Close resemblance of murine and human trials is essential to achieve the best predictive value of animal-based translational cancer research. Kras-driven genetically engineered mouse models of non-small-cell lung cancer faithfully predict the response of human lung cancers to systemic chemotherapy. Owing to development of multifocal disease, however, these models have not been usable in studies of outcomes following focal radiotherapy (RT). We report the development of a preclinical platform to deliver state-of-the-art image-guided RT in these models. Presence of a single tumour as usually dia

R-spondins are secreted ligands that bind cell surface receptors and activate Wnt/β-catenin signaling. Human mutations and gene inactivation studies in mice have revealed a role for these four proteins (RSPO1-4) in diverse developmental processes ranging from sex determination to limb development. Among the genes coding for R-spondins, only inactivation of Rspo3 shows early embryonic lethality (E10.5 in mice). Therefore, a conditional allele of this gene is necessary to understand the function of R-spondins throughout murine development. To address this need, we have produced an allele in whic

Corticotropin-releasing factor (CRF) orchestrates the mammalian endocrine, autonomic, and behavioral stress response and has been implicated in the pathophysiology of illnesses ranging from irritable bowel syndrome to mood and anxiety disorders. CRF is produced and released from a variety of cell types, making it difficult to distinguish the specific role of CRF from other neurotransmitters with which it colocalizes. To clarify the basic biology of the CRF neuron, we must be able to manipulate selectively CRFergic cells. Here we describe a novel transgenic mouse using 3.0 kb of the CRF promote

Accurate response to replication arrest is crucial to preserve genome stability and requires both the ATR and ATM functions. The Werner syndrome protein (WRN) is implicated in the recovery of stalled replication forks, and although an ATR/ATM-dependent phosphorylation of WRN was observed after replication arrest, the function of such modifications during the response to perturbed replication is not yet appreciated. Here, we report that WRN is directly phosphorylated by ATR at multiple C-terminal S/TQ residues. Suppression of ATR-mediated phosphorylation of WRN prevents proper accumulation of W

BACKGROUND: Dicer, an RNAse III endonuclease critical for processing of pre-microRNAs (miRNAs) into mature 22-nucleotide miRNAs, has proven a useful target to dissect the significance of miRNAs biogenesis in mammalian biology.
METHODS AND RESULTS: To circumvent the embryonic lethality associated with germline null mutations for Dicer, we triggered conditional Dicer loss through the use of a tamoxifen-inducible Cre recombinase in the postnatal murine myocardium. Targeted Dicer deletion in 3-week-old mice provoked premature death within 1 week accompanied by mild ventricular remodeling and dram

Megalin is an endocytic receptor highly expressed in the proximal tubules of the kidney. Recently, we demonstrated that this receptor is essential for the renal uptake and conversion of 25-OH vitamin D3 to 1,25-(OH)2 vitamin D3, a central step in vitamin D and bone metabolism. Unfortunately, the perinatal lethality of the conventional megalin knockout mouse model precluded the detailed analysis of the significance of megalin for calcium homeostasis and bone turnover in vivo. Here, we have generated a new mouse model with conditional inactivation of the megalin gene in the kidney by using Cre r

Immunoglobulin (Ig) and T-cell receptor (TCR) genes are rearranged in virtually all acute lymphoblastic leukemia (ALL) cases. However, the recombination patterns display several unusual features as compared to normal lymphoid counterparts. Cross-lineage gene rearrangements occur in more than 90% of precursor-B-ALL and in approximately 20% of T-ALL, whereas they are rare in normal lymphocytes. Approximately 25-30% of the Ig and TCR gene rearrangements at diagnosis are oligoclonal, and can undergo continuing or secondary recombination events during the disease course. Based on our extensive mole

We have investigated the capacity of precursor B cells from normal (BDF1) and V(D)J recombinase-deficient (RAG-27) or defective (SCID) mice to be induced by a CD40-specific monoclonal antibody and IL-4 to epsilon H chain gene transcription and to S mu-S epsilon switch recombination. In differentiating precursor B cells from all three strains of mice, the development of similar numbers of CD19+, CD23+, CD40+, and MHC class II+ expressing B lineage cells and similar levels of epsilon H chain gene transcription were induced. Efficient S mu-S epsilon switching occurred in normal and RAG-2-deficien

The V-(D) -J recombination that occurs during lymphoid differentiation is the basic event which settles the diversity of antigen receptors. Two recently cloned genes, RAG1 and RAG2, are sufficient to induce rearrangements of synthetic substrates when transfected into fibroblasts. In order to determine at which differentiation steps where the human RAG are expressed and in order to investigate the possible impairment of their expression by the leukemic process, we have recently studied a large series of B and T lymphoid neoplasias. We show that RAG are expressed in almost all sIg- B-ALL but not

In this short review, we analyse some recent data describing T-cell receptor gene rearrangement abnormalities occurring specifically in B-lineage ALLs. Numerous studies have shown that the lineage specificity of TCR gene rearrangements is lacking in malignant pre-B cells. The resulting inappropriate TCR gene recombinations use the recombinase machinery common to normal B and T cells and, with the exception of the TCRb locus, are similar to gene rearrangements associated with early stages of T-cell ontogeny. In most cases, these rearrangements are not efficiently transcribed. There is no clear

Chicken infectious anemia virus (CIAV) is a major immunosuppressive pathogen that causes huge economic losses in the poultry industry. The virus persists in the host for an extended period, making it difficult to eliminate and highlighting the need for rapid and accurate detection methods. In this study, we developed a recombinase polymerase amplification-lateral flow dipstick (RPA-LFD) kit for on-site detection of CIAV. Optimal primers and probes were designed and screened based on conserved regions of the full-gene sequences of 251 CIAV strains, ensuring high specificity, with no cross-react

Sirt7 is a member of the sirtuin family of proteins, which are NAD+-dependent deacetylases and ADP-ribosyltransferases. It is involved in a wide range of cellular processes. To study the specific role of Sirt7 in hematopoiesis during aging, the gene was specifically inactivated in hematopoietic stem cells (HSC). Vav1 promoter mediated expression of CRE recombinase in floxed Sirt7 mice resulted in specific inactivation of Sirt7 in the hematopoietic stem and progenitor cells. Young mice exhibited a normal peripheral blood count and no detectable hematological aberrancies. Peripheral blood of 19-

Pseudomonas plecoglossicida is the etiological agent of visceral white spot disease, which induces significant mortality in economically important fish such as the large yellow croaker. In this study, we integrated recombinase polymerase amplification (RPA) with CRISPR/Cas12a-mediated detection to establish a fluorescence-based assay for rapid identification of P. plecoglossicida. The complete single-tube, two-stage RPA-CRISPR/Cas12a workflow can be performed within ~45 min. Using purified genomic DNA, the assay achieved an analytical detection limit of 1.65 copies μl-1 and showed no cross-rea

Streptococcus agalactiae (Group B Streptococcus, GBS) is a major cause of neonatal and invasive infections, necessitating rapid and sensitive diagnostic approaches beyond conventional culture and PCR. Here, we developed an isothermal recombinase polymerase amplification (RPA)-MbCas12a CRISPR assay for fluorescence-based detection of GBS using a recently characterized MbCas12a variant from Moraxella bovoculi. The assay targets two conserved housekeeping genes, sodA and groL, and operates under isothermal conditions at 37 °C with minimal sample processing. Analytical sensitivity reached 1.6 × 10

Vibrio cholerae is an important foodborne pathogen that causes severe diarrhea and poses a serious threat to public health. In this study, we developed a novel visual biosensor platform that integrates recombinase-aided amplification (RAA) with lateral flow assay (LFA) technology for the rapid and sensitive detection of the Vibrio cholerae outer membrane protein W (OmpW) gene. The system used multiple phosphorylated primers in the RAA reaction to generate DNA amplicons of different lengths. Subsequently, the amplification products were treated with λ exonuclease, and the resulting single-stran

Severe fever with thrombocytopenia syndrome virus (SFTSV) is a tick-borne virus that primarily causes SFTS. Although a common testing route is available, a timely, conventional and accurate method for SFTSV detection is urgently needed. In the present study, we established a platform that combines the recombinase polymerase amplification (RPA) assay with the clustered regularly interspaced short palindromic repeats-CRISPR associated proteins (CRISPR/Cas) 12a technique in one step in one pot. The procedure can be completed within 45 min at a constant temperature without a sophisticated instrume

We describe a low-cost up scalable molecular test platform providing accessibility to frequent testing - the prerequisite to control infectious epidemics. The device uses the Multi-Spectral Digital Sensor AS7341 in a small fluorescence spectrometer which communicates with a Smartphone App via Bluetooth, which passes on the raw data to outsourced computation in the cloud via the internet. This very stable setup allows easy to use on site testing of individual samples and anonymised data analysis while providing the basis for direct result transmission of a rapid isothermal molecular diagnostic

Experimental access to cell types within the mammalian spinal cord is severely limited by the availability of genetic tools. To enable access to spinal motor neurons (SMNs) and SMN subtypes, we generated single-cell multiome datasets from mouse and macaque spinal cords and discovered putative enhancers for each neuronal population. We cloned these enhancers into adeno-associated viral vectors driving a reporter fluorophore and functionally screened them in the mouse. We extensively characterized the most promising candidate enhancers in rat and macaque and developed an optimized pan-SMN enhanc

Cancer evolution is a multifaceted process leading to dysregulation of cellular expansion and differentiation through somatic mutations and epigenetic dysfunction. Clonal expansion and evolution is driven by cell-intrinsic and -extrinsic selective pressures, which can be captured with increasing resolution by single-cell and bulk DNA sequencing. Despite the extensive genomic alterations revealed in profiling studies, there remain limited experimental systems to model and perturb evolutionary processes. Here, we integrate multi-recombinase tools for reversible, sequential mutagenesis from prema

Estrogen receptor α is commonly used in synthetic biology to control the activity of genome editing tools. The activating ligands, estrogens, however, interfere with various cellular processes, thereby limiting the applicability of this receptor. Altering its ligand preference to chemicals of choice solves this hurdle but requires adaptation of unspecified ligand-interacting residues. Here, we provide a solution by combining rational protein design with multi-site-directed mutagenesis and directed evolution of stably integrated variants in Saccharomyces cerevisiae. This method yielded an estro
