Epstein-Barr virus-encoded latent membrane protein 1 impairs G2 checkpoint in human nasopharyngeal epithelial cells through defective Chk1 activation.

Nasopharyngeal carcinoma (NPC) is a common cancer in Southeast Asia, particularly in southern regions of China. EBV infection is closely associated with NPC and has long been postulated to play an etiological role in the development of NPC. However, the role of EBV in malignant transformation of nasopharyngeal epithelial cells remains enigmatic. The current hypothesis of NPC development is that premalignant nasopharyngeal epithelial cells harboring genetic alterations support EBV infection and expression of EBV genes induces further genomic instability to facilitate the development of NPC. The

Modulation of microglia and CD8(+) T cell activation during the development of stress-induced herpes simplex virus type-1 encephalitis.

The central nervous system (CNS) has been shown to be vulnerable to a variety of insults in animals exposed to glucocorticoids. For example, psychological stress, a known inducer of glucocorticoid production, enhances the susceptibility of mice to herpes simplex virus type-1 (HSV-1) infection and results in the development of HSV-1 encephalitis (HSE). To determine the immune mechanisms by which stress promotes the development of HSE, we examined the role of the glucocorticoid receptor (GR) and the N-methyl-d-aspartate (NMDA) receptor in the development of HSE. Our findings demonstrate that blo

Distinct mechanisms of CD4+ and CD8+ T-cell activation and bystander apoptosis induced by human immunodeficiency virus type 1 virions.

Apoptosis of uninfected bystander T cells contributes to T-cell depletion during human immunodeficiency virus type 1 (HIV-1) infection. HIV-1 envelope/receptor interactions and immune activation have been implicated as contributors to bystander apoptosis. To better understand the relationship between T-cell activation and bystander apoptosis during HIV-1 pathogenesis, we investigated the effects of the highly cytopathic CXCR4-tropic HIV-1 variant ELI6 on primary CD4(+) and CD8(+) T cells. Infection of primary T-cell cultures with ELI6 induced CD4(+) T-cell depletion by direct cell lysis and by

Epstein-Barr virus nuclear protein 2 interacts with p300, CBP, and PCAF histone acetyltransferases in activation of the LMP1 promoter.

The Epstein-Barr virus (EBV) nuclear protein 2 (EBNA2) and herpes simplex virion protein 16 (VP16) acidic domains that mediate transcriptional activation now are found to have affinity for p300, CBP, and PCAF histone acetyltransferases (HATs). Transcriptionally inactive point mutations in these domains lack affinity for p300, CBP, or PCAF. P300 and CBP copurify with the principal HAT activities that bind to EBNA2 or VP16 acidic domains through velocity sedimentation and anion-exchange chromatography. EBNA2 binds to both the N- and C-terminal domains of p300 and coimmune-precipitates from trans

Enhanced transcription of the s-adenosylhomocysteine hydrolase gene precedes Epstein-Barr virus lytic gene activation in ganglioside-stimulated lymphoma cells.

Stimulation of Epstein-Barr virus (EBV) genome-positive Burkitt lymphoma cells with the ganglioside IV3NeuAc-nLcOse4Cer leads to the induction of cell differentiation processes and activates the EBV lytic viral cycle. In cells of the Burkitt lymphoma line Raji differential expression of host cell genes was analysed in the early phase (150 min) post stimulation with the ganglioside to display the cell activities that precede the activation of the EBV lytic cycle using the differential display reverse transcription-polymerase chain reaction technique. Activation of a ras-MAPK-dependent pathway by Epstein-Barr virus latent membrane protein 1 is essential for cellular transformation.

The latent membrane protein 1 (LMP1) of Epstein-Barr virus (EBV) is the only EBV protein which possesses the properties of an oncogene. In studies initiated to evaluate the mechanisms involved in EBV-induced malignant transformation, the extracellular response kinase (ERK) 1/2 were found to be activated 2 days after EBV infection of purified resting human B cells. Transfection studies in Rat-1 fibroblasts, an established rodent cell line, showed that LMP1 mediates ERK 1/2 activation. Cotransfection experiments with a dominant negative ras mutant demonstrated that such MAPK activation occurs vi

Recurring proviral integration suggests a role for proto-oncogene activation in thymomas induced with Mo-MuLV-rescued BCR/ABL virus.

Intrathymic injection of Moloney murine leukemia virus (Mo-MuLV)-pseudotyped bcr-abl retrovirus (bcr-abl/M) causes thymic lymphoma but only after a prolonged latent period similar to that seen after intrathymic injection of Mo-MuLV alone. Since thymomas induced by Mo-MuLV show recurring proviral integration near certain cellular proto-oncogenes, it was reasoned that if the pathogenesis of bcr-abl/M thymomas is affected by viral integration, then it may be possible to detect proviral insertion near common Mo-MuLV integration sites in bcr-abl-induced thymomas. A panel of thymomas induced by intr

Establishment of latent ganglionic infection with herpes simplex virus via maxillary gingiva and viral re-activation in vivo after trauma.

Herpes simplex virus can remain latent for months or years in sensory and automatic ganglia of animals and man, and can be re-activated in vivo by several procedures such as neurectomy, irritation of epithelial surfaces, and administration of immunosuppressive agents. The objective of this study was to determine whether dental stimuli can cause re-activation of the latent herpes simplex virus. Homogenization and explanation of ganglia from mice showed that herpes simplex virus (type 1) traveled from maxillary gingiva to trigeminal ganglia, and remained latent. It was also shown that mice passi

Effect of activation of the Epstein-Barr virus genome on expression of B cell differentiation antigens of Burkitt's lymphoma lines.

Using anti-human B cell monoclonal antibodies prepared against B1 (CD20), B2 (CD21), B4 (CD19), and BB-1 (B lymphoblast antigen-1), we compared the expression of B cell differentiation antigens on a Jijoye-P3HR-1 cell line family of Burkitt's lymphomas. The expression of BB-1 and B2 antigens was faint on P3HR-1 K cell line which is an Epstein-Barr virus (EBV) high producer. On the other hand, B1 and B4 antigens were strongly expressed on it. It was also found that BB-1 expression decreased on P3HR-1 cells after activation of intracellular EBV genes by treating chemically with tumor-promoting a

trans-Activation of the human T-cell leukemia virus long terminal repeat correlates with expression of the x-lor protein.

Cell lines established directly from adult T-cell leukemia-lymphoma patients or immortalized by human T-cell leukemia virus type I (HTLV-I) in vitro that do not produce complete HTLV virions were characterized both for the content of viral proteins and for the presence of trans-acting factors activating gene expression under the control of the HTLV long terminal repeat. The expression of the 42-kilodalton HTLV x-lor product correlated with trans-activation of the long terminal repeat. The implications of this study for understanding the role of the HTLV x-lor product in the initiation and main

Multimerization of secretory IgA enhances antiviral activity by aggregating influenza A virus particles.

Secretory immunoglobulin A (sIgA), the first line of defense against mucosal epithelial infections, is a multimer. Although multimeric sIgA shows higher antiviral activity than monomeric IgA, the mechanism underlying this enhancement and the functional differences among IgA subclasses remain unclear. Here, we generated recombinant monomeric and multimeric IgA1 and IgA2m2 antibodies from clone F045-092, which targets the head domain of influenza A virus (IAV) hemagglutinin (HA) and inhibits viral entry, and compared their functional properties. Monomeric IgA1 inhibited viral entry but not viral

Kidney transplantation after chronic active Epstein-Barr virus infection remission.

Chronic active Epstein-Barr virus is a rare but severe condition characterized by persistent Epstein-Barr virus activation, leading to chronic lymphoproliferation, significant morbidity, and mortality. We report the case of a 34-year-old male with autosomal dominant polycystic kidney disease who achieved chronic active Epstein-Barr virus remission through hematopoietic stem cell transplantation in childhood. Despite long-term remission, the patient developed end-stage renal disease and underwent ABO-incompatible living-donor kidney transplantation. Post-transplant monitoring over 7 months reve…

Quercetin exhibits antiviral activity against duck Tembusu virus in vitro and in vivo.

Duck Tembusu virus (DTMUV) is an avian pathogenic flavivirus that causes substantial economic losses to the poultry industry in China and Southeast Asia. The current antiviral drugs for the prevention and treatment of DTMUV infection are limited. Quercetin is a natural flavonoid widely found in fruits and vegetables and has been reported to exhibit antiviral activity against many viruses. However, whether quercetin possesses antiviral activity against DTMUV remains unknown. Therefore, the present study aimed to investigate the antiviral efficacy of quercetin against DTMUV. IFIT3 RNA-binding activity promotes influenza A virus infection and translation efficiency.

Host cells produce a vast network of antiviral factors in response to viral infection. The interferon-induced proteins with tetratricopeptide repeats (IFITs) are important effectors of a broad-spectrum antiviral response. In contrast to their canonical roles, we previously identified IFIT2 and IFIT3 as pro-viral host factors during influenza A virus (IAV) infection. During IAV infection, IFIT2 binds and enhances translation of AU-rich cellular mRNAs, including many IFN-stimulated gene products, establishing a model for its broad antiviral activity. However, IFIT2 also binds viral mRNAs and enh

Taurultam shows antiviral activity against SARS-CoV-2 and influenza virus.

BACKGROUND: SARS-CoV-2 and influenza virus are highly contagious respiratory viruses that continuously pose major threats to human and public health. The high frequency of viral mutations led to the emergence of resistant isolates and caused virus epidemics repeatedly, emphasizing the urgent need to develop new antivirals. Taurultam is a metabolite of taurolidine. Moreover, taurolidine has been shown to have potent antiviral activities against multiple viruses and to have antiviral effects through its metabolites. RESULTS: In this study, we sought to determine the antiviral activities of taur

Antiviral activity of newly synthesized pyrazole derivatives against Newcastle disease virus.

Newcastle disease virus (NDV) is a highly contagious poultry disease that affects the respiratory, nervous, and digestive systems, causing significant losses to the poultry industry. Pyrazole-based scaffolds had significant potential as antiviral agents targeting various pathogens. Thus, a series of 4-substituted pyrazole derivatives were synthesized by reacting 5-chloro-4-formyl-3-methyl-1-phenylpyrazole with some nitrogen and carbon-based nucleophiles. The antiviral efficacy of these compounds was evaluated against NDV by assessing their ability to inhibit virus-induced haemagglutination. No

Antiviral activity of eicosapentaenoic acid against zika virus and other enveloped viruses.

BACKGROUND: Zika virus (ZIKV) is an emerging flavivirus that may cause innate microcephaly or neurological disturbances. Yet no antiviral has been approved by FDA against ZIKV infection. It was shown that some unsaturated fatty acids could inactivate enveloped viruses including SARS-CoV-2. However, studies investigating the effect of eicosapentaenoic acid (EPA) on ZIKV infection are lacking. This study aims to evaluate the antiviral effect of EPA against ZIKV and other enveloped viruses. METHODS: We first explored the toxicities of EPA in vitro and in vivo. Then we examined the antiviral effe

Antiviral Activity of Ethyl Gallate Against Zika Virus: In Vitro and In Silico Studies.

Zika virus (ZIKV) remains a significant global public health concern, and growing resistance to existing antiviral drugs underscores the necessity of developing alternative therapeutic options. In this study, we investigated the inhibitory effects of ethyl gallate against ZIKV using antiviral activity evaluation, molecular docking, and molecular dynamic simulations. Treatment of ZIKV-infected Vero E6 cells with ethyl gallate resulted in dose-dependent suppression of viral infection without inducing cytotoxicity. In addition, ethyl gallate inhibited the increase in the expression of interferon-
