Comparison of the genomes of 15 avian herpes-virus isolates by restriction endonuclease analysis.

Differentiation of herpesvirus isolates has been performed mostly on the basis of biological properties and serology. In this study, 15 herp … ther be subdivided into four subgroups. Two other groups are represented by herpesvirus isolates of quail and crane, and by isolates of pigeon and owl. Duck plague virus, herpesvirus of turkey, infectious laryngotracheitis virus and a herpesvirus isolate from tragopan all exhibited distinct restriction patterns. In general, our results parallel earlier …

Laboratory markers for over-attenuation of mumps vaccine virus.

The properties of mumps vaccine virus (Leningrad-3 strain) gradually changed upon passaging in quail embryo fibroblasts, the substrate normally used for mumps vaccine production in the USSR. Alterations were extremely noticeable in the over-attenuated (38th passage) virus variant, and involved (a) poor, if any, antibody response in guinea-pigs, (b) turbid plaque formation, (c) lack of expression in cell culture of fusion protein and reduced expression of polymerase protein, and (d) enrichment by abnormally small, fusion-protein-deficient virus particles. Two other laboratory strains exhibited …

[Use of DEAE-Sephadex A-50 microcarriers for reproduction of the mumps virus].

Reproduction of the vaccine L-3 strains of mumps virus was studied in cultures of continuous Vero cells and in primary cultures of Japanese quail embryos (JQE) growing on DEAE-Sephadex A-50 microcarriers. The Vero cell culture multiplies actively on the microcarrier surface giving more than a 20-fold increase in 8 days. Mumps virus showed a high reproductive capacity in Vero cell culture and in primary JQE cells. Mumps virus-infected Vero cells produce 5-6 pools of virus-containing material with a mean infectious titre 8.2-8.3 lg HAE50/ml. The primary JQE culture infected with mumps virus can …

Effect of reserpine pretreatment on avian erythrocyte carbonic anhydrase activation by isoproterenol.

We studied the action of beta-adrenergic agonists on Japanese quail erythrocyte carbonic anhydrase (CA) in vitro. Earlier we had reported that epinephrine increased CA activity by 14%; the present study focused on an attempt to increase the size of this response. Washed erythrocytes from reserpine-treated (1 mg/kg daily i.m. for 3 days) and control birds were incubated for 40 min in the presence of isoproterenol 10(-6) mol/l. The activity of CA expressed as Wilbur-Anderson units/mg hemoglobin was increased by as much as 42% in reserpine-treated birds over the control depending on the condition…

[The calorigenic effect of glucagon].

The calorigenic action of glucagon was studied in several species by measuring its effect on the oxygen comsumption. The calorigenic effect was most pronounced in the quail and also evident in young adult rats of two different strains but not in mice and guinea pigs. The influence of various drugs on the calorigenic effect of glucagon was investigated in rats. Ganglionic blocking agents (hexamethonium, mecamylamine, chlorisondamine) as well as adrenergic blocking drugs (propranolol, butoxamine, phentolamine) greatly reduced the glucagon-induced calorigenesis but after pretreatment with reserpi…

The efficacy of a standardised product from dried leaves of Solanum glaucophyllum as source of 1,25-dihydroxycholecalciferol for poultry.

1. Chemical characterisation of an extract of Solanum glaucophyllum (SG) leaves affirmed the predominant presence of 1,25-dihydroxycholecalciferol (1,25(OH)2D3) glycosides. The compound 1-(ß-D-glucopyranosyl)-1α,25-dihydroxycholecalciferol was isolated for the first time from a natural source. 2. Vitamin D activity of the extract was confirmed by the calcaemic properties shown in a quail eggshell bioassay. The results suggested a 1,25(OH)2D3 bioavailability of approximately 15%. 3. A broiler feeding experiment replicated in time was carried out with 6 treatments. A basic control diet containin…

Sperm characteristics of Chukar partridge (Alectoris chukar) breeders as affected by the addition of calcitriol to the semen extender.

This study was carried out to determine the effect of supplementing the semen extender with calcitriol on in vitro sperm characteristics in Chukar partridges. A total of 60 male Chukar partridges were habituated for semen collection by abdominal massage. Pooled ejaculates from several males were extended (1 to 5 v/v ratio) in the Sexton's diluent containing 0, 24, 48, 96, or 192 μg calcitriol/mL. These concentrations represented 0-, 2-, 4-, 8-, and 16-fold levels of the mean seminal calcitriol concentration, respectively. A total of 12 subsamples from each treatment group were kept at 4 to 5°C

Novel tissue mechanics-guided cellular flows drive the formation of feather follicles.

Complex tissue architecture is achieved through multiple rounds of morphological transitions. Here, we analyzed cellular flows and tissue mechanics during avian skin development by employing chicken and transgenic quail skin explant models. We demonstrate how novel cellular flows initiate chemo-mechanical circuits that guide epithelial protrusion, folding, invagination, and spatial cell fate specification. During initial feather bud formation, stiff dermal condensates protrude vertically from the locally softened epithelial sheet. As the bud elongates, it stretches the epithelial cells at the …

Actin and myosin dynamics during epithelial remodeling in avian gastrulation.

Epithelial remodeling is powered by contractile forces exerted by the actomyosin cytoskeleton. In invertebrates, pulsatile contractile flows of the medio-apical actomyosin cortex have been shown to be crucial in promoting junction contraction, ultimately driving cell rearrangements, apical constriction and cell extrusion. However, how actomyosin dynamics drive epithelial remodeling in vertebrates, especially amniotes, remains poorly understood. In this study, we generated transgenic quail lines reporting actin and myosin, and investigated their dynamics in gastrulating embryos. We show that, d…

Barbiturate potentiation in mercury poisoning.

Barbiturate potentiation was observed in Japanese quail fed dietary levels of 4, 21, and 24 ppm Hg as methyl mercuric chloride. Gross symptoms of mercury poisoning were not observed until after BP was observed. After the initial increase in BP time was observed in the birds receiving 4 and 12 ppm mercury, there followed a plateau in response until those birds receiving 24 ppm Hg began to show gross symptoms of toxicity and the BP time markedly rose again. Pronounced BP persisted 7 weeks after removal of mercury from the diet reflecting the long biological half-life of mercury as the methyl der…

Quantitative live imaging reveals PRICKLE1 controls junctional neural tube morphogenesis independent of Planar Cell Polarity.

Neurulation, the process that forms the neural tube - the precursor to the brain and spinal cord - is frequently disrupted in congenital malformations. Primary and secondary neurulation are integrated at a junctional zone, yet the cellular dynamics linking these programs remain unknown. Using high-resolution quantitative live imaging in transgenic quail embryos, we show that the junctional neural tube forms through two coordinated processes: mediolateral convergence and EMT-driven ingression of medial neuroepithelial cells. We demonstrate that PRICKLE1, a core PCP protein, orchestrates these b…

Restoration of function by replacement of spinal cord segments in the rat.

Reconstruction of a severed mammalian spinal cord with restoration of function has so far not been achieved, although structural and functional restitution after spinal transection has been successful in some lower vertebrates. In quail-chick and chick-chick chimaeras, spinal cord segments were found to be functional after replacement by isotopic and isochronic grafting of the neural tube. Here we achieve such a replacement in neonatal rats under less restricted topological and temporal conditions than were necessary for the avian chimaeras. The replaced segments united with the host spinal co…

Comparative thermostability of West Nile, St. Louis encephalitis, and western equine encephalomyelitis viruses during heat inactivation for serologic diagnostics.

During the monitoring of arbovirus seroprevalence in wild birds collected in California, we inadvertently made two isolates of western equine encephalomyelitis virus (WEEV) from California quail sera being tested by plaque reduction neutralization assay for antibodies against St Louis encephalitis (SLEV) and West Nile (WNV) viruses despite heating the sera at 56 degrees C for 30 minutes. These data prompted us to examine the thermostability of these viruses during heat treatment. The flaviviruses, SLEV and WNV, at titers up to 10(6) plaque-forming units (PFU), were readily inactivated by the s…

Blood sampling from the venous occipital sinus of birds.

Venous occipital sinus blood sampling was demonstrated to be an effective nonlethal method in all species tested including ducks, geese, Chukar partridge, Japanese quail, pea-fowl, pheasants, parakeets, turkeys, and chickens. Maximum blood volume obtained from the occipital sinus of 1-day-old chicks equalled the volume obtained by cardiac puncture and exceeded the volume collected by decapitation. Venous occipital sinus puncture was successfully used to rapidly collect several successive blood samples from individual birds, thus indicating the method's utility for serial blood sampling. Hemato…
