Hydatid disease: research and control in Turkana, III. Albendazole in the treatment of inoperable hydatid disease in Kenya--a report on 12 cases.

The results of treatment of 12 cases of inoperable hydatid disease in Kenya using albendazole at a dose of 10 mg/kg body-weight for eight weeks are reported. In all the cases there was marked regression and collapse of the cysts but a further four weeks course of treatment at a higher dose of 15 mg/kg was given to two cases because they both still had a large number of demonstrable cysts on ultrasonography at eight weeks. No side effects of the drug were noted. Albendazole is likely to play an important role in the control programme in Turkana.

Human leukocyte extract enhances albendazole efficacy against Echinococcus multilocularis in mice by reducing parasitic microRNAs, myeloid and lymphoid immunosuppressive cells.

BACKGROUND: Echinococcus multilocularis induces a serious zoonotic infection in humans and growth of metacestodes is associated with immunosuppression and tolerance. Current treatment relies almost exclusively on albendazole, a parasitostatic drug with variable clinical efficacy. Human low molecular weight leukocyte extract (HLE) has been shown to modulate innate and adaptive immunity at various diseases and can serve as promising and safe adjuvant to chemotherapy. METHODOLOGY: In the present study we investigated the effects of albendazole (ABZ), human leukocyte extract (HLE) and their combi…

Safety Surveillance of Mass Praziquantel and Albendazole Co-Administration in School Children from Southern Ethiopia: An Active Cohort Event Monitoring.

Preventive chemotherapy (PC) with praziquantel and albendazole co-administration to all at-risk populations is the global intervention strategy to eliminate schistosomiasis and soil-transmitted helminth (STH) from being public health problems. Due to weak pharmacovigilance systems, safety monitoring during a mass drug administration (MDA) is lacking, especially in sub-Saharan Africa. We conducted large-scale active safety surveillance to identify the incidence, types, severity, and associated risk factors of adverse events (AEs) following praziquantel and albendazole MDA in 5848 school childre…

Safety of integrated mass drug administration of azithromycin, albendazole and ivermectin versus standard treatment regimens: a cluster-randomised trial in Ethiopia.

BACKGROUND: Neglected Tropical Disease (NTD) programs require separate and distinct drug regimens for treatment. This has required countries to undertake multiple independent mass drug administration (MDA) programmes, each targeting one or more diseases. The possibility of safely combining different drug regimens together in one MDA may offer several advantages to national programs. We conducted a study to assess the safety of combining ivermectin, albendazole and azithromycin in one integrated MDA. METHODS: We conducted an open-label, non-inferiority cluster-randomised trial comparing the fr…

Albendazole Microcrystal Formulations Based on Chitosan and Cellulose Derivatives: Physicochemical Characterization and In Vitro Parasiticidal Activity in Trichinella spiralis Adult Worms.

The oral route has notable advantages to administering dosage forms. One of the most important questions to solve is the poor solubility of most drugs which produces low bioavailability and delivery problems, a major challenge for the pharmaceutical industry. Albendazole is a benzimidazole carbamate extensively used in oral chemotherapy against intestinal parasites, due to its extended spectrum activity and low cost. Nevertheless, the main disadvantage is the poor bioavailability due to its very low solubility in water. The main objective of this study was to prepare microcrystal formulations …

Albendazole inhibits endothelial cell migration, tube formation, vasopermeability, VEGF receptor-2 expression and suppresses retinal neovascularization in ROP model of angiogenesis.

The angiogenic process begins with the cell proliferation and migration into the primary vascular network, and leads to vascularization of previously avascular tissues and organs as well to growth and remodeling of the initially homogeneous capillary plexus to form a new microcirculation. Additionally, an increase in microvascular permeability is a crucial step in angiogenesis. Vascular endothelial growth factor (VEGF) plays a central role in angiogenesis. We have previously reported that albendazole suppresses VEGF levels and inhibits malignant ascites formation, suggesting a possible effect …

Coacervated and Freeze-Dried Polysaccharides-Nanoparticle with Efficient Encapsulation of Albendazole for High-Performance Treatment of Monogenean Parasite Infestation in Tilapia Fish.

Monogenean parasite infestation in fish leads to economic losses in aquaculture, representing a veterinary challenge and an environmental concern. The common administration procedures of anthelmintics to treat monogeneans in fish have low efficiency and diverse drawbacks. In this study, we produced a nanoparticle using chitosan and alginate, biodegradable and biocompatible polysaccharides, as an oral drug delivery material of albendazole anthelmintic for parasite-infected fingerlings of Nile tilapia. The molecular interaction between the biopolymers was optimized and characterized by titration…
