E-cadherin to P-cadherin switching in lobular breast cancer with tubular elements.

Loss of E-cadherin expression due to mutation of the CDH1 gene is a characteristic feature of invasive lobular breast cancer (ILBC). Beta-catenin, which binds to the cytoplasmic domain of E-cadherin, is simultaneously downregulated, reflecting disassembly of adherens junctions (AJs) and loss of cell adhesion. E-cadherin to P-cadherin expression switching can rescue AJs and cell adhesion. However, P-cadherin has not been implicated in ILBC, so far. We aimed to characterize 13 ILBCs with exceptional histomorphology, which we termed ILBCs with tubular elements. The CDH1 mutational status was dete…

The glucosyltransferase Xiantuan of the endoplasmic reticulum specifically affects E-Cadherin expression and is required for gastrulation movements in Drosophila.

The majority of membrane and secreted proteins, including many developmentally important signalling proteins, receptors and adhesion molecules, are cotranslationally N-glycosylated in the endoplasmic reticulum. The structure of the N-glycan is invariant for all substrates and conserved in eukaryotes. Correspondingly, the enzymes are conserved, which successively assemble the glycan precursor from activated monosaccharides prior to transfer to nascent proteins. Despite the well-defined biochemistry, the physiological and developmental role of N-glycosylation and of the responsible enzymes has n

Ninjurin2 Promotes Pulmonary Endothelial Barrier Dysfunction Through VEGF/VEGFR2 Signaling Pathway.

Previous studies have established an association between Ninjurin2 and diseases related to vascular endothelial dysfunction, such as ischemic stroke and coronary heart disease. Given that impaired pulmonary endothelial barrier function is a critical feature of acute lung injury (ALI) and acute respiratory distress syndrome (ARDS), we hypothesized that Ninjurin2 may also participate in the regulation of lung vascular permeability. Therefore, this study investigates the role of Ninjurin2 in regulating endothelial cell function and dysfunction, aiming to elucidate the underlying mechanisms in ALI

Mechanobiological modulation: physiological pressure preconditioning boosts the paracrine-driven lymphangiogenic capacity of bone marrow mesenchymal stem cells.

INTRODUCTION: Lymphatic system remodeling and regeneration are essential for tissue repair and homeostasis, and these processes are precisel … ted that P-B-CM strengthened intercellular barrier integrity by increasing the length of continuous adherens junctions. Consistent with the in vitro cellular findings, ex vivo rat thoracic duct culture further verified that P-B-CM induced more robust lymphatic sprouting, confirming its promotive effects on lymphangiogenesis. CONCLUSION: Mechanical pressure precondi …

Apico-basal polarity in polycystic kidney disease epithelia.

Epithelial cell polarity is essential for the establishment and maintenance of morphological and functional asymmetries that underlie normal … ity cues. Specialized structural domains between adjacent cells and cells with their matrix, termed adherens junctions (AJ) and focal adhesions (FA), respectively, are formed that contain specific components of multi-molecular complexes acting as sites to recruit proteins and to activate intracellular mechano-transduction pathways. Regulation of these processes resu …

Prostaglandin E receptor-4 receptor mediates endothelial barrier-enhancing and anti-inflammatory effects of oxidized phospholipids.

Unlike other agonists that cause transient endothelial cell (EC) response, the products of 1-palmitoyl-2-arachidonoyl-sn-glycero-3-phosphocholine (PAPC) oxidation that contain cyclopenthenone groups, which recapitulate prostaglandin-like structure, cause sustained enhancement of the pulmonary EC barrier. The mechanisms that drive the sustained effects by oxidized PAPC (OxPAPC) remain unexplored. On the basis of the structural similarity of isoprostanoid moieties that are present in full-length oxygenated PAPC species, we used an inhibitory approach to perform the screening of prostanoid recept

Catenins in the rat epididymis: their expression and regulation in adulthood and during postnatal development.

Tight and adhering junctions are important in maintaining the integrity of the epididymal epithelium and formation of the blood epididymal barrier, which are crucial for sperm maturation and storage. The composition of the catenin-adhering junctional family of proteins and their relationship with tight junctions remain to be established in the epididymis. In the normal adult rat epididymis, immunostaining for three anticatenin antibodies (alpha, beta-, and p120ctn) was noted along the lateral plasma membranes (LPM) between adjacent epithelial cells. Although alpha-catenin and beta-catenin were

Pseudorabies virus infection increases the permeability of the mammalian respiratory barrier to facilitate Pasteurella multocida infection.

UNLABELLED: Interaction between viruses and bacteria during the development of infectious diseases is a complex question that requires continuous study. In this study, we explored the interactions between pseudorabies virus (PRV) and Pasteurella multocida (PM), which are recognized as the primary and secondary agents of porcine respiratory disease complex (PRDC), respectively. In vivo tests using mouse models demonstrated that intranasal inoculation with PRV at a sublethal dose induced disruption of murine respiratory barrier and promoted the invasion and damages caused by PM through respirato

Integrative Transcriptomic and Genomic Analysis Reveals the Regulatory Mechanisms Underlying Semen Quality Stability in Ganders.

Semen quality significantly affects the reproductive performance of ganders. As seasonal breeders, ganders typically experience a decline in semen quality over the breeding season; however, the mechanisms regulating semen quality stability during this period remain poorly understood. Maintaining high semen quality throughout the entire breeding season is crucial for improving production efficiency. In this study, we systematically investigated the semen quality parameters of Tianfu Meat Goose II ganders at three distinct stages of the female geese's annual laying cycle: early (36-week-old), pe

Endogenous EGF maintains Sertoli germ cell anchoring junction integrity and is required for early recovery from acute testicular ischemia/reperfusion injury.

Administration of exogenous epidermal growth factor (EGF) improves testicular injury after acute ischemia-reperfusion (IR) stress, but the molecular basis is poorly understood. The role of endogenous EGF in testicular recovery and the underlying intracellular signaling pathways involved were herein investigated. In mice, testicular IR injury significantly enhanced the expression level of endogenous Egf at the very beginning of reperfusion. Expression of EGF receptor (Egfr (ErbB1)) was accordingly upregulated 3  h after reperfusion. Deprivation of majority of circulated EGF by sialoadenectomy a

A preclinical study of device dependent therapeutic effects of cold atmospheric plasmas on atopic dermatitis induced by DNCB.

Atopic dermatitis (AD) is a skin disease resulting from a complex interplay between genetic factors, environmental influences, microbiota, skin barrier dysfunction, and immune system disorders. Despite the existence of various AD therapies, it is necessary to develop new therapies for AD treatment. Cold atmospheric plasma (CAP) is a promising non-invasive therapeutic alternative in dermatology. This study aims to investigate the device-dependent effects of CAP sources in treating a 1-chloro-2,4-dinitrobenzene(DNCB)-induced AD model in Balb/c mice, comparing its effects with an immunosuppressiv

Apolipoprotein M: Structural insights, functional roles, and therapeutic approaches in vascular disease.

Apolipoprotein M (ApoM) is a lipocalin predominantly associated with high-density lipoprotein (HDL) that transports sphingosine-1-phosphate (S1P) in circulation. Through its stable binding and selective delivery of S1P to the endothelial S1P receptors (S1PRs), ApoM orchestrates a spectrum of vasoprotective effects. This review summarizes the structural characteristics of ApoM and its unique function as a sphingolipid chaperone, focusing on its role in vascular biology, specifically endothelial barrier integrity, vascular tone, and inflammation. We examine the biased signaling of ApoM-HDL-S1P t

Abnormal Splicing in the Final Intron of PRX Results in Dominant Congenital Cataract Without Neurological Phenotype.

PURPOSE: Disrupted lens development can result in congenital cataract. Despite identifying over 50 genes linked to non-syndromic pediatric cataract, a causative mutation is found in only about 50% of affected families. METHODS: Exome and/or genome sequencing was undertaken in families with unexplained congenital cataract. RESULTS: Sequencing revealed heterozygous variants affecting splicing of the final intron of PRX in seven individuals from four families. PRX encodes periaxin, a protein necessary for the maintenance of peripheral nerve myelin and the proper organization of the lens cortex

N-cadherin: A diagnostic marker to help discriminate primary liver carcinomas from extrahepatic carcinomas.

Distinguishing primary liver cancer (PLC), namely hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (iCCA), from liver metastases is of crucial clinical importance. Histopathology remains the gold standard, but differential diagnosis may be challenging. While absent in most epithelial, the expression of the adherens junction glycoprotein N-cadherin is commonly restricted to neural and mesenchymal cells, or carcinoma cells that undergo the phenomenon of epithelial-to-mesenchymal transition (EMT). However, we recently established N- and E-cadherin expression as hallmarks of norm

ADAM10-Mediated Ectodomain Shedding Is an Essential Driver of Podocyte Damage.

BACKGROUND: Podocytes embrace the glomerular capillaries with foot processes, which are interconnected by a specialized adherens junction to ultimately form the filtration barrier. Altered adhesion and loss are common features of podocyte injury, which could be mediated by shedding of cell-adhesion molecules through the regulated activity of cell surface-expressed proteases. A Disintegrin and Metalloproteinase 10 (ADAM10) is such a protease known to mediate ectodomain shedding of adhesion molecules, among others. Here we evaluate the involvement of ADAM10 in the process of antibody-induced pod
