Developmental exposure to methylmercury alters behavioral sensitivity to D-amphetamine and pentobarbital in adult rats.

Female rats were exposed to 0, 0.5, or 6.4 ppm methylmercury in their drinking water before mating, and throughout gestation and lactation. When the female offspring were 4-6 months old, they were trained to respond under a multiple differential reinforcement of high rate (DRH) 9:4-- Extinction schedule of reinforcement. No differences among exposure groups were apparent in steady-state behavior. Drug challenges were conducted with multiple doses of D-amphetamine, scopolamine, pentobarbital, haloperidol, and dizocilpine, drugs selected for their different pharmacological effects. The ED(50) va…

Changes in the performance of schedule-induced polydipsia (SIP) in rats after arecoline and amphetamine treatments.

This study investigated the performance of schedule-induced polydipsia (SIP) rats in novel or intermittent-reward SIP sessions after arecoline (AREC) and amphetamine sulfate (AMPH) treatments. Either automatic monitors or observers extensively examined the functional changes of parameters in behavioral performance followed by increasing drug dosage. The parameters included locomotion and stereotyped behaviors in the novel sessions; schedule-induced licks, water intake, schedule-dependent nose-pokes, pellets earned and stereotyped behaviors of the facultative stage in the SIP sessions. It was f…

Contingent tolerance to amphetamine hypophagia: new insights into the role of environmental context in the expression of stereotypy.

A growing literature attests to the fact that the environment in which a drug is given can have a profound effect on the development and exp … forcement operating in the environment in which the drug is administered. For example, tolerance to amphetamine-induced hypophagia is contingent on having access to food while intoxicated [Carlton PL, Wolgin DL. Contingent tolerance to the anorexigenic effects of amphetamine. Physiol Behav 1971;7:221-223]. Such context-dependency can be explained in terms of …

Study of the behavioral effects of bilateral nucleus accumbens lesions on amphetamine and apomorphine in adult cats.

The aim of the present work was to study the effects of three different types of bilateral lesions performed on the nucleus accumbens, upon the behaviors elicited in adult cats by parenteral administration of amphetamine and apomorphine, and to obtain an understanding of the functional role played by the cited structure. To this end, 10 cats received bilateral injections of 6-OHDA, 18 microg in each accumbens; 8 cats received a similar treatment with ibotenic acid (20 microg), and 11 cats were submitted to bilateral electrolytic damage. Before and after performing these lesions, in separate se…

Sibutramine: a new weight loss agent without evidence of the abuse potential associated with amphetamines.

Sibutramine is a serotonin and norepinephrine reuptake inhibitor that has shown efficacy as a weight loss and weight maintenance agent. Because of the abuse liability and physical dependence potential of amphetamines and related antiobesity agents, this study evaluated the abuse potential of sibutramine and compared it with that of dextroamphetamine and placebo in recreational stimulant users. Thirty-one male recreational stimulant users participated in this single-site, Latin square crossover study that compared the effects of two doses of sibutramine (20 mg and 30 mg) to dextroamphetamine (2…

Development and reversal of sensitization to amphetamine-induced hypophagia: role of temporal, pharmacological, and behavioral variables.

This study shows that sensitization can develop to amphetamine-induced hypophagia and examines the stability of this effect following subsequent pharmacological and behavioral experience. Rats given 36 injections of either amphetamine (2.5 mg/kg; Group A) or saline (Group S) at 3-day intervals developed sensitization of hypophagia, as assessed by a shift to the left in the dose-response (DR) function. Group A also displayed sensitization of stereotypy, whereas Group S showed little change except at the highest dose. Subgroups from each group were then given daily injections of amphetamine (2 m…

Effects of perifornical hypothalamic microinjections of phenylpropanolamine and amphetamine on latency to feed and mash intake in rats.

To determine whether phenylpropanolamine (PPA) and amphetamine act on a common satiety mechanism, the present experiment compared the effect of unilateral microinjections (40, 80, and 160 nmol) of phenylpropanolamine hydrochloride (PPA: d,1-norephedrine) and of d-amphetamine sulfate within the perifornical hypothalamus (PFH) on consumption of a palatable sweetened-mash diet in adult male rats. Microinjection of d-amphetamine (40-160 nmol) within the PFH induced dose-dependent anorexia, whereas PPA microinjections were without effect on feeding. These results document that amphetamine and PPA d…

Effects of drugs on stimulus control of behavior. III. Analysis of effects of pentobarbital and d-amphetamine.

Key-peck responses of pigeons on one of two keys were reinforced intermittently under a multiple fixed-interval schedule. In one component o … ediate (3.0-10.0 mg/kg) doses that did not appreciably alter average rates of responding. Whereas d-amphetamine decreased stimulus control only at high doses (3.0-5.6 mg/kg) that also substantially decreased response rates. The results of subsequent studies suggested that the two drugs primarily affected stimulus control exerted by the colors of the response …

Effect of mazindol, d-amphetamine and diethylpropion on purine nucleotide binding to brown adipose tissue.

Amphetamine, diethylpropion and mazindol were administered to rats in both acute and chronic experiments to measure the changes in purine nucleotide (GDP) binding to the mitochondria from interscapular brown adipose tissue. There was a dose-dependent response to acute treatment with mazindol, but no such effect with diethylpropion. The effects of mazindol and amphetamine were present as early as 3 hours after treatment, and persisted for at least 48 hours, when compared to vehicle-injected rats when all rats were fasted from the time of injection until study. There was no effect when these dru…

Effects of low-level microwave irradiation on amphetamine hyperthermia are blockable by naloxone and classically conditionable.

In a series of experiments, we investigated the effects of pulsed low-level microwave irradiation on amphetamine-induced hyperthermia in the rat. Rats were irradiated in a 2,450-MHz cylindrical waveguide exposure system at 1 mW/cm2, 2 mus pulses, 500 pps, average SAR of 0.6 W/kg. Acute (45 min) exposure to microwaves attenuated amphetamine-induced hyperthermia. This effect was blocked by pretreatment of the animals with the narcotic antagonist naloxone. In another experiment, rats were subjected to ten daily sessions of microwave exposure (45 min/session). On day 11, amphetamine-induced hypert…

Failure of naloxone to modify the effects of chlorpromazine and d-amphetamine on avoidance behavior in the squirrel monkey.

Lever-pressing by squirrel monkeys was maintained under a continuous avoidance schedule in which each response postponed for 30 s the delivery of an electric shock to the tail. Dose-response curves were determined for chlorpromazine (0.03--0.3 mg/kg) and d-amphetamine (0.03--1.0 mg/kg) administered alone and administered concomitantly with 1.0 or 10 mg/kg of naloxone. The dose-response curves for chlorpromazine and d-amphetamine were similar to those previously reported for monkeys under other schedules of shock-maintained behavior: Chlorpromazine decreased responding in a dose-related manner …

Effect of tolerance to morphine, ethanol and barbiturate on amphetamine circling in rats with a striatal dopamine lesion.

The (+)-amphetamine circling rate of rats with unilateral 6-OHDA lesions in the striatum was recorded. Morphine tablets were implanted subcutaneously for chronic treatment. In the morphine-dependent animal the circling rate to amphetamine given 4 days after morphine was first implanted was depressed but after withdrawal with naloxone a day later the rate increased, returning to normal after 21 days. Barbiturate physical dependence was induced by adding increasing amounts of barbitone to the drinking water of lesioned rats over four weeks after which the amphetamine circling response was depres…

Species differences in the in vitro metabolic reduction of the amphetamine metabolite, 1-phenyl-2-propanone.

Marked differences were observed, in the ability of fortified 9000 g liver homogenate supernatants from three species to reduce 1-phenyl-2-propanone to the corresponding alcohol. This metabolic keto-reduction was negligible in homogenates from the rat and extensive in the rabbit; guinea-pig liver homogenates had intermediate ability. Metabolic oxidation of 1-phenyl-2-propanol was negligible in all three species. The amount of deamination of amphetamine and of N-n-propylamphetamine was approximately equal, in vitro, in rats and guinea-pigs but two to three times greater in liver homogenates fro…

Comparison of the anorectic and motor activity effects of some aminoindanes, 2-aminotetralin and amphetamine in the rat.

1. The anorectic and motor activity effects of 1-aminoindane, 2-aminoindane, some N-substituted 2-aminoindanes, 2-aminotetralin, amphetamine and fenfluramine were determined in rats. 2. The two compounds with structures most like the extended conformation of amphetamine, 2-aminotetralin and 2-aminoindane, were potent anorectics. At dosages which halved the intake of food over 1 h, amphetamine increased motor activity, 2-aminotetralin had no effect, and 2-aminoindane reduced motor activity. 3. Both the anorectic and central stimulant actions of 2-aminoindane were absent in N-ethyl- and N-isopro…

Urinary excretion of 3-methoxy-4-hydroxyphenylglycol in depressed patients: modifications by amphetamine and lithium.

The excretion of 3-methoxy-4-hydroxyphenylglycol (MHPG), the urinary metabolite which may best reflect brain norepinephrine, was measured in eight depressed patients on a longitudinal basis during the administration of placebo, d- and 1-amphetamine (30 mg/day), lithium carbonate (0.9 - 2.1 g/day), and d- and 1-amphetamine together with lithium. d-Amphetamine significantly decreased MHPG excretion. No significant change in MHPG was observed during administration of 1-amphetamine or when d- or 1-amphetamine was given together with lithium carbonate. Six patients responded with behavioral activat…

α-Synuclein regulates monoamine transporter capacity and amphetamine-induced efflux via modulation of transporter expression.

α-Synuclein (α-syn) is a presynaptic protein implicated in Parkinson disease and related synucleinopathies, but its physiological function r … diated knockout of α-syn reduced transporter expression and decreased uptake capacity. In addition, amphetamine-induced transporter-mediated efflux was significantly attenuated in α-syn-deficient cells. These findings identify α-syn as a positive regulator of monoamine transporter expression and transport capacity. By controlling transporter abundance, α-syn …
