Bacterial mutualists can modulate the biochemical capacity of animals. Highly coevolved nutritional mutualists do this by synthesizing nutrients missing from the host's diet. Genomics tools have advanced the study of these partnerships. Here we examined the endosymbiont Xiphinematobacter (phylum Verrucomicrobia) from the dagger nematode Xiphinema americanum, a migratory ectoparasite of numerous crops that also vectors nepovirus. Previously, this endosymbiont was identified in the gut, ovaries, and eggs, but its role was unknown. We explored the potential role of this symbiont using fluorescenc

We have used site-directed and in-frame deletion mutational analysis in order to explore the structural features of the IL-6 portion of the diphtheria toxin-related interleukin-6 (IL-6) fusion toxin DAB389-IL-6 that are essential for receptor-binding and subsequent inhibition of protein synthesis in target cells. Deletion of the first 14 amino acids of the IL-6 component of the fusion toxin did not alter either receptor binding affinity or cytotoxic potency. In contrast, both receptor binding and cytotoxic activity were abolished when the C-terminal 30 amino acids of the fusion toxin were dele

IDO is the rate-limiting enzyme in the kynurenine pathway, catabolizing tryptophan to kynurenine. Tryptophan depletion by IDO-expressing tumors is a common mechanism of immune evasion inducing regulatory T cells and inhibiting effector T cells. Because mammalian cells cannot synthesize tryptophan, it remains unclear how IDO(+) tumor cells overcome the detrimental effects of local tryptophan depletion. We demonstrate that IDO(+) tumor cells express a novel amino acid transporter, which accounts for ∼50% of the tryptophan uptake. The induced transporter is biochemically distinguished from the co

Soybean and pea protein isolates contain essential amino acid that may support bone health, skeletal function and muscle performance. The current research examines the amino acid profile of SPI and PPI and evaluates their impact on bone health in vivo and in silico. Female Sprague-Dawley rats were randomly assigned to four groups: T0 (Standard diet + without osteoporosis), T1 (Standard diet + osteoporosis), T2 (2 g/kg bw of SPI+ 2 g/kg bw of PPI), and T3 (3 g/kg bw of SPI+ 3 g/kg bw of PPI). Biochemical parameters and histopathology of bone (femur and tibia), liver, and kidney were evaluated.

Urea synthesis is an irreversible, essential for maintenance of health and life, and highly regulated liver function with a very high capacity for production of the end-product urea-nitrogen. The set-point of urea synthesis in relation to its overall substrate, the prevailing blood concentration of L-α-amino acids, contributes to determine whole-body nitrogen balance and the size and composition of the plasma free amino acid pool. Ammonia is definitively eliminated from the body by urea synthesis. Ammonia is released by all tissues as part of their nitrogen metabolism and accumulation of ammon

BACKGROUND: Celiac disease (CeD) is an autoimmune condition characterized by an aberrant immune response triggered by the ingestion of gluten, which damages epithelial cells lining the small intestine. Small intestinal epithelial cells (sIECs) play key roles in the enzymatic digestion and absorption of nutrients, maintaining gut barrier integrity, and regulating immune response. Chronic inflammation and tissue damage associated with CeD disrupt the intricate network of metabolic processes in sIECs that support these functions, impairing their ability to perform their essential roles. However,

Methotrexate (MTX) is commonly used as a therapeutic agent in the treatment of malignancies and autoimmune disorders. Risk of subsequent infertility following MTX administration has been reported as a significant side effect due to testicular toxicity. The aim of the study was to evaluate the modulatory effects of Ginkgo biloba (standardized extract, EGb 761) on MTX-induced testicular oxidative stress, energy deficits and spermatogenic status in rats. All groups received intraperitoneal injection of MTX (0.5 mg/kg) twice weekly for four weeks except the control group that received the correspo

UNLABELLED: The fungus Candida albicans is a frequent colonizer of humans but also an opportunistic pathogen causing superficial to severe infections, especially in vulnerable individuals. Its broad metabolic flexibility is key for the fungal adaptation to host environments, evasion from immune attack, and virulence. Amino acid metabolism and homeostasis, in particular, are critical for fungal fitness-illustrated by a rapid metabolic shift in response to amino acid starvation to restore intracellular metabolic balance. To investigate the cellular mechanisms underlying such compensatory metabol

Tuberculosis (TB) remains a global health threat aggravated by the emergence of drug-resistant strains. Mycobacterium tuberculosis promotes metabolic remodeling within host cells to support its survival. Type II alveolar epithelial cells have been established as a primary niche for bacterial infection, adherence, and invasion. However, compared to macrophages, metabolic interactions within these cells, particularly the strain-specific exploitation of host metabolism, remain inadequately explored. To address this, we integrated intracellular bacterial growth and host cell viability assays with

The proline-rich Akt substrate of 40 kilodaltons (PRAS40) was identified as a raptor-binding protein that is phosphorylated directly by mammalian target of rapamycin (mTOR) complex 1 (mTORC1) but not mTORC2 in vitro, predominantly at PRAS40 (Ser(183)). The binding of S6K1 and 4E-BP1 to raptor requires a TOR signaling (TOS) motif, which contains an essential Phe followed by four alternating acidic and small hydrophobic amino acids. PRAS40 binding to raptor was severely inhibited by mutation of PRAS40 (Phe(129) to Ala). Immediately carboxyl-terminal to Phe(129) are two small hydrophobic amino ac

We established six T cell clones specific for pyruvate dehydrogenase complex (PDC)-E2 peptides from four different patients with primary biliary cirrhosis using 33 different peptides of 17-20 amino acid residues corresponding to human PDC-E2 as stimulating antigens. The minimal T cell epitopes of these six T cell clones were all mapped to the same region of the PDC-E2 peptide 163-176 (GDLLAEIETDKATI), which corresponds to the inner lipoyl domain of PDC-E2. The HLA restriction molecules for this epitope were all identified as HLA DRB4 0101. The common essential amino acids of this epitope for t

OBJECTIVE: Plasmablasts play important roles in autoimmune diseases, including systemic lupus erythematosus (SLE). Activation of mechanistic target of rapamycin complex 1 (mTORC1) is regulated by amino acid levels. In patients with SLE, mTORC1 is activated in B cells and modulates plasmablast differentiation. However, the detailed mechanisms of amino acid metabolism in plasmablast differentiation remain elusive. We undertook this study to evaluate the effects of methionine in human B cells.
METHODS: Purified CD19+ cells from healthy donors (n = 21) or patients with SLE (n = 35) were cultured

Ferroptosis is a newly identified form of cell death that is regulated by many metabolic pathways, including iron, lipid and amino acids. Recent two studies reveal that the mitochondria and energy stress could also mediate ferroptosis. Gao et al. report that mitochondria play an essential role in ferroptosis induced by cysteine deprivation. In addition, Lee et al. show that energy stress depressed ferroptosis partly through activation of AMP-activated protein kinase. These findings provide potential therapeutic strategies for treating ferroptosis-related diseases, such as cancer, tissue injury

A male patient, born in 1999, was diagnosed with ornithine transcarbamylase deficiency as neonate and was managed with a strict low-protein diet supplemented with essential amino acids, L-citrulline, and L-arginine as well as sodium benzoate. He had an extensive history of hospitalizations for hyperammonemic crises throughout childhood and early adolescence, which continued after the addition of sodium phenylbutyrate in 2009. In December 2013 he was switched to glycerol phenylbutyrate, and his metabolic stability was greatly improved over the following 7 months prior to liver transplant.

Halitosis is a very common condition which may affect up to 30% of the population. In about 90% of the cases, halitosis originates in the mouth due to inadequate plaque control, periodontal disease, dry mouth, faulty restorations, and in particular due to excessive bacterial growth. Oral malodor is mainly caused by a microbial degradation of amino acids into volatile, bad-smelling gases (volatile sulfur compounds - VSCs). Management of oral malodor is directed primarily at managing and reducing the VSC-producing bacteria count as well as masking the odor. Essential oils have been used for this

We have determined the nucleotide sequence of a segment of chromosome III contained in the right part of the lambda PM3270 clone, for a total of 8824 bp. This sequence contains an unusual long open reading frame, YCR601, of 6501 bp that encodes for a protein of 2167 amino acids that show no homology with other known proteins. YCR601 was disrupted by internal deletion and insertion of LEU2 gene and is a non-essential gene, however, it is transcribed during vegetative growth yielding a polyadenylated mRNA of approximately 7 kb.

It has been shown that in rats with alimentary hypovitaminosis A, with a sufficiently pronounced decrease in the content of free retinol in blood serum (below 140 ng/ml) and in the liver tissue (below 360 ng/g), there is a complete regression of Pliss's lymphosarcoma. Under the conditions indicated, retinoic acid, in contradistinction to retinol, does not promote the tumor growth. A relative and absolute reduction in blood plasma of the essential amino acids, arginine, methionine and lysine, might be one of the reasons for tumor regression under hypovitaminosis A.
