[Survey on vaccination strategies used in different Italian Regions, for the 7-valent conjugate pneumococcal, meningococcal C and varicella vaccines].

A survey was performed to obtain information regarding regional vaccination strategies in Italy, for 7-valent conjugate pneumococcal, meningococcal C and varicella vaccines. A questionnaire was sent to the person in charge of infectious disease control and vaccination activities in each of the 21 regions. All 21 regions replied to the questionnaire. Twenty regions offer pneumococcal 7-valent conjugate vaccine free only to high-risk children, while 9 of these regions offer it to all newborns as well. Seventeen regions offer meningococcal C vaccine only to high-risk individuals while 9 regions o

Immune response to octavalent diphtheria- and tetanus-conjugated pneumococcal vaccines is serotype- and carrier-specific: the choice for a mixed carrier vaccine.

BACKGROUND: Development of protein-conjugated pneumococcal vaccines for infants has led to formulations that are immunogenic in the age group at highest risk for pneumococcal diseases. This study focuses on the search for an optimal formulation. METHODS: In a randomized trial Icelandic infants (n = 160) were immunized at age 3, 4 and 6 months with one of two octavalent pneumococcal conjugate vaccines (serotypes 3, 4, 6B, 9V, 14, 18C, 19F and 23F conjugated to diphtheria toxoid (PncD) or tetanus protein (PncT) followed with a booster of either the same conjugate or 23-valent polysaccharide vac…

A phase 3 trial of safety, tolerability, and immunogenicity of V114, 15-valent pneumococcal conjugate vaccine, compared with 13-valent pneumococcal conjugate vaccine in adults 50 years of age and older (PNEU-AGE).

BACKGROUND: Pneumococcal conjugate vaccines (PCVs) have greatly reduced the incidence of pneumococcal disease, yet unmet medical need remains due to increased disease caused by non-vaccine serotypes (STs). V114 (VAXNEUVANCETM, Merck Sharp & Dohme Corp., a subsidiary of Merck & Co., Inc., Kenilworth, NJ, USA) is a 15-valent PCV containing 13 serotypes in licensed PCV13 and 2 additional serotypes (22F, 33F) which significantly contribute to pneumococcal disease burden. This phase 3 trial compared safety, tolerability, and immunogenicity of V114 to PCV13 in adults ≥50 years of age. METHODS: Adul

Effectiveness and impact of 7 and 13-valent pneumococcal conjugate vaccines in Chinese children: A systematic review and meta-analysis.

Pneumococcal conjugate vaccines (PCVs) are highly effective against pneumococcal diseases, yet coverage in China remains low. We conducted a systematic review of observational studies published between 2006 and 2024 in mainland China, Hong Kong, and Taiwan to evaluate the effectiveness and impact of PCV7 and PCV13. Twenty-seven studies were identified, of which 17 were included in meta-analyses. PCVs showed strong protection against invasive pneumococcal disease (pooled effectiveness: 88.8%, 95%CI: 80.7-96.8%) and significantly reduced the incidence of community-acquired pneumonia (14.3%, 95%C

Enantiomer-dependent and modification-free DNA matrix as an adjuvant for subunit vaccines against SARS-CoV-2 or pneumococcal infections.

The emergence of novel infectious disease has intensified demand for more advanced vaccine development and more potent adjuvants to enhance immunogenicity. Here we introduce a dynamic DNA supramolecular matrix assembled from five unmodified, short DNA single strands, serving as a safe, multifaceted adjuvant platform. This DNA matrix elicits a robust humoral response with minimal adverse effects, generating potent neutralizing antibodies and conferring robust protection against SARS-CoV-2 and Streptococcus pneumoniae infections. Its dynamic colloidal feature prolongs the in vivo retention of bo

High genetic diversity of Staphylococcus aureus strains colonising the nasopharynx of Gambian villagers before widespread use of pneumococcal conjugate vaccines.

BACKGROUND: With the global efforts of reducing pneumococcal disease through widespread introduction of pneumococcal vaccines, concerns have emerged on the potential increase of morbidity and mortality from S. aureus disease. Little is known however, of the carriage rates of S. aureus or of its' relationship with carriage of S. pneumoniae in rural Africa, and West Africa in particular where very high rates of carriage of S. pneumoniae have been reported. This study aims to evaluate the prevalence, antibiotic susceptibility patterns and genotypes of S. aureus isolated from the nasopharynx of he…

Impact of pneumococcal conjugate vaccines on nasopharyngeal carriage and invasive disease among unvaccinated people: review of evidence on indirect effects.

BACKGROUND: Invasive disease due to Streptococcus pneumoniae remains an important worldwide cause of morbidity and mortality, particularly in young children and the elderly. The development and use of pneumococcal conjugate vaccines (PCVs) have had a dramatic impact on rates of vaccine-type invasive pneumococcal disease (IPD) not only in the pediatric population targeted for vaccination but in non-vaccinated age-groups as well. This indirect effect is directly mediated by a reduction of vaccine-type nasopharyngeal carriage and thus transmission by vaccinated children. Current PCV licensing pro

CpG oligodeoxynucleotides act as adjuvants for pneumococcal polysaccharide-protein conjugate vaccines and enhance antipolysaccharide immunoglobulin G2a (IgG2a) and IgG3 antibodies.

Pneumococcal polysaccharide-protein conjugate vaccines elicit antipolysaccharide antibodies, but multiple doses are required to achieve protective antibody levels in children. In addition, the immunogenicity of experimental multivalent pneumococcal conjugate vaccines varies with different polysaccharide serotypes. One strategy to improve these vaccines is to incorporate an adjuvant to enhance their immunogenicity. Synthetic oligodeoxynucleotides containing unmethylated CpG motifs (CpG ODN) are adjuvants that promote T-cell and T-dependent antibody responses to protein antigens, but it has been

The Role of Pneumococcal and Human Papillomavirus Vaccination in Preventing Otolaryngological Diseases: Implications for Pediatric Practice.

Vaccinations are a cornerstone of primary prevention in pediatric otolaryngology and play a critical role in reducing the burden of pediatric infectious diseases, particularly acute otitis media (AOM) and HPV-related infections. The introduction and widespread use of pneumococcal conjugate vaccines (PCVs) have been associated with substantial changes in the epidemiology of pediatric AOM, particularly through reductions in vac-cine-serotype disease and complicated forms, while contributing to changes in pathogen distribution with an increased role of non-vaccine serotypes and non-pneumococcal b

What is driving the resurgence and persistence of vaccine-targeted pneumococcal serotypes?-Serotype 19F as the paradigm.

Over the past 25 years, pneumococcal conjugate vaccines (PCVs) have markedly reduced both pneumococcal disease and nasopharyngeal carriage caused by vaccine serotypes among the more than 100 known pneumococcal serotypes. In the United States, the transition from the original 7-valent formulation (PCV7) to the 13-valent vaccine (PCV13) occurred approximately a decade after the initial introduction of PCVs, whereas several other countries implemented higher-valency formulations over shorter time intervals. More recently, next-generation PCVs targeting 15 (PCV15) or 20 (PCV20) serotypes have been

[Screening for iron deficiency and pneumococcal vaccination: What role do they play in community cardiology?].

This article presents a single-center, cross-sectional, descriptive epidemiological field study evaluating the screening for iron deficiency and pneumococcal prevention by community cardiologists in patients with heart failure. In total, 90% of the practitioners surveyed reported being familiar with the European Society of Cardiology's recommendations regarding iron deficiency screening, but only 60% perform it at least once a year. In contrast, 90% do not assess the patient's pneumococcal vaccination status. Nearly all respondents stated they are willing to collaborate with an advanced practi

Assessing the impact of childhood pneumococcal vaccination on pneumonia mortality in Colombia: a 14-year analysis.

BACKGROUND: The 10-valent pneumococcal conjugate vaccine (PCV10) has been offered to all infants through Colombia’s National Immunization Program (NIP) since 2012, with catch-up vaccination until age 5. However, pneumococcal vaccination is not currently included in the NIP for other age groups, such as those ≥ 5 years with medical conditions or older adults. This study assessed the pediatric PCV10 effect on pneumonia mortality rate (MR) trends across different age groups from 2006 to 2019. METHODS: This retrospective time-series study utilized the national death registration data. Deaths cause

Evolution of hospitalizations of adults for Community-Acquired Pneumonia in Portugal and the impact of pediatric pneumococcal vaccination.

In Portugal, universal and free paediatric pneumococcal vaccination began in January 2015 with the inclusion of the 13-valent conjugate vaccine in the National Immunisation Programme (NIP). To characterise the evolution of adult hospitalisations for Community-Acquired Pneumonia (CAP) and to assess the impact of children pneumococcal vaccination on adult hospitalisations, we conducted a retrospective analysis of all adult hospitalisations in the National Health Service in Portugal from 2010 to 2019. A comparison with the period from 2000 to 2009 was also performed. The analysis utilised coded i

Pneumococcal serotypes and resistance profiles among children with and without PCV13 vaccination in Shenzhen, China.

Streptococcus pneumoniae remains a major cause of lower respiratory tract infections (LRTIs) in children, and the introduction of pneumococcal conjugate vaccines has altered serotype circulation in many regions. In China, PCV13 has been available since 2016 as a voluntary, self-paying vaccine, resulting in uneven coverage across populations. This retrospective study analyzed 425 children under 14 years of age with culture-confirmed S. pneumoniae LRTIs treated at Shenzhen Children's Hospital between 2021 and 2022, comparing serotype distribution and antibiotic resistance profiles according to P

Immunogenicity of the 23-valent polysaccharide pneumococcal vaccine in children with steroid sensitive nephrotic syndrome.

BACKGROUND: Idiopathic nephrotic syndrome (INS) is the most common chronic kidney disease in childhood, with steroid sensitive nephrotic syndrome (SSNS) as the predominant form. Children with SSNS are at increased risk for invasive pneumococcal disease due to disease-related and treatment-induced immunosuppression. Evidence on immunogenicity of the 23-valent pneumococcal polysaccharide vaccine (PPV23) in this group is limited. This prospective study is the first to evaluate PPV23 exclusively in children with SSNS during remission, directly compared with healthy controls. METHODS: Thirty patie

Evolving pneumococcal epidemiology and vaccine impact in the Netherlands, 2004-2024: Carriage and invasive disease.

OBJECTIVES: Since pneumococcal conjugate vaccines (PCVs) were introduced into the Dutch childhood National Immunization Program in 2006, colonization and invasive disease of Streptococcus pneumoniae have declined, alongside shifts in serotype prevalence towards non-vaccine serotypes (NVTs). To monitor changes and estimate the added value of higher-valent PCVs, serotype-specific carriage, invasive pneumococcal disease (IPD) and invasive disease potential were analyzed. METHODS: During the winters of 2018/2019 and 2022/2023 pneumococcal carriage was determined among 24-month-old children and pa
