Role of anatomic architecture in sustained atrial reentry and double potentials.

To determine the role of anatomic architecture in atrial flutter, electrophysiologic findings were correlated with anatomic features in a modified model of atrial flutter with ligation of the crista terminalis. Crista ligation in the middle right atrium prolonged intraatrial conduction time in a rate-dependent manner in 12 dogs, particularly in the low right atrium. With burst atrial pacing, unidirectional block occurred either in the low right atrium or in the interatrial septal region near the superior vena cava, leading to initiation of atrial flutter. Atrial activation mapping revealed a s…

Vascular architecture of the human optic chiasm and bitemporal field defects.

The system of blood supply of the optic chiasma was studied on 85 human fresh bodies by several methods. The optic chiasma is supplied by the Willis' circle, arterioles from which, after forming anastomosis at two levels, enter the chiasma and break into capillaries. The authors found that the median portion of chiasma is a weak point of microcirculation, as demonstrated by rarefied and transverse capillaries. When the blood supply of chiasma is compromised by lesions near the sella turcica, this weak point of microcirculation is apt to be disturbed first and becomes ischemic, causing disorder

Longitudinal changes in left ventricular architecture and function in peripartum cardiomyopathy.

Left ventricular (LV) function was quantitated in 14 patients with peripartum cardiomyopathy, in 10 using 2-dimensional (2-D) echocardiography and in 4 radionuclide ventriculography, and values were compared with those in 11 normal women in the immediate postpartum period. LV end-diastolic and end-systolic volume indexes, LV wall mass index and ejection fraction were calculated during the acute phase of the illness and serially through long-term follow-up (mean 24 months). During the acute phase of illness there was marked LV dilatation: mean end-diastolic volume index was 95 +/- 22 ml/m2, vs

Multiscale Design of Porous Architectures for Highly Efficient Oxygen Electrocatalysis.

Driven by the global imperative for carbon neutrality and sustainable energy development, oxygen electrocatalysis, especially the oxygen evolution reaction (OER) and oxygen reduction reaction (ORR), has emerged as a key link between renewable electricity and clean energy conversion devices. However, the sluggish kinetics of these reactions severely limit energy efficiency, a challenge exacerbated by insufficient active-site utilization and restricted tri-phase mass transport. This review highlights recent advances in multiscale porous architectures for oxygen electrocatalysis, with particular …

Filtration-Dominant Water Treatment Architectures for High-Purity Applications.

Water quality is a critical control variable for healthcare device processing and other high-purity applications. Many facilities rely on chemical treatments (e.g., salt-based softeners, sodium bisulfite for dechlorination), but these methods increase system complexity, generate chemical waste, and introduce risks to staff, patients, and downstream wastewater systems. These challenges are particularly noteworthy in healthcare settings, where both safety and environmental performance are tightly regulated. Filtration-based alternatives that reduce dependence on chemical dosing were evaluated in

Amphiphilic Lipid-Polymer Triblock Architectures as Nanocarriers for Therapeutic Delivery.

This study investigates the formation and characterization of nanocarriers derived from the self-assembly of amphiphilic lipid-polymer hybrids (LPHs) engineered with a symmetric ABA block copolymer architecture. In this design, the terminal A-blocks comprise branched hydrophobic fatty acid (FA) moieties, while the central B-block consists of a hydrophilic polymer segment, either polyethylene glycol (PEG) or a polypeptide-based glycine-lysine (GK) construct. By systematically varying the degree of branching within the hydrophobic blocks and the chemical identity of the hydrophilic segments, thi…

Role of Metabolic Acids in Shaping Bone-like Apatite Architectures.

Bone mineral forms both inside and between collagen fibrils in the extracellular matrix. While the morphology of intrafibrillar bone mineral has been hypothesized to be primarily controlled by the size and shape of the restricted spaces inside collagen fibrils within which the mineral forms, what controls the architecture of the extrafibrillar mineral is still an open question. While bone mineral is primarily apatitic in composition, it also contains significant quantities of cell respiration metabolites, in particular, carbonate, citrate, and lactate. An as-yet unanswered question is what, if…

Complex subtelomeric architectures in a complete rhesus macaque reference genome.

We present T2T-MMU8v2.0, a near-perfect telomere-to-telomere assembly of the rhesus macaque (Macaca mulatta), representing high base-level accuracy reported in a primate genome. Our optimized assembly strategy exposes subtelomeric satellite-rich regions as the principal bottleneck in obtaining full assemblies, owing to long-read technological biases and limitations in hybrid assembly frameworks. By resolving ∼8 Mbp of SATR satellite arrays and discovering 268 previously unannotated repeat families, we define four distinct SATR genomic architectures, each with unique satellite composition, segm…

Genetic Architecture of Myopia and Its Implications for Risk Stratification and Prognosis.

Myopia is a prevalent ocular condition with marked heterogeneity in onset and progression. Although diagnosis is straightforward, predicting disease trajectories and identifying risks of high or pathologic myopia remain main clinical challenges. Advances in human genetics have substantially reshaped current understanding of myopia, revealing a complex architecture involving common polygenic susceptibility, rare high-impact variants, and cumulative genetic risk burden. Large-scale genome-wide association studies demonstrate that myopia-related variants are enriched in regulatory and signaling p…

Key Role of Pro230 in the Hinge Region on the Architecture and Function of IgG1.

Immunoglobulin G (IgG) is a multifunctional glycoprotein essential for immune defense and widely used as a therapeutic due to its antigen specificity and effector functions. However, the inherent flexibility of its hinge region complicates structural characterization and obscures the molecular basis of its mechanism of action. To clarify the hinge's role, we performed systematic amino acid substitutions. Notably, deletion of Pro230 led to the formation of a half-IgG1 species lacking inter-heavy chain interactions. Structural analysis using nuclear magnetic resonance (NMR), negative-stain EM, a

Comparative analysis of manufacturability and binding of various bispecific antibody architectures.

Bispecific monoclonal antibodies (BsAbs) enable novel therapeutic mechanisms with multiple binding specificities, but their architecture can influence manufacturability and function. We compared the production of five formats of a BsAb recognizing a viral epitope with an added function through binding to the transferrin receptor (TfR) for blood-brain barrier passage. They had symmetric and asymmetric architectures, with additional binding domains fused to either the heavy or light chains, and a dual-variable domain immunoglobulin (DVD). We characterized growth kinetics, productivity, downstrea…

Ancient Somatosensory Circuit Architectures Employ Flexible Molecular Strategies.

The extent to which conserved neural circuit architectures depend on shared molecular specification programs remains unclear. Here, we address this question by examining the somatosensory system of the little skate, Leucoraja erinacea , an early-diverging vertebrate that retains ancestral features of both finned and limb-based body plans. We show that core features of somatosensory circuit organization, including laminar organization of the spinal cord and dorsally restricted targeting of sensory afferents, are deeply conserved. Unexpectedly, the molecular programs specifying dorsal root gangl…

An isolation ward's architectural blueprint for prompt infectious disease prevention.

BACKGROUND: The community epidemic prevention in the post COVID-19 requires the prevention to respond quickly to changes in the epidemic and immediately initiate on-site isolation. A system for rapid prevention and control of infectious diseases is built systematically, forming a comprehensive epidemic prevention method, and to achieve the application of the system by proposing a new architecture scheme (isolation wards). METHODS: The paper starts from the theoretical framework of epidemic prevention across preventive medicine, architecture, and mechanical engineering. Through interviews with…

Architectural and biochemical advances in understanding microbial CO2 concentrating mechanisms.

CO2 concentrating mechanisms (CCMs) are ubiquitous in phototrophs. They are found in the micro as well as the macro world. Different domains of life have developed strikingly different approaches to concentrate inorganic carbon around Rubisco, the most prevalent enzyme on earth. While prokaryotes have developed carboxysomes, icosahedral shell protein cages, microbial eukaryotic phototrophs such as green algae, on the other hand, have developed pyrenoids, liquid-liquid phase separated organelles. In contrast to carboxysomes and pyrenoids, which represent biophysical CCMs, some vascular plants (

Research on the protection and restoration technology of glazed components in ancient architecture.

In traditional Chinese architecture, glazed tile components are a significant decorative element that are frequently utilized on walls and roofs. However, glazed tile components typically show variable degrees of deterioration and damage owing to long-term exposure to the natural environment. This study examines and evaluates the preservation quality and primary forms of deterioration of historic architectural glazed tile components. The damage development process of glazed tile components under the combined influences of temperature, humidity fluctuations, and acidic environments was discover…

Molecular architecture and spatial organization of proteasomes in the human sperm nucleus.

Proteasomes are fundamental for protein homeostasis and genome integrity and essential in spermatogenesis and fertilization. However, their presence, composition and role within the sperm nucleus are a subject of debate. Here we use in situ cryo-electron tomography in human sperm cells to elucidate the molecular architecture of nuclear proteasomes, which cluster in DNA-free, nuclear cavities within the sperm nucleus. We show that the main population of proteasomes consists of 20S core particles, with a smaller fraction of 20S capped by PA200 activator. Using single-particle cryo-electron micro…

The Genetic and Epigenetic Architecture of Keratoconus: Emerging Pathways and Clinical Implications.

BACKGROUND: Keratoconus (KC) is a progressive corneal ectasia and a leading cause of corneal transplantation in young adults. Once regarded as a biomechanical disorder, KC is now recognized as a complex disease driven by genetic predisposition, epigenetic modulation, and environmental triggers. Advances in genomics and transcriptomics have begun to elucidate the molecular mechanisms underlying corneal thinning and ectasia. OBJECTIVES: This review synthesizes two decades of evidence on the genetic and epigenetic architecture of keratoconus, highlights key molecular pathways implicated by these…
