Medicinal Chemistry Optimization of a Diaminopurine Chemotype: Toward a Lead for Trypanosoma brucei Inhibitors.

Human African trypanosomiasis (HAT), or sleeping sickness, is caused by the protozoan parasite Trypanosoma brucei and transmitted through the bite of infected tsetse flies. The disease is considered fatal if left untreated. To identify new chemotypes against Trypanosoma brucei, previously we identified 797 potent kinase-targeting inhibitors grouped into 59 clusters plus 53 singleton compounds with at least 100-fold selectivity over HepG2 cells. From this set of hits, a cluster of diaminopurine-derived compounds was identified. Herein, we report our medicinal chemistry investigation involving t…

Epidemiology of human African trypanosomiasis.

Human African trypanosomiasis (HAT), or sleeping sickness, is caused by Trypanosoma brucei gambiense, which is a chronic form of the disease … ology of the disease is mediated by the interaction of the parasite (trypanosome) with the vectors (tsetse flies), as well as with the human and animal hosts within a particular environment. Related to these interactions, the disease is confined in spatially limited areas called "foci", which are located in Sub-Saharan Africa, mainly in remote rural areas. The …

Study on the prevalence and associated risk factors of bovine trypanosomiasis in Zaba Gazo Woreda, Southern Ethiopia.

In tsetse-infested areas of Ethiopia, African Animal Trypanosomiasis is the most serious threat to livestock productivity. A cross-sectional study was conducted from May 2021 to September 2021 in the Zaba Gazo district to assess the prevalence and associated risk factors of bovine trypanosomiasis through parasitological and hematological techniques. In the current study, the overall prevalence of trypanosomiasis was found to be 21%. Among anticipated risk factors, the origin of animals has shown a statistically significant association (p < 0.05) with the occurrence of bovine trypanosomiasis. O

Contemporary and emerging strategies for eliminating human African trypanosomiasis due to Trypanosoma brucei gambiense: review.

OBJECTIVES: To review current and emerging tools for Gambiense HAT control and elimination, and propose strategies that integrate these tools with epidemiological evidence. METHODS: We reviewed the scientific literature to identify contemporary and emerging tools and strategies for controlling and eliminating Gambiense HAT. Through an iterative process involving key stakeholders, we then developed comprehensive scenarios leading to elimination, considering both established and new tools for diagnosis, case treatment and vector control. RESULTS: Core components of all scenarios include detect

Human African trypanosomiasis in endemic populations and travellers.

Human African trypanosomiasis (HAT) or sleeping sickness is caused by the protozoan parasites Trypanosoma brucei (T.b.) gambiense (West African form) and T.b. rhodesiense (East African form) that are transmitted by the bite of the tsetse fly, Glossina spp.. Whereas most patients in endemic populations are infected with T.b. gambiense, most tourists are infected with T.b. rhodesiense. In endemic populations, T.b. gambiense HAT is characterized by chronic and intermittent fever, headache, pruritus, and lymphadenopathy in the first stage and by sleep disturbances and neuro-psychiatric disorders i

Epidemiological study of canine trypanosomosis in an urban area of Ivory Coast.

Following confirmed cases of trypanosomosis in military working dogs, a cross-sectional study was undertaken to evaluate the source of infection and determine the prevalence of canine infection with Trypanosoma congolense in the urban focus of Abidjan, Ivory Coast. Blood from 123 dogs were collected and subjected to PCR using specific primers for Trypanosoma congolense "forest type". In addition, an entomological study was conducted in an urban area near the forest surronding the military camp. The observed prevalence was 30.1% and PCR positivity to Trypanosoma congolense was not significantly

The flagellum of Trypanosoma brucei: new tricks from an old dog.

African trypanosomes, i.e. Trypanosoma brucei and related sub-species, are devastating human and animal pathogens that cause significant human mortality and limit sustained economic development in sub-Saharan Africa. T. brucei is a highly motile protozoan parasite and coordinated motility is central to both disease pathogenesis in the mammalian host and parasite development in the tsetse fly vector. Therefore, understanding unique aspects of the T. brucei flagellum may uncover novel targets for therapeutic intervention in African sleeping sickness. Moreover, studies of conserved features of th

Multiple-strain infections of Trypanosoma brucei across Africa.

It is becoming increasingly clear that parasitic infections frequently contain multiple strains of the same parasite species. This may have important consequences for the parasite dynamics in the host and thus alter disease and transmission dynamics. In Trypanosoma brucei, the causal agent of human African trypanosomiasis (sleeping sickness), multiple-strain infections have previously been demonstrated to occur. Here, we analyzed field isolates of T. b. gambiense, T. b. rhodesiense, and T. b. brucei, isolated throughout Africa to assess the commonness of multiple-strain infections across the n

[Distribution and spread of human African trypanosomiasis: value of genetic identification of the trypanosomes].

Numerous factors extrinsic to trypanosome populations have been implicated in the distribution and spread of human African trypanosomiasis (HAT), but quantification of these factors has proven difficult. An easier method of monitoring HAT consists of tracking parasites by genetic identification of trypanosomes in hosts and vectors. This method requires distinction between Trypanosoma brucei rhodesiense and Trypanosoma brucei gambiense followed by determination of the genotype of each subspecies in the host and vector. The role of vertebrate hosts in the distribution and spread of HAT has been

Can remotely sensed meteorological data significantly contribute to reduce costs of tsetse surveys?

A 0.125 degree raster or grid-based Geographic Information System with data on tsetse, trypanosomiasis animal production, agriculturerkina> and land use has recently been developed in Togo. This paper addresses the problem of generating tsetse distribution and abundance maps from remotely sensed data, using a restricted amount of field data. A discriminant analysis model is tested using contemporary tsetse data and remotely sensed, low resolution data acquired from the National Oceanographic and Atmospheric Administration and Meteosat platforms. A split sample technique is adopted where a rand

Measurement of risk in endemic areas of human African trypanosomiasis in Côte d'Ivoire.

An index of epidemiological risk was developed for the foci of human African trypanosomiasis (HAT) in the forest zone of Côte d'Ivoire, based on the following characteristics of Glossina palpalis palpalis populations: daily survival rate, apparent density of teneral males and females, and frequency of human-fly contact. The index agreed well with HAT prevalence. It varied according to ethnic groups and with seasonal changes in agricultural activities and fell rapidly to zero following the start of an anti-vector control campaign. Further studies in different biogeographical zones are desirable

Epidemiology of bovine trypanosomiasis in the Ghibe valley, southwest Ethiopia. 2. Factors associated with variations in trypanosome prevalence, incidence of new infections and prevalence of recurrent infections.

An average of 840 East African Zebu cattle from nine herds in the Ghibe valley, southwest Ethiopia were monitored from January 1986 to April 1990. Each month blood samples were collected for analysis of packed red cell volume (PCV) and detection of trypanosomes. Animals found to be parasitaemic and with a PCV less than 26% were treated with diminazene aceturate at a dose of 3.5 mg/kg body weight. The majority of infections were associated with Trypanosoma congolense (84% of infections in adult cattle and 71% in cattle less than 24 months of age), and the mean percentage of adult animals detect

Man-fly contact in the Gambian trypanosomiasis focus of Nola-Bilolo (Central African Republic).

A study using bipyramid tetse fly traps in the Nola-Bilolo sleeping sickness focus (Central African Republic) reveals ecological and behavioural differences between two vectors, Glossina palpalis palpalis and G. fuscipes fuscipes. The latter species inhabits mainly open water sites and surrounding forest, whereas G. p. palpalis occurs mainly in coffe plantations near villages. Consequently, the man-fly contact differs considerably according to the species. The intensity of trypanosomiasis transmission, estimated by the probable distribution of cases, showed significant positive correlation wit

[New data on the control of African animal trypanosomiasis].

The glossina control turns towards cheaper, ecological, easier techniques: trapping, which the efficacy might be increased by odour attractants; synthetic pyrethrinoids (impregnated targets; dipping or application of "Pour-on" formulations on skin of cattle). Only one new trypanocide was set up during the last 25 years: Melarsamine, arsenical derivative, effective in camels, cattle, horses infected by Trypanozoon (3.5 mg/kg b. w. by IM or SC routes). The trypanotolerant cattle is characterized by a better control of parasitemia and anaemia, dominant gene A of hemoglobin and gene F of albumin,
