infect all homeotherms and potentially cause acute fatal disease in naive species. In the Galapagos (Ecuador), an archipelago composed of more than a hundred islets and islands, the presence of T. gondii can be attributed to human-introduced domestic cats, but little is known about its transmission in wildlife populations. We compared the prevalence of antibodies against T. gondii in sympatric Galapagos wild bird species that differ in diet and contact with oocyst-

Characterization of grain carotenoids in global sorghum germplasm to guide genomics-assisted breeding strategies.

BACKGROUND: Crop biofortification is a successful strategy to ameliorate Vitamin A deficiency. Sorghum is a good candidate for vitamin A biofortification, as it is a staple food in regions with high prevalence of vitamin A deficiency. β-carotene-the main provitamin A carotenoid-is below the target concentration in sorghum grain, therefore biofortification breeding is required. Previous studies found evidence that sorghum carotenoid variation is oligogenic, suggesting that marker-assisted selection can be an appropriate biofortification method. However, we hypothesize that sorghum carotenoids h

Cloprostenol enhances sexual behaviour and semen quality in growing lambs more effectively than Dinoprost.

Analogues of PGF2α have been used in boars with positive results on sexual behaviour, but limited impact on semen quality. The objective of this study was to determine whether the administration of Dinoprost tromethamine or D-Cloprostenol enhances the sexual behaviour and semen quality of sexually inexperienced growing lambs during the onset of the breeding season. The study lasted 10 weeks, beginning during the non-breeding season, with 24 sexually inexperienced Katahdin lambs (5.1 ± 0.5 months old). Three groups of eight lambs each received: (1) vehicle treatment (group CON); (2) 10 mg of Di

Fertility prediction in dairy goats from Murciano-Granadina breed: The role of sperm evaluation and female traits.

Fertility is one of the most economically important traits in farm animals, due to the direct and indirect costs associated to low pregnancy rates. Thus, one of the priority goals in animal reproduction is to predict the performance that the semen doses will have in vivo based on the quality values obtained in laboratory assays. Attempts have been made for getting a predictive model of fertility of frozen-thawed sperm in dairy goats, but similar studies have not been conducted for chilled goat buck sperm doses that are mostly used for artificial insemination in many countries including Spain.

Establishment of a Captive Cave Nectar Bat (Eonycteris spelaea) Breeding Colony in Singapore.

Bats are known natural reservoirs of several highly pathogenic zoonotic viruses, including Hendra virus, Nipah virus, rabies virus, SARS-like coronaviruses, and suspected ancestral reservoirs of SARS-CoV-2 responsible for the ongoing COVID-19 pandemic. The capacity to survive infections of highly pathogenic agents without severe disease, together with many other unique features, makes bats an ideal animal model for studying the regulation of infection, cancer, and longevity, which is likely to translate into human health outcomes. A key factor that limits bat research is lack of breeding bat c

Discovery of a defence-response gene for Eldana saccharina resistance in sugarcane.

Eldana saccharina is a major stalk boring pest of sugarcane in South Africa, causing substantial yield and revenue losses. Developing sugarcane cultivars with durable resistance to E. saccharina is therefore a priority for breeding programmes. In this study, we employed RNA sequencing (RNA-seq) to characterise the transcriptional responses to E. saccharina by comparing infested plants with uninfested controls in sugarcane genotypes displaying contrasting resistance phenotypes. Differential expression analysis, combined with functional annotation, revealed clear and contrasting defence response

Ovicidal, repellent, adulticidal and field evaluations of plant extract against dengue, malaria and filarial vectors.

Mosquitoes are insect vectors responsible for the transmission of parasitic and viral infections to millions of people worldwide, with substantial morbidity and mortality. Infections transmitted by mosquitoes include malaria, yellow fever, chikungunya, filariasis and other arboviruses. Insecticides of botanical origin may serve as suitable alternative biocontrol techniques in the future. The adulticidal activities of crude hexane, benzene, ethyl acetate, acetone and methanol leaf extracts of Acalypha alnifolia were assayed for their toxicity against three important vector mosquitoes, viz., Aed

Integrating multi-omics reveals the mechanisms of elevated [CO2] enhances salt tolerance of tobacco plants with a photosynthetic regulatory network.

Abiotic stresses often occur concurrently with global changes, especially the combination of salt stress and elevated CO2 concentration ([CO2]) is a common phenomenon in salt-affected soils throughout the world. The synergistic regulatory mechanisms of tobacco (Nicotiana tabacum L.) plants in response to combined salt stress and elevated [CO2] are still poorly understood. This study aimed to reveal the potential mechanisms of elevated [CO2] modulating leaf photosynthesis of tobacco plants subjected to salt stress by integrating physiological, transcriptional, and metabolomics analyses. Functional divergence of two soybean cytosolic serine hydroxymethyltransferases in development and defense against soybean cyst nematode.

Serine hydroxymethyltransferase (SHMT) is an enzyme essential for one-carbon metabolism. In higher plants, multiple SHMT genes code for isoforms that function in the cytosol, nucleus, mitochondria, and chloroplasts. The soybean genome contains two cytosolic SHMTs, GmSHMT05 and GmSHMT08, sharing high sequence identity and similar expression throughout soybean development. In certain soybean genotypes, two amino acid substitutions negatively impact GmSHMT08's ability to bind to tetrahydrofolate (THF), leading to a gain-of-function in resistance to the soybean cyst nematode (SCN). Whether this pe

Northern Light Reviews - Rethinking Plant Stress Biology in a Multifactorial World.

The accelerated pace of climate change, soil degradation, environmental pollution, and pathogen outbreaks subjects trees and crops growing in different parts of the world to multiple abiotic/biotic stress conditions, simultaneously or sequentially. Unfortunately, most studies conducted to date, including studies of two-stress combinations, do not mimic the complex multifactorial stress combination (MFSC) environment plants experience in nature. Here, we summarize evidence from recent MFSC studies of three or more stresses simultaneously impacting plants, and argue that environmental complexity

Rapeseed WRKY53 regulates pattern-triggered immunity (PTI) through modulating the transcription of salicylic acid biosynthetic genes.

Plants have developed a few major immunity systems to combat extraneous pathogens. These systems include pattern-triggered immunity (namely PTI, basal resistance), effector-triggered immunity (called ETI) and systemic acquired resistance (called SAR). Quite a few members of WRKY transcription factor family are famous for their roles in regulating plant immunity through transcriptional reprogramming in Arabidopsis and other plants. Among these, AtWRKY53 plays a pivotal role in leaf senescence and also positively regulate basal resistance against the bacterial pathogen Pseudomonas syringae. Howe
