Small Intestinal Bacterial Overgrowth: Comprehensive Review of Diagnosis, Prevention, and Treatment Methods.

Small intestinal bacterial overgrowth (SIBO) is a commonly diagnosed gastrointestinal disorder affecting millions of individuals throughout the United States. It refers to a condition in which there is an excess and imbalance of small intestinal bacteria. Despite its prevalence, it remains underdiagnosed due to the invasive nature of diagnostic testing. Symptoms observed in SIBO, including abdominal distension, bloating, diarrhea, and gas formation, are nonspecific and can overlap with other gastrointestinal disorders. Frequently cited predisposing factors include gastric acid suppression, dys

Molecular Pathogenesis of MALT Lymphoma.

Approximately 8% of all non-Hodgkin lymphomas are extranodal marginal zone B cell lymphoma of mucosa associated lymphoid tissue (MALT), also known as MALT lymphoma, which was first described in 1983 by Isaacson and Wright. MALT lymphomas arise at a wide range of different extranodal sites, with the highest frequency in the stomach, followed by lung, ocular adnexa, and thyroid, and with a low percentage in the small intestine. Interestingly, at least 3 different, apparently site-specific, chromosomal translocations and missense and frameshift mutations, all pathway-related genes affecting the N

Enhanced biodegradation of carbamazepine after UV/H2O2 advanced oxidation.

Carbamazepine is one of the most persistent pharmaceutical compounds in wastewater effluents due to its resistance to biodegradation-based conventional treatment. Advanced oxidation can efficiently degrade carbamazepine, but the toxicity and persistence of the oxidation products may be more relevant than the parent. This study sets out to determine whether the products of advanced oxidation of carbamazepine can be biotransformed and ultimately mineralized by developing a novel methodology to assess these sequential treatment processes. The methodology traces the transformation products of the

Isolation of scFvs that inhibit the NS3 protease of hepatitis C virus by a combination of phage display and a bacterial genetic screen.

The need for inhibitors for enzymes linked with microbial infection, specifically the NS3 protease of hepatitis C virus (HCV), inspired us to develop a unique, rapid and easy color-based method described herein. The NS3 serine protease of HCV has a role in processing viral polyprotein and it has been implicated in interactions with various cell constituents, resulting in phenotypic changes including malignant transformation. NS3 is currently regarded a prime target for antiviral drugs. We established a genetic screen that is based on coexpression of NS3, a beta-galactosidase reporter that is cl

Induction of hydroxyl radical production in Trametes versicolor to degrade recalcitrant chlorinated hydrocarbons.

Extracellular hydroxyl radical ((*)OH) production via quinone redox cycling in Trametes versicolor, grown in a chemically defined medium, was investigated to degrade trichloroethylene (TCE), perchloroethylene (PCE), 1,2,4- and 1,3,5-trichlorobenzene (TCB). The activity of the enzymes catalyzing the quinone redox cycle, quinone reductase and laccase, as well as the rate of (*)OH production, estimated as the formation of thiobarbituric acid reactive substances (TBARS) from 2-deoxyribose, increased rapidly during the first 2-3 days and then remained at relatively constant levels. Under quinone re

Expression of a modified Mannheimia haemolytica GS60 outer membrane lipoprotein in transgenic alfalfa for the development of an edible vaccine against bovine pneumonic pasteurellosis.

The GS60 antigen is one of the protective antigens of Mannheimia haemolytica A1. GS60 contains conserved domains belonging to the LppC family of bacterial outer membrane lipoproteins. A high antibody titer to GS60 has been shown to be significantly correlated with resistance to pneumonic pasteurellosis. Calves vaccinated with a commercial vaccine (Presponse) and demonstrating protection against M. haemolytica A1 produced antibodies directed against GS60. Alfalfa was chosen as the platform for an edible vaccine. Agrobacterium tumefaciens was used to mediate the transformation of alfalfa with se

Immune and Stem Cell-Mediated Bactericidal Amplification and Bone Remodeling for Infection Clearance and Osteointegration.

Implant-related infections reduce mesenchymal stem cell accumulation near implant sites while suppressing the viability and osteogenic capacity of these stem cells. Here, we show that pH-responsive biomaterial interfaces exploit infection-induced acidification to coordinate antimicrobial action with stem and immune cell functional reprogramming. The system can release bactericidal peptides for direct bacterial elimination, whereas indirect bactericidal enhancement occurs in the presence of magnesium ions and cell-targeting peptides, which enable a positive modulation of immune responses and re

A novel sensing probe based on AuNPs-Apt for the detection of enrofloxacin.

Enrofloxacin (ENR), as a synthetic broad-spectrum antibiotic is widely utilized in veterinary medicine to treat animal diseases and promote livestock growth, it can inhibit bacterial DNA gyrase subunit A, thereby preventing bacterial DNA replication and exerting its antibacterial effect. However, excessive use of enrofloxacin poses significant risks to ecological balance and human health due to residual contamination. We have developed a novel ENR aptamer sensor based on the gold nanoparticles/aptamer (AuNPs-Apt) complexes, in which AuNPs were synthesized via the seed method and functionalized

A Microbial Cocaine Bioreporter.

The continuous emergence of new illegal compounds, particularly psychoactive chemicals, poses significant challenges for current drug detection methods. Developing new protocols and kits for each new drug requires substantial time, effort, and dedicated manpower. Whole-cell bacterial bioreporters have been proven capable of detecting diverse hazardous compounds in both laboratory and field settings, identifying not only single compounds but also chemical families. We present the development of a microbial bioreporter for the detection of cocaine, the nervous system stimulant that is the second

Synthesis and structure-activity relationships of teixobactin.

The discovery of antibiotics has led to the effective treatment of bacterial infections that were otherwise fatal and has had a transformative effect on modern medicine. Teixobactin is an unusual depsipeptide natural product that was recently discovered from a previously unculturable soil bacterium and found to possess potent antibacterial activity against several Gram positive pathogens, including methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococci. One of the key features of teixobactin as an antibiotic lead is that resistance could not be generated in a laborato

In-situ oxygen-supplying ROS nanopurifier for enhanced healing of MRSA-infected diabetic wounds via microenvironment modulation.

Hypoxia, high ROS levels and chronic inflammation are the main factors that hinder the healing of diabetic wounds. Long-term exposed wounds are prone to bacterial infection, especially MRSA infection, which exacerbates the complex wound microenvironment of diabetes and threatens patients' lives. Here, we developed a ROS nanopurifier (CSVNP), which was prepared by loading superoxide dismutase (SOD), catalase (CAT) and vancomycin into nanogels through in-situ polymerization. CSVNP can effectively increase enzyme loading and stability, and improve cascade reaction efficiency between enzymes throu

Stabilizing Genetically Unstable Simple Sequence Repeats in the Campylobacter jejuni Genome by Multiplex Genome Editing: a Reliable Approach for Delineating Multiple Phase-Variable Genes.

Hypermutable simple sequence repeats (SSRs) are major drivers of phase variation in Campylobacter jejuni. The presence of multiple SSR-mediated phase-variable genes encoding enzymes that modify surface structures, including capsular polysaccharide (CPS) and lipooligosaccharide (LOS), generates extreme cell surface diversity within bacterial populations, thereby promoting adaptation to selective pressures in host environments. Therefore, genetically controlling SSR-mediated phase variation can be important for achieving stable and reproducible research on C. jejuni. Here, we show that natural "

Retention and biosynthesis of diverse nitroarenes in Escherichia coli after combinatorial nitroreductase gene deletions.

The instability of nitroaromatic compounds or nitroarenes in the presence of microbial cells has hindered the ability to biomanufacture these industrially and pharmaceutically relevant chemicals. Although nitroreductase (NTR) gene deletions have been linked to nitroarene stability, the extent of this effect is unclear. Here we perform a comprehensive analysis of bacterial NTR activity in cells by engineering strains of Escherichia coli that contain up to 15 knockouts of known and candidate NTR genes. We evaluate the stability of over 20 exogenously supplemented nitroaromatic compounds to wild-

Signed, sealed, delivered: a generalizable model for living biotherapeutic dosing and metabolism.

Living Biotherapeutic Products (LBPs) offer a promising therapeutic strategy for metabolic disorders rooted in gut microbiome dysfunction, yet quantitative frameworks for predicting their efficacy remain underdeveloped. We introduce the Bacterial Compartment Absorption and Transit (BCAT) model, a pharmacokinetic-pharmacodynamic framework that couples probiotic transit, endogenous microbiome metabolism, and enzymatic transformation within a unified dose-optimization setting. Building on the classical CAT model, BCAT incorporates mechanistically-derived colon compartments and treats dosing time

The inflammation-epigenetics axis in gastric cancer: Helicobacter-driven field defects, immune escape, and therapeutic frontiers.

Gastric cancer provides a clinically relevant model of the inflammation-epigenetics axis because Helicobacter pylori can drive prolonged mucosal inflammation before malignant transformation. Inflammatory signaling remodels DNA methylation, histone states, chromatin accessibility and RNA-regulatory programs, generating epigenetic field defects that may persist after bacterial eradication. Epstein-Barr virus-positive gastric cancer represents a distinct hypermethylated and immune-adapted state. We organize these processes using four coordinates: inflammatory driver, dominant epigenetic lesion, c

Designing ligands for tuning the active site of novel artificial vanadium-based enzymes for sulfoxidation.

Artificial metalloenzymes, which are formed by inserting inorganic complexes into biological scaffolds, have attracted interest as a potential biotechnological alternative of natural enzymes. In this study, vanadium complexes were incorporated as artificial active sites for sulfoxidation catalysis within NikA, a bacterial Ni(II) import protein. Two of the designed metal complexes, namely VOL1 and VOL3, which are based on either an amino alcohol or an amino acid ligand, respectively, were spectroscopically and structurally characterized, demonstrating transformation of a VOL3 into a VO(L3)2 spe
