Medication treatment of different types of alcoholism.

Alcoholism remains a serious cause of morbidity and mortality despite progress through neurobiological research in identifying new pharmacol … d associated with mood symptoms, may help in the selection of optimal therapy. Emerging adults with binge drinking patterns also might be aided by selective treatments. Although preliminary work on the pharmacogenetics of alcoholism and its treatment has been promising, the assignment to treatment still depends on clinical assessment. Brief behavioral interventi …

Identifying drinking patterns in a middle-aged population and their association with coronary heart disease, stroke, and all-cause mortality risk: the Danish Health Examination Survey.

Alcohol drinking patterns encompass various dimensions of consumption. However, most studies have focused narrowly on the quantity of alcohol consumed. This study aimed to identify alcohol-drinking patterns in middle-aged population using alcohol-related variables and to examine their relationship with coronary heart disease (CHD), stroke, and all-cause-mortality risk. The present population-based study included adults from the Danish Health Examination Survey (DANHES) cohort who were ≥40 years old and free of CHD and stroke at baseline (n = 46 141). Latent Class Analysis (LCA) was used to ide

A Biphasic Effect of Alcohol on Endothelial Plasticity Through Regulation of Endothelial-to-Mesenchymal Transition.

BACKGROUND: Alcohol consumption influences cardiovascular disease, but whether it does so by affecting endothelial plasticity is unknown. We tested whether alcohol regulates endothelial-to-mesenchymal transition (EndMT) to influence arterial pathology. METHODS: HCAEC and HUVEC were exposed to inflammatory cytokines (TGFβ ± IL1β) or hypoxia in the presence of ethanol (0-100 mM). EndMT was assessed by changes in cell marker expression, SNAIL levels, and migration assays. In vivo, carotid ligation was performed in mice gavaged with/without either daily moderate ethanol (2-drink equivalent/d) or

Eating disorder animal model.

PURPOSE OF REVIEW: The aim of this review is to describe animal models that simulate the features of eating disorders. The literature pertaining to animal models that are of relevance for clinical eating disorders and the possible underpinning mechanisms was reviewed using PubMed, Ovid database and Google Scholar. RECENT FINDINGS: New refinements of the circuits regulated by neurotransmitters and neuropeptides which instigate eating behaviours and the various feedback pathways which monitor acute and chronic nutrient status continue to be discovered. Moreover, work with the animal models that

Hepatic injury and inflammation alter ethanol metabolism and drinking behavior.

While liver injury is commonly associated with excessive alcohol consumption, how liver injury affects alcohol metabolism and drinking preference remains unclear. To answer these questions, we measured the expression and activity of alcohol dehydrogenase 1 (ADH1) and acetaldehyde dehydrogenase 2 (ALDH2) enzymes, ethanol and acetaldehyde levels in vivo, and binge-like and preferential drinking behaviors with drinking in the dark and two-bottle choice in animal models with liver injury. Acute and chronic carbon tetrachloride (CCl4), and acute LPS-induced liver injury repressed hepatic ALDH2 acti

Intermittent Exposure to a Single Bottle of Ethanol Modulates Stress Sensitivity: Impact of Age at Exposure Initiation.

Alcohol use during adolescence is a serious public health problem, with binge drinking and high-intensity drinking being particularly harmful to the developing adolescent brain. To investigate the adverse consequences of binge drinking and high-intensity adolescent drinking, adolescent rodents were intermittently exposed to ethanol through intragastric gavage, intraperitoneal injection, or vapor inhalation. These models revealed the long-lasting behavioral and neural consequences of adolescent intermittent ethanol (AIE) exposure. The present study was designed to characterize a different AIE m…

Examining the role of psychological distress in linking childhood maltreatment and alcohol use in young adulthood.

BACKGROUND AND OBJECTIVES: Childhood maltreatment is related to alcohol use as well as psychological distress in young adulthood. Few studies have examined whether psychological distress mediates the relationship between child maltreatment and alcohol use. We examined the role of psychological distress in linking child maltreatment subtypes (ie, emotional abuse, physical abuse, sexual abuse, neglect) to four patterns of alcohol use, including frequency of alcohol use, binge drinking, alcohol-related problems, and alcohol dependence. METHODS: We used a community sample of young adults (N = 337…

Activation of a locus coeruleus to ventral tegmental area noradrenergic pathway blunts binge-like ethanol intake and promotes aversive unconditioned responses in TH-ires-Cre mice.

Ethanol comprises innately aversive properties that may act as a deterrent to overconsumption. We have previously found that chemogenetic activation or inhibition of a noradrenergic locus coeruleus (LC) to rostromedial tegmental nucleus (RMTg) pathway bidirectionally modulates binge-like ethanol intake and aversive reactivity in male and female TH-ires-Cre mice. We previously hypothesized that noradrenergic RMTg circuitry may modulate ethanol intake and aversion through inhibitory inputs to the ventral tegmental area (VTA), a region that is both densely innervated by the RMTg and proposed to c

Alcohol consumption among low-income pregnant Latinas.

BACKGROUND: Due to changing cultural norms, Latinas of childbearing age residing in the U.S. may be at increasing risk of drinking harmful levels of alcohol during pregnancy, and may also be unaware of the risks for Fetal Alcohol Spectrum Disorders associated with this behavior. We assessed the prevalence of alcohol consumption in a sample of low-income pregnant Latinas and examined risk factors for alcohol use in the periconceptional period. METHODS: As part of a larger intervention trial, a cross-sectional in-home interview study was conducted among a sample of 100 pregnant low-income Latin

High-Intensity Drinking Is Linked to Greater Same-Day Positive and Negative Consequences in Young Adults.

OBJECTIVE: Young adults who report high-intensity drinking (HID; 8+/10+ drinks for females/males) are more likely to experience negative alcohol consequences than those reporting heavy episodic drinking (HED; 4+/5+ drinks for females/males). Identifying for whom and when HID is associated with same-day negative (and positive) consequences may inform intervention approaches. We hypothesized that negative consequences would be more likely on HID versus HED days, and when HID levels were reached more quickly. METHOD: Young adults reporting prior HID (N = 203, 57% female) completed a 28-day ecolo

Aggression and repeated alcohol drinking-in-the-dark: dissecting subtypes in outbred mice.

RATIONALE: Repeated bouts of alcohol consumption increase the risk for developing alcohol use disorders and influence social dynamics, including aggression. Intermittent, weekly binge-like drinking-in-the-dark (DID) can escalate such intake in mice, potentially altering the trajectory of adaptive social behaviors. OBJECTIVE: To examine individual patterns of alcohol intake over repeated weeks of DID with corresponding measures of offensive aggression in outbred Swiss Webster (CFW) resident mice. METHODS: CFW resident male mice were given 2 h of access to either water or 16% ethanol (w/v) for t

Gender Differences in Heavy-Episodic Drinking Among Justice-Involved Youth: Testing for Cognitive vs. Social Mediation.

BACKGROUND: There exist gender disparities in heavy-episodic drinking, with male adolescents reporting elevated risk for the behavior. Variation in cognitive development and exposure to antisocial peers may explain these disparities. Deviant peer association may provide a social mechanism and the imbalance in development of dual systems constructs of impulse control and sensation-seeking may provide a cognitive mechanism. Deviant peer association and dual systems imbalance were examined as mediators of the relationship between gender and heavy-episodic drinking among a sample of justice-involv

Alcohol use disorders and the heart.

Alcohol use is an important preventable and modifiable cause of non-communicable disease, and has complex effects on the cardiovascular system that vary with dose. Observational and prospective studies have consistently shown a lower risk of cardiovascular and all-cause mortality in people with low levels of alcohol consumption when compared to abstainers (the 'J'-shaped curve). Maximum potential benefit occurs at 0.5 to one standard drinks (7-14 g pure ethanol) per day for women (18% lower all-cause mortality, 95% confidence interval (CI) = 13-22%) and one to two standard drinks (14-28 g etha

A New Perspective on "Drinking" Self: A Network Approach to Characterizing Drinking-Related Changes in Personality.

BACKGROUND: Previous research suggests that self-knowledge, particularly perceptions of oneself as a "drinker," may influence the development and progression of alcohol use and related problems, such as binge drinking. While existing studies have provided empirical evidence for the importance of assessing drinking self-perception within the five-factor model of personality framework, further examination with novel analytic methods, such as network analyses, could improve understanding of the drinker self-concept. Methods: Five factor traits of openness to experience, conscientiousness, extrave…
