Plague is an infectious disease of humans and animals caused by the bacterium Yersinia pestis. During the Middle Ages millions of people in Europe died from plague, whose current mortality-if untreated-ranges from 50% to 90%. The plague has been a great protagonist in history because it has often been grimly present in the collective events of humans. Its plurisecular history, tied to the whole chain of ecological balance, has had a strong influence on the collective imagination on account of its sudden occurrence and unavoidable mortality. In the past, the passage from contagion to illness en

An overview is given of studies on diseases of mites. Knowledge of diseases of mites is still fragmentary but in recent years more attention … of mite populations in citrus and peach orchards, respectively, but application of these viruses as biological control agents does not seem feasible. A putative iridovirus has been observed in association with Varroa mites in moribund honeybee colonies. The virus is probably also pathogenic for honeybees and may be transmitted to them through this parasitic mite. Few bacte …

High-risk non-muscle-invasive bladder cancer (HR-NMIBC) represents a biologically aggressive disease state with substantial risk of recurrence, progression, and need for radical cystectomy. While intravesical Bacillus Calmette-Guérin (BCG) remains first-line for HR-NMIBC is standard of care, both recurrence and progression have led to the development of novel intravesical agents therapies. This article summarizes key clinical trials that are reshaping the management of HR-NMIBC, with particular emphasis on BCG-unresponsive disease. Approved intravesical and systemic agents demonstrate meaningf

BACKGROUND: Given the rising prevalence of diabetes mellitus (DM) and the limited data on its effect on the course of inflammatory bowel disease (IBD), we characterized multiyear patterns of disease severity in a cohort of IBD patients with coexistent DM.
METHODS: Data of consented IBD patients followed prospectively in a natural history registry at a tertiary center between 2009 and 2017 were analyzed. Patients with ≥3 years of clinical follow-up were included. Patients identified with a diagnosis of DM were compared with 400 consecutive IBD controls without a diagnosis of DM, no laboratory

Severe asthma is a difficult-to-control airway disease with pronounced heterogeneity in both clinical manifestations and underlying inflammatory mechanisms. This review examines the mechanisms, biomarkers, and biologic therapies of severe asthma, with a focus on biomarker-guided treatment selection and emerging precision strategies for type 2-high (T2-high) and type 2-low (T2-low) disease. The development of biologic therapies has changed the treatment paradigm, particularly for patients with T2-high inflammation. By targeting immunoglobulin E (IgE), interleukin-5 (IL-5), interleukin-4 recepto

The Early Alerting and Reporting (EAR) project, launched in 2008, is aimed at improving global early alerting and risk assessment and evaluating the feasibility and opportunity of integrating the analysis of biological, chemical, radionuclear (CBRN), and pandemic influenza threats. At a time when no international collaborations existed in the field of event-based surveillance, EAR's innovative approach involved both epidemic intelligence experts and internet-based biosurveillance system providers in the framework of an international collaboration called the Global Health Security Initiative, w

PURPOSE: Immunomodulatory agents, including conventional immunosuppressive treatment and biologics, are the mainstay of treating chronic uveitis. Janus kinase (JAK) inhibitors, one of the newest biologics, have shown successful outcomes in treating autoimmune diseases such as rheumatoid arthritis and inflammatory bowel diseases by suppressing the JAK/signal transducers and transcription (STAT) pathway. We present two cases of recalcitrant chronic uveitis with significant improvement in intraocular inflammation by using upadacitinib, a selective JAK1 inhibitor.

Non-infectious uveitis (NIU) represents one of the leading causes of blindness in developed countries. The therapeutic strategy aims to rapidly control intra-ocular inflammation, prevent irremediable ocular damage, allow corticosteroid sparing and save the vision, and has evolved over the last few years. Anterior NIU is mostly managed with topical treatment in adults. However, for intermediate, posterior and pan-uveitis, notably when both eyes are involved, systemic treatment is usually warranted. Biotherapies are recommended in case of inefficacy or non-tolerance of conventional immunosuppres

Peptide nucleic acids (PNAs) are a promising platform targeting disease-associated microRNAs due to their high binding affinity, excellent sequence specificity and resistance to enzymatic degradation. However, their therapeutic application is often limited by inefficient cellular uptake. In this study, we present the design, synthesis and physicochemical characterisation of cell-penetrating peptide (CPP)-PNA conjugates that target the oncogenic microRNA miRNA-221-5p, as well as their preliminary biological evaluation. Anti-miRNA-221-5p PNAs functionalised for conjugation with CPPs via Cu(I)-ca

Ion transport across biological membranes, facilitated by naturally occurring ion channels and pumps, is crucial for many biological functions. Many of these transport systems are gated, such that ion transport is regulated by a range of external stimuli, including light, small molecule ligand binding, and membrane potential. Synthetic ion transport systems, including those with similar gating mechanisms, have garnered significant attention due to their potential applications in targeted therapeutics as anticancer agents or to treat channelopathies. In this work, we report stimuli-responsive a

Antibiosis is a key feature widely exploited to develop biofungicides based on the ability of biological control agents (BCAs) to produce fungitoxic compounds. A less recognised attribute of plant-associated beneficial microorganisms is their ability to stimulate the plant immune system, which may provide long-term, systemic self-protection against different types of pathogens. By using conventional antifungal in vitro screening coupled with in planta assays, we found antifungal and non-antifungal Bacillus strains that protected the ornamental plant Kalanchoe against the soil-borne pathogen Fu…

OBJECTIVE: Still's disease (SD) is increasingly being treated with biologic disease-modifying antirheumatic drugs (bDMARDs) according to a treat-to-target approach, with switching between these agents an integral part of this strategy. However, guidance for the clinician on switching between biologic agents in SD is limited. Here we review the evidence in the literature relating to switching between bDMARDs in SD.
METHODS: A systematic review was performed for articles published since 2014 which related to the biologic treatment of SD. Case reports and studies with under 5 patients and non-En

INTRODUCTION: Psoriasis (PsO) is a common inflammatory dermatological condition with a substantial negative impact on patient quality of life. Several biological agents are available for the treatment of PsO, and clinicians and patients must consider various factors when deciding on the most appropriate biological agent.
METHODS: Here, we report a set of consensus statements developed by an Italian PsO advisory board on use of the anti-interleukin-17A biological secukinumab in routine clinical practice.
RESULTS: The statements were developed under three main topics, the first of which was "c

Fusarium oxysporum is a significant threat to agriculture and One Health, requiring advanced molecular tools for functional genomic analyses and biological control agent development. Existing gene-editing methods are hampered by costly protoplast preparation protocols and by CRISPR-Cas9 limitations, such as restricted protospacer adjacent motif (PAM) sequences and complex guide RNA requirements. We engineered an efficient CRISPR/Cpf1 system that overcomes these issues through three main innovations: small-scale protoplast generation using filter column-based methods that greatly reduce enzyme

BACKGROUND: In response to the evolving measles epidemic in the United States, the Centers for Disease Control and Prevention recommended that some adults be revaccinated against measles because they may have inadequate immunity against the virus. Patients receiving biologic medications for psoriasis face a clinical dilemma because they may be at an increased risk of developing severe measles; however, vaccination with the measles-mumps-rubella (MMR) vaccine is not recommended for those on biologic therapy according to the American Academy of Dermatology-National Psoriasis Foundation guideline
