UNC45B Reduction With Aging: A Myofiber-Intrinsic Promoting Factor for Sarcopenia.

Skeletal muscle mass and force decline with age, and the loss of muscle force precedes muscle atrophy. However, the underlying mechanisms remain unclear. Here, we investigated the role of the myosin co-chaperone, uncoordinated mutant number-45 myosin chaperone B (UNC45B), in regulating muscle mass and force. UNC45B expression decreased in mouse gastrocnemius muscle with age, particularly at 24 months old, and adeno-associated virus vector-mediated knockdown of Unc45b in 3-month-old mouse triceps surae muscle first reduced plantar flexor torque and then decreased gastrocnemius muscle mass. In a

The role of static and dynamic evaluation of sarcopenia in liver transplant candidates.

BACKGROUND: Sarcopenia, characterized by the loss of skeletal muscle mass, strength, and function, is a prevalent and severe complication of liver cirrhosis, irrespective of its etiology. Despite its early onset in cirrhosis, the impact of sarcopenia on liver transplantation (LT) outcomes is often underestimated. OBJECTIVES: This study evaluates the role of both static and dynamic assessments of sarcopenia in LT recipients, focusing on cardiopulmonary performance and the likelihood of prolonged ICU stays. MATERIAL AND METHODS: We studied 54 LT recipients (median age: 53.5 years, 59% female p…

Sarcopenia and sepsis fuel a self-perpetuating cycle of immunometabolism decline.

Sepsis and sarcopenia are intertwined clinical challenges characterized by profound immunometabolic dysregulation. Traditionally viewed as a passive reservoir, skeletal muscle is now recognized as an active immunometabolic rheostat that dynamically influences systemic inflammation and homeostasis. This review synthesizes recent advances that redefine our understanding of muscle in critical illness, moving beyond simplistic views of catabolism to encompass complex adaptive strategies. This review summarizes recent advances and discusses septic autocannibalism, a process in which muscle proteoly…

The ageing microenvironment in sarcopenia: early alterations and targeted therapeutic strategies.

Sarcopenia is a progressive skeletal muscle disorder associated with ageing and characterized by declines in muscle strength, mass and function. Increasing evidence suggests that its development is not driven solely by myofiber atrophy or impaired protein metabolism, but also by early remodeling of the ageing skeletal muscle microenvironment. This Review summarizes key early alterations in sarcopenia, including dysregulated immune homeostasis, cellular senescence and the senescence-associated secretory phenotype, depletion and dysfunction of muscle satellite cells, disruption of the regenerati…

Sarcopenia in MASLD-Eat to Beat Steatosis, Move to Prove Strength.

The connections between sarcopenia and various chronic conditions, including type 2 diabetes (T2DM), metabolic syndrome (MetS), and liver disease have been highlighted recently. There is also a high occurrence of sarcopenia in metabolic dysfunction-associated steatotic liver disease (MASLD) patients, who are often disregarded. Both experimental and clinical findings suggest a complex, bidirectional relationship between MASLD and sarcopenia. While vitamin D, testosterone, and specific drug therapies show promise in mitigating sarcopenia, consensus on effective treatments is lacking. Recent focu…

A lower atherogenic index of plasma was associated with a higher incidence of sarcopenia.

Sarcopenia is an age-related muscle senescence disease that leads to functional limitations, physical disability and premature death in older adults. Atherogenic index of plasma (AIP) is a novel indicator of atherosclerotic status based on triglycerides and high-density lipoprotein cholesterol. The aim of this study was to investigate the association between AIP and new-onset sarcopenia and its components among middle-aged and older adults in a Chinese community. This cohort study included 7,992 participants who were free of sarcopenia in 2011 in the China Health and Retirement Longitudinal St…

The Roles of myomiRs in the Pathogenesis of Sarcopenia: From Literature to In Silico Analysis.

Senile sarcopenia is a condition of age-associated muscular disorder and is a significant health issue around the world. In the current review, we curated the information from the NCBI, PubMed, and Google Scholar literature and explored the non-genetic and genetic causes of senile sarcopenia. Interestingly, the myomiRs such as miR-1, miR-206, miR-133a, miR-133b, miR-208b, and miR-499 are skeletal muscle's critical structural and functional regulators. However, very scattered information is available regarding the roles of myomiRs in different skeletal muscle phenotypes through a diverse list o…

Influence of Klotho Protein Levels in Obesity and Sarcopenia: A Systematic Review.

The Klotho gene is recognized for its anti-aging properties. Its downregulation leads to aging-like phenotypes, whereas overexpression can extend lifespan. Klotho protein exists in three forms: α-klotho, β-klotho and γ-klotho. The α-klotho has two isoforms: a membrane-bound form, primarily in the kidney and brain, and a secreted klotho protein present in blood, urine, and cerebrospinal fluid. Klotho functions as a co-receptor for fibroblast growth factor-23 (FGF23), regulating phosphate metabolism. The membrane-bound form controls various ion channels and receptors, while the secreted form reg

POCUS ultrasound: A tool for the detection and monitoring of sarcopenia in peritoneal dialysis.

UNLABELLED: The prevalence of Sarcopenia varies between 1.5% and 68% among patients with chronic kidney disease (CKD). This wide range is due to the different diagnostic tools and the cut-off points used. Early diagnosis and intervention of sarcopenia are essential because of its association with poorer quality of life (QoL), higher hospitalization rates, and adverse events. The "European Working Group on Sarcopenia in Older People (EWGSOP2)" consensus review suggests ultrasound as an emerging tool to quantify muscle mass. Our objective is to describe the prevalence of sarcopenia risk in perit…

Higher prognostic nutritional index is associated with lower incidence of sarcopenia.

Sarcopenia is a syndrome of progressive, generalized decline in skeletal muscle function and muscle mass. As one of the key factors affecting sarcopenia, nutritional status plays an important role in the occurrence and development of sarcopenia. Therefore, this study selected the National Health and Nutrition Examination Survey (NHANES) database to study the effect of prognostic nutritional index (PNI) on sarcopenia. A total of 8058 adults were included in this study. In multiple linear regression analysis, PNI levels were significantly negatively associated with sarcopenia [0.89 (0.85,0.93)];…
