Characterization of novel macrophage cell lines derived from goat and sheep blood and their susceptibility to ruminant virus infections.

Macrophages are present in all vertebrate tissues and play a key role in both the prevention and pathogenesis of infectious diseases. In vitro cultures of macrophages provide a valuable tool for studying the innate immune functions of macrophages and macrophage-pathogen interactions in specific animal species. We previously demonstrated that porcine macrophages were effectively immortalized by transferring the simian virus 40 large T antigen (SV40LT) gene and porcine telomerase reverse transcriptase (pTERT) gene using lentiviral vectors. In the present study, we investigated whether this immor

Time to Positivity of Neonatal Blood Cultures for Early-onset Sepsis.

BACKGROUND: In newborns at risk for early-onset sepsis, empiric antibiotics are often initiated while awaiting the results of blood cultures. The duration of empiric therapy can be guided by the time to positivity (TTP) of blood cultures. The objective of the study was to determine the TTP of neonatal blood cultures for early-onset sepsis and the factors which may impact TTP. METHODS: Observational study of blood cultures growing pathogenic species obtained within 72 hours of birth from infants born at 23-42 weeks gestation, at 19 hospitals in Northern California, Boston, and Philadelphia. Molecular characterization of Ehrlichia canis and Babesia vogeli reveals multiple genogroups associated with clinical traits in dogs from urban areas of Colombia.

Ehrlichia canis and Babesia vogeli are vector-borne pathogens that infect blood cells and produce the diseases Canine Monocytic Ehrlichiosis (CME) and Babesiosis in dogs. Considering the lack of studies on these pathogens in Colombia, this study aims to determine the molecular prevalence and genetic characterization of E. canis and Babesia spp., in dogs from the Metropolitan Area of Bucaramanga (MAB), Santander, a region with one of the greatest pet densities in Colombia. One hundred eighty-five dogs were surveyed and analyzed through molecular, clinical, and hematological approaches. The mole

Molecular and microscopic detection of haemoprotozoan diseases in dogs from Haryana, India.

Haemoparasitic infections are frequently observed in dogs from tropical regions, including India. The present investigation combined microscopic blood smear examination and PCR assays to assess the occurrence of canine tick-borne diseases (CTBD) from suspected dogs in and around Hisar, Haryana. Using the Giemsa-stained peripheral thin blood smear examination, 15 (12.5%) of the 120 dogs were infected with CTBD, with 5.8%, 3.3%, 2.5%, and 0.8% dogs testing positive for Hepatozoon canis, Ehrlichia canis, Babesia vogeli, and Babesia gibsoni, respectively. Using the PCR assay, CTBD was found to be

Detrimental role for CCAAT/enhancer binding protein δ in blood-borne brain infection.

BACKGROUND: The most frequent pathogen that causes bacterial meningitis is the Gram-positive bacterium Streptococcus (S.) pneumoniae. CCAAT/enhancer binding protein δ is a transcription factor that has recently been hypothesized to play a detrimental role in outcome of meningitis caused by S. pneumoniae. Here, we studied the role of C/EBPδ prior to the development of pneumococcal meningitis. METHODS: Wild-type and C/EBPδ-deficient mice (C/EBPδ-/-) were intraveneously infected with S. pneumoniae and sacrificed after 24 or 48 h. cebpδ expression, bacterial loads, inflammatory response and patho

[Postdonation information: the French fourth hemovigilance sub-process].

Postdonation information is the knowledge of information about the donor or his donation, occurring after it, which challenges quality or safety of the blood products stemming from this or other donations. Classical hemovigilance sub-processes concerning donors or recipients adverse events do not cover this topic. France is just about to make it official as a fourth sub-process. Less formal management of postdonation information is already set up for more than ten years. French data of the year 2013 are presented, including the regional notification level and the national reporting one. A sign

Prevention of AIDS transmission through screening of the blood supply.

Shortly after the first cases of AIDS were reported in 1981, it became apparent that this disease was caused by a blood-borne infectious agent that could be transmitted by blood transfusion. Early reports documented a reduced ratio of CD4+ to CD8+ T cells not only in AIDS patients but also in likely carriers of the AIDS pathogen. On this basis, from July 1983 to June 1985, our blood center utilized flow cytometry to test each donated unit for the ratio of CD4 to CD8 T cells; we did not transfuse blood from donors with CD4/CD8 < 0.85. Despite available data supporting the utility of this or oth

Infection Transmission Associated with Point of Care Testing and the Laboratory's Role in Risk Reduction.

Lack of knowledge and confusion exists regarding safe and appropriate use of blood glucose monitoring equipment. Increasing numbers of diabetics, and exponential growth in blood glucose monitoring presents increased opportunities for infection transmission between patients. Diabetics have increased exposure to blood and blood borne pathogens from frequent blood glucose monitoring. Risk factors have been identified in infectious outbreaks and by analysis of testing practice. Point of care blood glucose meters are frequently contaminated by blood. Bacterial and viral organisms survive on surface

Insects and the Transmission of Bacterial Agents.

Arthropods are small invertebrate animals, among which some species are hematophagous. It is during their blood meal that they can transmit pathogenic microorganisms that they may be harboring to the vertebrate host that they parasitize, which in turn will potentially develop a vector-borne disease. The transmission may occur directly through their bite, but also through contaminated feces. Zoonotic diseases, diseases that can naturally be transmitted between humans and animals, are a considerable part of emerging diseases worldwide, and a major part of them are vector-borne. Complete Inhibition of Vertical Mosquito Landing through Topographical Surface Design.

Mosquito-borne diseases such as dengue, chikungunya, and malaria have long been a health and economic burden in our society. Such illnesses develop after the pathogen, here arboviruses, are transmitted to humans by female mosquitoes during blood meals. In the case of dengue and chikungunya, such pathogens are transmitted to humans by infected Aedes aegypti females. Prior to feeding the insects rest on vertical surfaces. In this work, a surface roughness threshold was observed for live Aedes aegypti colonies, and below a root-mean-squared roughness of Sq < 0.124 μm the mosquitoes were physicall

AG129 Mice as a Comprehensive Model for the Experimental Assessment of Mosquito Vector Competence for Arboviruses.

Arboviruses (an acronym for "arthropod-borne virus"), such as dengue, yellow fever, Zika, and Chikungunya, are important human pathogens transmitted by mosquitoes. These viruses impose a growing burden on public health. Despite laboratory mice having been used for decades for understanding the basic biological phenomena of these viruses, it was only recently that researchers started to develop immunocompromised animals to study the pathogenesis of arboviruses and their transmission in a way that parallels natural cycles. Here, we show that the AG129 mouse (IFN α/β/γ R-/-) is a suitable and com

Assessment of TLR-2 concentration in tick-borne encephalitis and neuroborreliosis.

The aim of the study was to check whether measurement of TLR-2 in serum or cerebrospinal fluid (CSF) can help differentiate between neuroborreliosis (NB) and tick-borne encephalitis (TBE). Eighty patients with meningitis and meningoencephalitis were divided into two groups: Group I - patients with NB (n = 40) and Group II - patients with TBE (n = 40). Diagnosis was based on the clinical picture, CSF examination and presence of specific antibodies in serum and CSF. The control group (CG) consisted of healthy blood donors (n = 25) and patients in whom inflammatory process in central nervous syst

Travel patterns and risk behavior in solid organ transplant recipients.

BACKGROUND: International travel is associated with an increased risk of enteric, vector-borne and bloodborne infections. The risk of acquiring travel-related illness is higher in those who are immunocompromised. However, little is known about travel practices and risk behaviors in transplant recipients who travel. We herein profile transplant recipients who travel, and characterize their pre-travel precautions, travel activities, burden of illness, and exposure history. METHODS: With the use of a standardized and validated questionnaire, patients attending a transplant clinic were surveyed r

Comparison of DNA Extraction and Amplification Techniques for Use with Engorged Hard-Bodied Ticks.

Tick-borne infections are a serious threat to humans, livestock, and companion animals in many parts of the world, often leading to high morbidity and mortality rates, along with decreased production values and/or costly treatments. The prevalence of the microbes responsible for these infections is typically assessed by the molecular identification of pathogens within the tick vectors. Ticks sampled from animals are often engorged with animal blood, presenting difficulties in the amplification of nucleic acids due to the inhibitory effects of mammalian blood on the enzymes used in polymerase c

Ticks and Tick-Borne Pathogens Associated with Dromedary Camels (Camelus dromedarius) in Northern Kenya.

Ticks and tick-borne pathogens (TBPs) are major constraints to camel health and production, yet epidemiological data on their diversity and impact on dromedary camels remain limited. We surveyed the diversity of ticks and TBPs associated with camels and co-grazing sheep at 12 sites in Marsabit County, northern Kenya. We screened blood and ticks (858 pools) from 296 camels and 77 sheep for bacterial and protozoan TBPs by high-resolution melting analysis and sequencing of PCR products. Hyalomma (75.7%), Amblyomma (17.6%) and Rhipicephalus (6.7%) spp. ticks were morphologically identified and con
