A novel mutation in the mitochondrial tRNA for tryptophan causing a late-onset mitochondrial encephalomyopathy.

BACKGROUND: Mitochondrial DNA (mtDNA) mutations are increasingly being recognized as causes of late-onset disease. We report a patient with a late-onset mitochondrial encephalomyopathy caused by a novel G > C transition in mtDNA at position 5556 in the gene encoding the tRNA for tryptophan (MTTW). AIMS: To investigate the cause of disease and assess the pathogenicity of this new mutation. METHODS: Clinical, histopathological and gene sequencing studies. Quantification of the mutation was performed in different tissues from the patient and two relatives and in single muscle fibres. RESULTS: …

Protein-source tryptophan as an efficacious treatment for social anxiety disorder: a pilot study.

Until recently, intact protein that is rich in tryptophan was not seen as an alternative to pharmaceutical-grade tryptophan because protein also contains large neutral amino acids (LNAAs) that compete for transport sites across the blood-brain barrier. Recent evidence indicates that when deoiled gourd seed (a rich source of tryptophan with approximately 22 mg/g protein) is combined with glucose (a carbohydrate that reduces serum levels of competing LNAAs) a clinical effect similar to that of pharmaceutical-grade tryptophan is achieved. Objective and subjective measures of anxiety in those suff…

Mouse tryptophan hydroxylase isoform 2 and the role of proline 447 in enzyme function.

Tryptophan hydroxylase (TPH) is the rate-limiting enzyme in the synthesis of the neurotransmitter serotonin (5-HT). Once thought to be a single gene product, TPH is now known to exist in two isoforms. Isoform 1 (TPH1) is found in the pineal gland and gut, and isoform 2 (TPH2) is selectively expressed in brain. A single-nucleotide polymorphism in TPH2 results in a proline-to-arginine mutation at residue 447 and substantially lowers catalytic activity. In view of the importance of TPH in determining brain 5-HT function, we cloned TPH2 and produced the P447R mutant to assess the importance of thi…

[Determination of tyrosine- and tryptophan-containing peptides in plasma by absorption in UV spectral region].

A method for determining the cumulative concentration of tyrosine- and tryptophan-containing peptides (TSP and TPP) in blood plasma: 0.2 ml of blood plasma is added 0.1 ml of 1.8 M chloric acid, the sample is centrifuged, the supernatant (0.2 ml) is added 1.8 ml of 0.7 M NaOH and the optic density is measured at 290 nm in the quarts or plastic flask. Equations were derived to calculate separately the concentrations of TSP and TPP, whose normal values are 1.28 +/- 0.31 and 0.25 +/- 0.06 mmol/l, respectively. The increasing total content of peptides in thyrotoxicosis was shown to be mainly condi…

Effects of acute tryptophan depletion on cognitive function in Alzheimer's disease and in the healthy elderly.

BACKGROUND: The cholinergic system is profoundly impaired in senile dementia of Alzheimer type (SDAT) and replacement therapy produces only modest clinical benefits. The serotonergic system is also impaired and may contribute both to cognitive and non-cognitive symptoms in SDAT. To investigate this further we assessed the effects of lowering brain serotonin using the technique of acute tryptophan depletion on cognitive function in patients with SDAT and in age matched control subjects. METHOD: Sixteen patients with probable SDAT and 17 healthy elderly subjects received two amino acid drinks i…

Metabolites of the kynurenine pathway of tryptophan metabolism in the cerebrospinal fluid of Malawian children with malaria.

A retrospective study of 100 Malawian children (87 with malaria and 13 with a diagnosis other than malaria) was conducted to determine the relationship between levels of metabolites of the kynurenine pathway in cerebrospinal fluid (CSF) and disease outcome. Three metabolites were measured: quinolinic acid (QA), an excitotoxin; kynurenic acid (KA), a neuroprotective receptor antagonist; and picolinic acid (PA), a proinflammatory mediator. Elevated levels of QA and PA in CSF were associated with a fatal outcome in Malawian children with cerebral malaria (CM). QA was associated with a history of

Effect of tryptophan depletion on impulsive behavior in men with or without a family history of alcoholism.

This study investigated the effects of acute serotonin depletion on two measures of impulsive behavior in healthy men with a family history … men, half of whom had an alcoholic father, participated in a two-session study. Subjects ingested a tryptophan-depleting diet on one session and a balanced diet on the other session, and completed tasks measuring behavioral inhibition and delay discounting. Tryptophan depletion impaired performance on the behavioral inhibition task in the males with a positi …

A structural approach into human tryptophan hydroxylase and its implications for the regulation of serotonin biosynthesis.

Tryptophan hydroxylase (TPH) catalyzes the 5-hydroxylation of tryptophan, which is the first step in the biosynthesis of indoleamines (serotonin and melatonin). Serotonin functions mainly as a neurotransmitter, whereas melatonin is the principal hormone secreted by the pineal gland. TPH belongs to the family of the aromatic amino acid hydroxylases, including phenylalanine hydroxylase (PAH) and tyrosine hydroxylase (TH), which all have a strict requirement for dioxygen, non-heme iron (II) and tetrahydrobiopterin (BH4). During the last three years there has been a formidable increase in the amou…

Synthesis of fluorochromes and pigments in Malassezia furfur by use of tryptophan as the single nitrogen source.

A new minimal medium consisting only of L-tryptophan (L-Trp) and a lipid source induced formation of brown pigmentation only in the species Malassezia furfur, which diffuses into the agar. Strains of the species M. sympodialis and M. pachydermatis failed to grow on this medium. On mDixon medium, however, after replacement of peptone by L-Trp, growth of all three Malassezia species was achieved. Under these conditions pigment production was observed with all M. furfur strains tested, although the results for M. pachydermatis strains were inconsistent. M. sympodialis strains showed no pigment pr…

Resolution of the fluorescence of the buried tryptophan in yeast 3-phosphoglycerate kinase using succinimide.

The heterogeneous fluorescence of yeast 3-phosphoglycerate kinase, a hinge-bending enzyme with two tryptophans, has been resolved into two approximately equal components, one accessible and one inaccessible to the relatively inefficient quencher succinimide. The inaccessible component is blue-shifted and exhibits a heterogeneous fluorescence decay which has a temperature-dependence and steady-state acrylamide quenching properties typical of a single tryptophan in a buried environment. This component is therefore assigned to the buried tryptophan W333. The presence of succinimide greatly simpli…

[Role of tryptophan in the enzymatic activity of histidine decarboxylase from Micrococcus sp. n].

The effect of N-bromosuccinimide (BSI) on micrococcal histidine decarboxylase in 0.07 M phosphate buffer, pH 5.6 was studied. Data from spectral and amino acid analyses suggest that at 20-fold molar excess of BSI three of 12 tryptophane residues undergo selective modification, resulting in 80-85% loss of the enzyme activity. Using fluorescent method and polyacrylamide gel electrophoresis, it was shown that modification of these reactive tryptophane residues does not cause structural changes of the enzyme. Presumably tryptophane residue responsible for enzymatic activity are either located in t…
