Substance P and the Neurokinin-1 Receptor: The New CRF.

Substance P (SP) is an 11-amino acid neuropeptide of the tachykinin family that preferentially activates the neurokinin-1 receptor (NK1R). First isolated 85 years ago and sequenced 40 years later, SP has been extensively studied. Early studies identified a role for SP and the NK1R in contraction of intestinal smooth muscle, central pain processing, and neurogenic inflammation. An FDA-approved NK1R antagonist, aprepitant, is used clinically for the treatment of chemotherapy-induced nausea, as the NK1R influences the activity of the brain stem emesis centers. More recently, SP and the NK1R have

Hooked on hormones? An anabolic steroid addiction hypothesis.

Widespread illicit anabolic steroid use has recently been reported. A review of available evidence suggests that elevations of serum levels of steroid hormones, including anabolic steroids, have profound psychological effects. Long-term, high-dose anabolic steroid use may lead to a preoccupation with drug use, difficulty stopping despite psychological side effects, and drug craving. Reductions in serum levels of steroid hormones appear to result in acute hyperadrenergic withdrawal symptoms that respond to steroid replacement or to agents that also ameliorate withdrawal symptoms in alcohol and

A quick and simple screening method for pathological and problem gamblers in addiction programs and practices.

Despite high rates of comorbidity among pathological gambling, substance use disorders, and other psychiatric conditions, health professionals rarely screen their clients for gambling problems. We report on the performance of the NODS-CLiP, an existing brief, three-item screen for problem and pathological gambling, and an alternative four-item screen that demonstrates improved sensitivity, good positive and negative predictive power, and invariance across key demographic groups . Given the high rates of comorbidity, routine and accurate identification of gambling-related problems among individ

Neuronal pathways linking substance P to drug addiction and stress.

Accumulating evidence suggests that the neuropeptide substance P (SP) and its principal receptor neurokinin 1 (NK1) play a specific role in the behavioral response to opioids and stress that may help to initiate and maintain addictive behavior. In animal models, the NK1 receptor is required for opioids to produce their rewarding and motivational effects. SP neurotransmission is also implicated in the behavioral response to stress and in the process of drug sensitization, potentially contributing to vulnerability to addiction or relapse. However, SP neurotransmission only plays a minor role in

Acupuncture for the treatment of drug addiction.

Over the last three decades there has been an increasing interest in acupuncture treatment of substance abuse around the world. Three important steps can be identified in this field. Dr. Wen of Hong Kong was the first (1972) to report that acupuncture at 4 body points and 2 ear points combined with electrical stimulation can relieve opioid withdrawal signs in the addicts. The second major step was made by Dr. M. Smith in New York, the head of the National Acupuncture Detoxification Association (NADA) of the USA, who finalized a protocol (1985), using only ear points without electrical stimulat

The structure of psychopathology in adolescence and its common personality and cognitive correlates.

The traditional view that mental disorders are distinct, categorical disorders has been challenged by evidence that disorders are highly comorbid and exist on a continuum (e.g., Caspi et al., 2014; Tackett et al., 2013). The first objective of this study was to use structural equation modeling to model the structure of psychopathology in an adolescent community-based sample (N = 2,144) including conduct disorder, attention-deficit/hyperactivity disorder (ADHD), oppositional-defiant disorder (ODD), obsessive-compulsive disorder, eating disorders, substance use, anxiety, depression, phobias, and

Addictive Disorders in Adolescents.

Addictive disorders in youth represent a dynamic field characterized by shifting patterns of substance use and high rates of experimentation, while retaining the risky behaviors and negative outcomes associated with established drug classes. Youth/adolescents are also at the forefront of use of new technologies, and non-substance-related disorders are pertinent. These disorders present with similar pictures of impairment, and can be diagnosed following the same principles. An underlying mental disorder and the possibility of a dual diagnosis need to be assessed carefully, and optimal treatment…

A Comparison of Opioid and Nonopioid Substance Users in Residential Treatment for Co-Occurring Substance Use and Mental Disorders.

The past decade has seen a marked increase in the illicit use of opioids, as well as a doubling of the percentage of individuals seeking treatment for opioid use disorders. However, little is known about the differences between opioid users and nonopioid users in residential treatment. Further, no studies have been published that compare opioid users and nonopioid users in treatment for co-occurring substance use and mental disorders. To address this gap, this study examined differences between opioid and nonopioid substance users in residential treatment for co-occurring disorders. Data was d

The effects of repetitive transcranial magnetic stimulation on sleep in individuals with substance use disorders: A systematic review.

INTRODUCTION: Substance use disorders (SUDs) are associated with significant sleep disturbances, which contribute to increased relapse risk and poorer treatment outcomes. Repetitive transcranial magnetic stimulation (rTMS) has emerged as a promising intervention for SUDs and has been shown to improve sleep in other clinical populations. However, its influence on sleep in individuals with SUDs has not been systematically examined. Understanding this relationship is critical, as improving sleep could enhance treatment outcomes and reduce relapse risk. MATERIALS AND METHODS: A systematic literat

The therapeutic use of psychedelic drugs: Legal, policy, and neuroscientific perspectives.

After many years of stigma and neglect, there is a resurgence of interest in the therapeutic use of psychedelic drugs. Anecdotal and evidence-based reports indicate psychedelics as a possible treatment for depression, anxiety, PTSD, substance abuse, and other disorders resistant to conventional medical interventions. The available data, however, are limited and the use of psychedelic substances in therapy remains controversial. This paper presents a collection of reports based on the talks of eighteen renowned experts in science, policy, and law who spoke at a recent symposium organized by Ame

Pharmacological treatments for co-occurring PTSD and substance use disorders: A systematic review.

INTRODUCTION: Post-traumatic stress disorder and substance use disorders commonly co-occur and are associated with worse health outcomes. Currently, only psychosocial therapies are specifically recommended for use in the co-occurring population, but these come with numerous barriers to access and engagement. This study aims to identify potential pharmacological treatments to enhance treatment options and outcomes for this population. METHODS: This systematic review identified studies on pharmacological treatment of co-occurring PTSD and SUD in humans, using validated outcome measurements, wit
