Valproic acid downregulates heme oxygenase-1 independently of Nrf2 by increasing ubiquitination and proteasomal degradation.

AIMS: Heme oxygenase-1 (HO-1; HMOX1 in human, Hmox1 in mice) is an antioxidative enzyme affecting wide range of sub-cellular processes. It was shown to modulate tumor growth or vascular-related diseases, thus being putative molecular target for tailored therapies. Therefore it is of importance to elucidate novel compounds regulating HO-1 activity/expression and to delineate mechanisms of their action. In the present study we aimed to understand mode of action of valproic acid (VA), an antiepileptic drug, on HO-1 expression. RESULTS: We demonstrated that HO-1 expression is decreased by VA at p

Inhibition of protein-tyrosine phosphatase 1B (PTP1B) mediates ubiquitination and degradation of Bcr-Abl protein.

Chronic myelogenous leukemia (CML) is a myeloproliferative disorder characterized at the molecular level by the expression of Bcr-Abl, a chi … PTP1B elicits tyrosine phosphorylation of Bcr-Abl that triggers the degradation of Bcr-Abl through ubiquitination via the lysosomal pathway. The degradation of Bcr-Abl consequently inhibits tyrosine phosphorylation of Bcr-Abl substrates and the downstream production of intracellular reactive oxygen species. Furthermore, PTP1B inhibition reduces cell viability a …

Ubiquitination and proteolysis in limb and respiratory muscles of patients with chronic obstructive pulmonary disease.

Peripheral muscle dysfunction associated with chronic diseases is undeniably a growing problem as one of its main causes, chronic obstructive pulmonary disease (COPD), progresses. Among others, muscle atrophy is one component building the concept of muscle dysfunction. Muscle atrophy has a significant impact on patient clinical status, independent of the impairment in lung function. A lot of effort has been devoted lately to increasing our understanding of the relationship between COPD and the initiation and the development of muscle atrophy. A growing body of evidence is showing that the ubiq

Effect of HSV-1 ICP0 E3 ubiquitin ligase on CIN85 endosomes.

Herpes simplex virus-1 (HSV-1) has infected more than 67% of the world population causing diseases that range in severity from benign cold sores to encephalitis. To infect and persist in the host, HSV-1 has evolved sophisticated strategies to counteract antiviral responses. The viral immediate early protein ICP0 (Infected Cells Protein 0) plays a fundamental role in this process. ICP0 is an E3 ubiquitin ligase that is required for successful onset of lytic infection and productive reactivation of viral genomes from latency. Most ICP0 studies have been focused on its nuclear functions, but late

RING finger E3 ubiquitin ligases: novel therapeutic opportunities in melanoma.

Melanoma is an aggressive type of cancer that is prone to developing resistance to targeted therapies and immunotherapies, so it is necessary to seek novel therapeutic opportunities. RING finger E3 ubiquitin ligases (RNFs) play a crucial role in the ubiquitin-proteasome system and are important in regulating the development of melanoma by orchestrating various pathways. In this review, we analyze the structural and functional characteristics of the RNF subfamily to clarify their mechanisms of action in melanoma and to compare the functional differences among various RNFs. Additionally, we syst

Ubiquitin-Proteasome System-Related Prognostic Model and Immune Landscape in Glioblastoma.

BACKGROUND: Glioblastoma (GBM) is a highly aggressive brain tumor with poor prognosis. This study is aimed at establishing an ubiquitin-proteasome system (UPS)-related prognostic model and investigating its link to immune infiltration and therapy response. MATERIALS AND METHODS: GBM datasets were obtained from public databases. Ubiquitin-proteasome system-related genes (UPSGs) were identified from literature. Consensus clustering defined UPS-based GBM subtypes. Differentially expressed genes (DEGs) were screened, and a prognostic model was constructed using univariate Cox, least absolute shri

Ubiquitin-specific peptidase 51: A potential target in cancer progression and therapy.

Ubiquitination, a reversible post-translational modification regulated by deubiquitinating enzymes, is pivotal for cancer initiation and progression. USP51, an emerging member of the USP family, plays essential roles in maintaining genomic stability and cell cycle homeostasis under physiological conditions, while its dysregulation drives multiple cancer-related biological processes. This review systematically synthesizes the substrate-dependent mechanisms underlying USP51's functional diversity across cancer types, its synergistic interactions with other USP family members via shared substrate…

A novel mechanism of ubiquitin-charging a bivalent E2:RING E3 complex.

Protein ubiquitination results from a cascade of enzyme interactions that transfer ubiquitin from one covalent bond to another. First, an E1 is activated by attaching ubiquitin to itself through a thioester bond. Next, in E2 charging, activated E1 interacts with an E2 and transfers ubiquitin to a thioester bond on the E2. Finally, an E3 mediates the transfer of ubiquitin from a charged E2 to an isopeptide bond on a protein target. RING E3s, the largest E3 family in eukaryotes, function as scaffolds rather than enzymes and facilitate direct transfer of ubiquitin from a charged E2 to a protein t…

Potential Role for the Ubiquitin Ligase SIAH2 in Modulating Bone Morphology and Adiposity.

Marrow adipose expansion, with obesity or ageing, increases fracture risk. Skeletal stem cells can commit to osteoblast or adipocyte endpoints, an inverse relationship influenced by peroxisome proliferator-activated receptor gamma (PPARγ). Seven in absentia homologue 2 (SIAH2) is a ubiquitin ligase that acts in peripheral adipose tissue stromal progenitors, promoting PPARγ activity and adipogenesis. The pivotal role of PPARγ in bone marrow adipose tissue accumulation prompted us to evaluate SIAH2's role in the balance between osteogenesis and adipogenesis. Using adherent skeletal stem and/or p

Ubiquitin-like modifications in adipose tissue development and obesity: New insights.

Obesity involves abnormal adipose tissue remodeling, including increased fat cell number (hyperplasia) and size (hypertrophy), along with dysregulated adipogenesis, function, and turnover of white, beige, and brown fat cells. These changes cause chronic inflammation and insulin resistance, leading to metabolic diseases. Post-translational modifications (PTMs), especially ubiquitin and ubiquitin-like modifications (UBLs), regulate adipose development by attaching to proteins, affecting their activity, stability, and localization through a cascade involving E1, E2, and E3 enzymes. Although UBL m

FLT3-ITD signals for CEBPA and p53 proteolysis by the ubiquitin-proteosome pathway.

Internal-tandem-duplication of the receptor tyrosine kinase FLT3 (FLT3-ITD) generates ligand-independent signaling and is highly recurrent in acute myeloid leukemias (AMLs). One way signaling pathways can quickly influence cell fates is by phosphorylating key fate-determining proteins to trigger their proteolysis. We investigated the master transcription factor (MTF) driver of granulo-monocytic lineage-fates, CEBPA, for regulation by this mechanism because we found high CEBPA mRNA but little CEBPA protein in FLT3-ITD versus FLT3-wildtype AML cells, and inhibiting FLT3-ITD signaling with tyrosi

Identification of E3 Ubiquitin Ligases Associated with Survival in Soft Tissue Sarcomas.

Proteasome inhibitors are approved to treat multiple myeloma and mantle cell lymphoma. Recent reports suggest sarcomas also display proteasome addiction. Mechanistic explanations cite proteotoxic stress. In sarcoma patients, we analyzed the impacts of 377 human E3 ubiquitin ligases on sarcoma patient overall survival (OS) and recurrence-free survival (RFS), identified substrates of E3 ligases with the most significant and robust effects, and performed enrichment analyses. High expression of 102 E3 ligases was associated with shortened OS. Thirteen of these shortened OS by >40 months, six with

LRRC58 defines an E3 ubiquitin ligase complex sensitive to cysteine abundance.

Adaptation to fluctuating nutrient supply is essential for organismal survival, but how human cells monitor the abundance of many critical nutrients remains undefined. Characterizing the conditional degradation of CDO1, the critical enzyme responsible for cysteine catabolism, here we identify a Cullin-RING E3 ligase complex defined by the substrate adaptor LRRC58 that is sensitive to cysteine abundance. When cysteine is replete, LRRC58 activity is restrained through ubiquitination and proteasomal degradation. Upon cysteine deprivation, LRRC58 is stabilized to permit CDO1 degradation. Through s…

The Fbxw11-Dpysl2 ubiquitin axis restrains the progression of acute myeloid leukemia.

Ubiquitination regulation is essential for maintaining protein homeostasis. Dysregulated ubiquitination is increasingly recognized to participate in malignant progression. Fbxw11, a substrate-recognition subunit of SCF-type E3 ligases, is involved in tumorigenesis and progression. However, its role and molecular mechanism in acute myeloid leukemia (AML) have not been elucidated. Here we demonstrat that lower levels of Fbxw11 were detected in AML patients and correlated with worse prognosis. Furthermore, overexpression of Fbxw11 delayed AML progression by reducing leukemia stem cell (LSC) frequ…
