Herpes simplex 1 stomatitis after cleft palate repair: a case report and guidelines for management.

Herpes simplex virus (HSV) primary infection and reactivation has been associated with the inflammation and transient decrease in immunocompetence after surgery and local trauma. In addition, HSV infection is known to impair wound healing, increase risk of scarring, and impede connective tissue graft transplantation. To our knowledge, this is the first case of HSV infection complicating cleft palate repair presented in literature. In this report, we present a case of primary HSV infection occurring in a healthy 26-month-old patient after repair of the secondary cleft palate with mucoperichondr

Serum Th1, Th2 and Th17 cytokine profiles and alpha-enolase levels in recurrent aphthous stomatitis.

BACKGROUND: All aspects of aetiopathogenesis of recurrent aphthous stomatitis (RAS) have not been elucidated. RAS and Behçet's disease (BD) have clinical and immunological characteristics in common. Although T17 cytokines and alpha-enolase have been shown to play effective roles in BD and many other autoinflammatory diseases recently, their roles in RAS have not been studied extensively. In the present study, we investigated levels of several Th1, Th2 and Th17 pathways related cytokines and alpha-enolase to elucidate pathogenesis of RAS and to obtain data about possible treatment alternatives …

Association between MEFV gene mutations and recurrent aphthous stomatitis in a cohort of Turkish patients.

Recurrent aphthous stomatitis (RAS) has a multifactorial etiopathogenesis, an interaction between predisposing factors and/or systemic conditions and immunological components in genetically predisposed subjects. The Mediterranean fever (MEFV) gene has already been identified as being responsible for familial Mediterranean fever. Because the association between MEFV gene mutations and Behçet's disease has been reported before in several studies, we considered that the role of MEFV gene mutations should be studied in patients with RAS, because of the clinical similarities of both diseases. The a…

Preparation of vesicular stomatitis virus-G (VSV-G) conjugate and its use in gene transfer.

The fusiogenic envelope G glycoprotein of the vesicular stomatitis virus (VSV-G) that has been used to pseudotype retrovirus and lentivirus vectors can be used alone as an efficient vehicle for gene transfer. VSV-G protein is secreted into the culture medium as sendimentable vesicles from cells transfected with a VSV-G expression plasmid in the absence of other viral components. The VSV-G vesicles in the conditioned medium can be partially purified by pelleting through sucrose cushion ultracentrifugation. Protein-DNA complexes are formed by mixing the VSV-G vesicles with naked plasmid DNA. Suc…

Comparison of colchicine versus prednisolone in recurrent aphthous stomatitis: A double-blind randomized clinical trial.

PURPOSE: Recurrent aphthous stomatitis (RAS) is one of the most common ulcers of the oral cavity with a reported prevalence of 5- 50%. There is still no definitive treatment for RAS; however, immunosuppressive and immunomodulant agents have been proposed. In this study, we compared the therapeutic effects of 5 mg/d prednisolone with 0.5 mg/d colchicine in the treatment of RAS. METHODS: In a double-blind randomized clinical trial, 34 patients with RAS were randomly divided into two groups for treatment with prednisolone or colchicine. All patients took the medication for three months and were …

Pseudotyped vesicular stomatitis virus for analysis of virus entry mediated by SARS coronavirus spike proteins.

Severe acute respiratory syndrome (SARS) coronavirus (CoV) contains a spike (S) protein that binds to a receptor molecule (angiotensin-converting enzyme 2; ACE2), induces membrane fusion, and serves as a neutralizing epitope. To study the functions of the S protein, we describe here the generation of SARS-CoV S protein-bearing vesicular stomatitis virus (VSV) pseudotype using a VSVdeltaG*/GFP system in which the G gene is replaced by the green fluorescent protein (GFP) gene (VSV-SARS-CoV-St19/GFP). Partial deletion of the cytoplasmic domain of SARS-CoV S protein (SARS-CoV-St19) allowed efficie…

Oral mucosal diseases in the office setting--part I: Aphthous stomatitis and herpes simplex infections.

This article is an update and review of the most common nontraumatic ulcerative and vesicular lesions of the oral cavity. Details concerning their etiology, pathogenesis, clinical presentation, differential diagnosis, and management are included. Comparisons are made between the various forms of aphthous ulcerations and their viral counterparts. Lesions of herpes simplex origin are described for both primary and recurrent or secondary forms and differentiation from aphthous ulcerations is made. Treatment options for both the chronic and more acute forms of this condition are discussed.

Unconventional mechanism of mRNA capping by the RNA-dependent RNA polymerase of vesicular stomatitis virus.

All known eukaryotic and some viral mRNA capping enzymes (CEs) transfer a GMP moiety of GTP to the 5'-diphosphate end of the acceptor RNA via a covalent enzyme-GMP intermediate to generate the cap structure. In striking contrast, the putative CE of vesicular stomatitis virus (VSV), a prototype of nonsegmented negative-strand (NNS) RNA viruses including rabies, measles, and Ebola, incorporates the GDP moiety of GTP into the cap structure of transcribing mRNAs. Here, we report that the RNA-dependent RNA polymerase L protein of VSV catalyzes the capping reaction by an RNA:GDP polyribonucleotidylt…

Phase III controlled evaluation of sucralfate to alleviate stomatitis in patients receiving fluorouracil-based chemotherapy.

PURPOSE: Stomatitis is a major dose-limiting toxicity of bolus fluorouracil (5FU)-based chemotherapy regimens, despite the use of oral cryotherapy. Pursuant to preliminary data that suggested a sucralfate oral solution could alleviate chemotherapy-induced oral mucositis, we developed a prospective trial to test this contention. PATIENTS AND METHODS: A phase III, double-blind, placebo-controlled clinical trial was designed. Patients were entered onto the study at the time of the first cycle of 5FU-based chemotherapy. All patients received oral cryotherapy for 30 minutes with each dose of 5FU. …

Lytic effects of serum and mononuclear leukocytes on oral epithelial cells in recurrent aphthous stomatitis.

A radioisotope-release assay, utilizing 51Cr-labeled epithelial cells derived from non-keratinizing oral mucosa, was developed to investigate in vitro cytolytic reactions correlating with recurrent aphthous stomatitis (RAS). The cytolytic effects of sera and mononuclear leukocytes from patients in the early stage of ulceration were compared with those from matched RAS-negative control subjects. RAS sera induced significantly more cytolysis than did matched control sera. Heating the RAS or control sera for 30 min at 56 degrees C abrogated their cytotoxic activity. RAS mononuclear leukocytes, li…

Altered G-protein glycosylation in vesicular stomatitis virus-infected glucose-deprived baby hamster kidney cells.

The glycosylation of the G-protein was analyzed in vesicular stomatitis virus-infected baby hamster kidney cells incubated in the absence of glucose. The results indicate that the G-protein in glucose-starved cells is initially glycosylated from a lipid donor with a glucosylated oligosaccharide which is resistant to endo-beta-N-acetylglucosaminidase H and partially susceptible to alpha-mannosidase. With longer times, the protein-bound carbohydrate chain becomes much more sensitive to alpha-mannosidase while remaining endo-beta-N-acetylglucosaminidase H-resistant. Purified virions from glucose-…

ABA and Stomatal Biology: From Transcriptional and Biosynthesis to Multicellular Coordination Across Cell Types.

Abscisic acid (ABA) was first identified in plants over six decades ago and is now recognized as one of the central phytohormones in plants, regulating growth, development, and physiological adaptation to environmental changes. In this Centennial Update, we use two influential ABA-related articles published in Plant Physiology in the early 2000s and discuss complementary advances in stress-responsive transcription and ABA biosynthetic regulation. We extend the Update from ABA-relevant gene discovery to recent advances in multicellular coordination of stomatal control. Growing evidence supports

Candidate genes with antagonistic roles in stomatal development are associated with population-wide variation in apple.

A sustainable approach to address climate change and increasing water demand in agriculture is breeding for plant functional traits that conserve water and enhance climate resilience. Stomata regulate plant-water relations and are promising targets for crop improvement. Here, we investigate the variation in stomatal density (SD) in a diverse apple population (Malus domestica Borkh.) consisting of 269 accessions. Genome-wide association studies identified robust associations with SD on chromosomes 2, 9, and 10 (classified as SNPs with a P-value higher than adjusted Bonferroni threshold of -log1

ABA and ABA signaling mediate Arabidopsis stomatal response to CO2 via H2O2 and NO production.

The augmentation of atmospheric CO2 concentrations induces reduction in foliar stomatal aperture, thereby affecting photosynthetic capacity and transpiration- mediated cooling in plants. Elucidating the mechanism through which CO2 modulates stomatal dynamics is crucial for understanding plant adaptability to future fluctuations in CO2 levels. Although abscisic acid (ABA) is a pivotal phytohormone regulating stomatal movements, its role in CO2-induced stomatal closure remains unclear. Here, we demonstrate that elevated CO2 failed to induce stomatal closure in wild-type Arabidopsis treated with

Coordinated Responses of Barley in Photosynthesis, Antioxidative System, and Stomatal Regulation Under Flufenoxuron Stress.

Flufenoxuron is extensively applied in agricultural pest control; however, its effects on plant physiological processes remain insufficiently understood. Hence, this study evaluated the effects of flufenoxuron on photosynthetic processes and stomatal regulation in barley. The findings indicated that flufenoxuron significantly reduced photosynthetic efficiency and gas-exchange capacity. Furthermore, flufenoxuron exposure markedly increased abscisic acid (ABA) in leaves but not in roots. Nonetheless, reactive oxygen species (ROS) generation and membrane lipid peroxidation were triggered by flufe

Efficacy of topical erythropoietin gel and mouthwash for recurrent aphthous stomatitis in a randomized clinical trial.

Erythropoietin (EPO) has anti-inflammatory, antioxidant, and wound-healing properties that could be valuable in managing Recurrent Aphthous Stomatitis (RAS). This study aimed to evaluate its clinical effectiveness in either gel in orabase or mouthwash, on pain reduction, ulcer healing, recurrence rates, and salivary IL-2 level in patients with RAS. In this randomized clinical trial, 60 patients with RAS were equally divided into 3 groups: EPO oral gel, EPO mouthwash, or a placebo. Ulcer size, the Pain Visual Analog Scale (P-VAS), and salivary IL-2 levels were evaluated at baseline, day 3, and …

Metabolite profiling of saliva for the discrimination of Behcet's disease, Sjögren's syndrome, and recurrent aphthous stomatitis.

Oral manifestations can be the initial sign of systemic diseases such as Behcet's disease (BD) and Sjogren's syndrome (PSS). Their frequency and morphology vary widely, and recurrent aphthous stomatitis (RAS) often mimics BD ulcers, complicating differentiation on clinical grounds. Nonspecific aphthoid lesions are also seen in PSS. This study aimed to identify distinctive metabolic patterns in saliva that could discriminate BD, PSS, and RAS using global metabolite profiling. Saliva samples were collected from 43 patients (BD, n = 24; PSS, n = 10; RAS, n = 9) after fasting and abstaining from o…
