Matrix Metalloproteinase-3 -1171 5A/6A Polymorphism (rs35068180) is Associated with Risk of Periodontitis.

Matrix metalloproteinase-3 (MMP3) plays a key role in tissue degradation in periodontitis. The relationship between the MMP3 -1171 5A/6A pol … -smokers subgroups. Moreover, MMP3 rs35068180 polymorphism might be associated with a lower risk of aggressive periodontitis (AgP) in Asians (allelic genetic model: OR = 0.66, 95% CI: 0.48-0.91, P(heterogeneity) = 0.945), and CP in Caucasians and Brazilians. In conclusion, this meta-analysis demonstrates that MMP3 -1171 5A/6A polymorphism may be associated with decreased …

Subgingival human cytomegalovirus correlates with increased clinical periodontal parameters and bacterial coinfection in periodontitis.

BACKGROUND: Viruses from the Herpesviridae family may be implicated in the pathogenesis of periodontal disease. The aim of this investigation was to compare the subgingival frequency of human cytomegalovirus (HCMV) in subjects affected by periodontitis to periodontally healthy subjects and to assess the correlation of HCMV with periodontal clinical parameters and periodontopathic bacteria. METHODS: Thirty subjects with periodontitis (20 with chronic periodontitis and 10 with aggressive periodontitis) were included in the study. A group of 22 periodontally healthy individuals served as control…

Variance component modeling of attachment level measurements.

The purpose of this study was to investigate the influence of within subject and within tooth variability of attachment-level measurements on hypothesis testing. Full-mouth attachment-level measurements were obtained at 4 sites per tooth in early onset periodontitis subjects (both localized N = 89 and generalized N = 139) and adult periodontitis subjects (N = 309). Variance component models utilizing iterative generalized least squares were employed to estimate the % of variance distributed at the level of the site, tooth and subject in the 3 subject populations. The data indicate that a signi

Different contribution of BRINP3 gene in chronic periodontitis and peri-implantitis: a cross-sectional study.

BACKGROUND: Peri-implantitis is a chronic inflammation, resulting in loss of supporting bone around implants. Chronic periodontitis is a risk indicator for implant failure. Both diseases have a common etiology regarding inflammatory destructive response. BRINP3 gene is associated with aggressive periodontitis. However, is still unclear if chronic periodontitis and peri-implantitis have the same genetic background. The aim of this work was to investigate the association between BRINP3 genetic variation (rs1342913 and rs1935881) and expression and susceptibility to both diseases. METHODS: Perio…

The correlation of gingival crevicular fluid interleukin-8 levels and periodontal status in localized juvenile periodontitis.

Localized juvenile periodontitis (LJP) is an early-onset periodontal disease characterized by progressive bone loss involving the permanent first molar and incisor teeth. Approximately 70% to 75% of LJP patients have impaired neutrophil chemotaxis towards a number of chemoattractants including N-formyl-methionyl-leucyl-phenyl-alanine, complement fragment C5a, leukotriene B4, and interleukin 8 (IL-8). The aim of the present study was to observe the role of IL-8 in the pathogenesis of LJP. Fourteen individuals who were systemically and periodontally healthy and 24 systemically healthy individual

Progressive cervical root resorption related to tetracycline root conditioning.

Root resorption is reported as a microscopic finding in trials attempting to regenerate the periodontium using tetracycline or citric acid root conditioning. This report deals with a case of progressive cervical root resorption in a female patient who had been successfully treated by tetracycline root conditioning. The article emphasizes the possibility of this adverse effect, and discusses a possible mechanism inducing this phenomenon.

No female preponderance in juvenile periodontitis after correction for ascertainment bias.

Juvenile periodontitis (JP) is generally recognized to exist in 2 clinical forms: localized and generalized. Historically, females have been reported to be affected by both forms of JP at rates of 2 to 10 times greater than males. However, evidence suggests that females are more likely than males to seek dental care. If this is true, females will be diagnosed with JP more often than males even if juvenile periodontitis is equally prevalent among males and females in the general population. Thus, previous reports of a female predominance for JP may simply reflect this selection bias. The purpos

Diagnosis and classification of the periodontal diseases.

A classification for gingivitis and periodontitis has been proposed based on clinical observations and immunologic parameters (summarized in Table 10). Overlapping clinical situations and exceptions to the rule certainly exist and pose challenges to clinicians during diagnosis, prognosis, and treatment planning. The clinical significance of making the diagnosis of either rapidly progressive periodontitis, juvenile, or prepubertal forms of periodontitis compared with adult periodontitis must be considered. A diagnosis of early-onset disease may (1) modify the periodontal treatment plan and may

Comparison between 2 surgical techniques for the treatment of early-onset periodontitis.

The objective of this study was to compare guided tissue regeneration (GTR) and osseous surgery (OS) in treatment of early-onset periodontitis. Six patients participated in this research, presenting 15 paired defects randomly assigned to 1 of the surgical groups. Pre-operative clinical parameters (probing depth and attachment loss) between the two groups were not significantly different. Results 1 year following surgery showed probing depth reduction (2.60 +/- 1.30 mm) and clinical attachment gain (2.20 +/- 1.42 mm) were significantly greater in the GTR group than in the osseous surgery group

Genetic approach to the study of epidemiology and pathogenesis of Actinobacillus actinomycetemcomitans in localized juvenile periodontitis.

Actinobacillus actinomycetemcomitans isolates from periodontal pockets were examined for restriction fragment-length polymorphism using a characterized 4.7-kb DNA probe. A total of 6 patterns of RFLP was found in 133 isolates originating from 12 subjects. No relatedness was found between RFLP types and serotypes. Different periodontal sites within the same subject and different individuals within the same family sometimes showed only one type of A. actinomycetemcomitans RFLP. When members among the same family showed 2 RFLP types, children were always infected with the A. actinomycetemcomitans

The decreased antibacterial activity of oral polymorphonuclear leukocytes coincides with the occurrence of virus-carrying oral lymphocytes and epithelial cells.

Patients suffering from periodontitis or periodontosis were selected for the study. Further subdivision of these groups was based on the presence or absence of herpes and/or adenoviruses in their oral lymphocytes and epithelial cells. The phagocytic and bactericidal activities of oral leukocytes isolated from the same individuals were compared with virus carriage. In the periodontitis group, 60.5%, and in the periodontosis group 61.5% of patients carried viruses, while this was established only in 21.1% of control cases. On the other hand, emigration and sulcular gathering of the less viable p

Enamel-renal-gingival syndrome and FAM20A mutations.

The enamel-renal syndrome of amelogenesis imperfecta (AI) and nephrocalcinosis, and the amelogenesis imperfecta-gingival fibromatosis syndrome have both been associated with mutations in FAM20A. We report on two unrelated Thai patients with three novel and one previously reported mutations in FAM20A with findings suggesting both disorders, including hypoplastic AI, gingival fibromatosis, unerupted teeth, aggressive periodontitis, and nephrocalcinosis/nephrolithiasis. Additional findings consisted of a supernumerary premolar, localized aggressive periodontitis, thin alveolar bone, vitamin D def…

Mechanisms Involved in the Association between Periodontitis and Complications in Pregnancy.

The association between periodontitis and some of the problems with pregnancy such as premature delivery, low weight at birth, and preeclampsia (PE) has been suggested. Nevertheless, epidemiological data have shown contradictory data, mainly due to differences in clinical parameters of periodontitis assessment. Furthermore, differences in microbial composition and immune response between aggressive and chronic periodontitis are not addressed by these epidemiological studies. We aimed to review the current data on the association between some of these problems with pregnancy and periodontitis,

Histological differences between teeth with adult periodontitis and prepubertal periodontitis.

Thirty-nine permanent teeth from three siblings aged 11 to 14 years were extracted because of generalized prepubertal periodontitis (PP) and studied histologically with light microscopy. Twenty healthy teeth (HT) removed for orthodontic procedures from 10 children of similar ages and 22 teeth affected by adult periodontitis (AP) extracted from patients over 50 years old were used as controls. The cementum of the PP teeth was much thicker than that of the healthy teeth. A high percentage of resorption, unrelated to inflammation, was found in all the teeth studied. In 24 PP teeth the cementum wa

Microscopic characterization of root surface-associated microbial plaque in localized juvenile periodontitis.

The purpose of this investigation was to characterize by scanning electron microscopy, the microbial morphotypes associated with the roots of teeth from patients exhibiting localized juvenile periodontitis (LJP). Eighteen teeth were examined from 10 adolescent patients who satisfied the standard clinical diagnostic criteria for LJP. The microscopic evaluation was facilitated by dividing that portion of the root exposed to the periodontal pocket into coronal, middle, and apical one-thirds. Controls consisted of an equal number of teeth matched as to pocket depth and anatomical type obtained fro

Clinical, microbiological and immunological features associated with the treatment of active periodontosis lesions.

Clinical, microbiological and immunological factors were examined using data from a subject with periodontosis. The subject was monitored at bimonthly intervals for 26 months at 6 sites per tooth for redness, plaque, suppuration, bleeding on probing, pocket depth, and attachment level. Using attachment level measurements and the tolerance method of analysis, sites with active disease and control (inactive) sites of equal pocket depth were selected. Subgingival plaque samples were taken from these sites for predominant cultivable and dark field evaluation before, and 5 and 13 months after treat

A six-site method for the evaluation of periodontal bone loss in cone-beam CT images.

OBJECTIVES: In contrast to two-dimensional planar images, a measuring point is hardly repeatedly determined in a CBCT image when alveolar bone loss is assessed. Thus, the aim of the present study was to propose a six-site measuring method, which is closely related to anatomical structure, for the evaluation of alveolar bone loss in CBCT images. METHODS: 150 measuring points in 11 molars and 14 premolars from 6 patients (2 males and 4 females) were included. CBCT images of the teeth were acquired prior to periodontal surgery. Four observers measured the distances between cemento-enamel junctio

Periodontitis: Consensus report of workgroup 2 of the 2017 World Workshop on the Classification of Periodontal and Peri-Implant Diseases and Conditions.

A new periodontitis classification scheme has been adopted, in which forms of the disease previously recognized as "chronic" or "aggressive" are now grouped under a single category ("periodontitis") and are further characterized based on a multi-dimensional staging and grading system. Staging is largely dependent upon the severity of disease at presentation as well as on the complexity of disease management, while grading provides supplemental information about biological features of the disease including a history-based analysis of the rate of periodontitis progression; assessment of the risk
