[Diaphyseal dysplasia (Camurati-Engelmann syndrome) with progressive loss of vision: 30-year observations and the effect of prednisolone treatment (author's transl)].

Radiological changes in the skeleton had been present for 30 years in a now 42-year-old man with diaphyseal dysplasia (Camurati-Engelmann syndrome). Because of threatened blindness by bony narrowing of the optic canal prednisolone was administered over several years. Biochemical, calcium-kinetic and ophthalmological studies demonstrated a favourable effect of this treatment.

Difficulties in classification of the short rib-polydactyly syndromes.

A case of "short rib-polydactyly" (SRP) syndrome (type II-Saldino-Noonan) with a detailed study of the bones, muscles and peripheral nerves in the extremities is presented. Analysis of the literature (35 cases with different forms of SRP) and the comparison of clinical, genetic and morphological data lead us to consider that different types of SRP may be related to different alleles of the same gene. Genetic heterogeneity (mutations of different genes) is also possible.

Acrodysostosis. A case of peripheral dysostosis, nasal hypoplasia, mental retardation and impaired hearing.

Acrodysostosis--a rare congenital malformation syndrome--is described in a 4 1/2 year old boy with peripheral dysostosis, nasal hypoplasia, mental retardation (PNM syndrome) and impaired hearing. The differential diagnosis includes pseudo (PH)--and pseudo-pseudohypoparathyroidism (PPH). The patient described here had severe peripheral dysostosis, typical of PNM as opposed to the above-mentioned conditions with only moderate peripheral dysostosis. Furthermore, there were no soft tissue calcifications and no intracranial calcification as can be seen in PH and PPH. Laboratory findings were normal

Autosomal recessive inheritance of metaphyseal dysplasia (Pyle disease).

Two young adults with Pyle disease have been investigated in a large Afrikaner kindred in South Africa. Consanguinity was present in the family, and it is likely that the condition was inherited as an autosomal recessive. This contention is supported by the radiographic demonstration of minor degrees of widening of the distal femora in obligatory and potentially heterozygous relatives. Apart from genu valgus of moderate degree, the patients enjoyed good health and their gross radiographic skeletal abnormalities contrasted with the innocuous clinical presentation. Differentiation of Pyle diseas

[Which films should be taken? Suspicion for congenital skeletal dysplasia (author's transl)].

Clinical signs and roentgenologic alterations are important for classification of congenital skeletal dysplasis. It is summarized which skeletal parts are most important for the roentgenographic determination of bone dysplasia. A minimal survey includes films of the vertebral column (lateral projection of the pelvis, hand, knee and skull (lateral projection). A schematic presentation demonstrates the vertebral column lesions in the most important congenital skeletal dysplasias.

Epiphyseo-metaphyseal cupping following infantile scurvy.

A five year old girl presented with short femora, cupped distal metaphyses and flexion deformity of the knees suggesting previous epiphyseo-metaphyseal damage. The aetiology was suggested by finding a history of previous infantile scurvy with separated distal femoral epiphyses. The rarity of this complication and possible contributing factors are discussed.

Exomphalos, inguinal hernia, renal agenesis, skeletal dysplasia, heart disease--a "new" syndrome?

We report a growth retarded, male child with exomphalos, bilateral direct inguinal hernias, unilateral renal agenesis, congenital heart defect, costovertebral dysplasia, microcephaly, ventricular septal defect, and paroxysmal supraventricular tachycardia associated with Wolff-Parkinson-White syndrome. The natural history of that syndrome is poorly understood and provisional suggestion of a new syndrome will depend on further similar observations.

Communicating cartilage canals of the physis of the distal part of the ulna of growing swine and their potential role in healing of metaphyseal dysplasia of osteochondrosis.

The distal parts of ulnae of 22 pigs ranging in age from 1 day to 6 months were studied histologically. Communicating cartilage canals passing from the epiphyseal vasculature through the physis and anastomosing with the metaphyseal vasculature were identified in neonatal pigs. In older pigs, the communicating cartilage canals had become smaller, as evidence by the presence of only capillaries. Eosinophilic matrical strekas and patches containing scattered elongated nuclei and centrally located accumulations of erythrocytes within the physis became more numerous in groups of old pigs. These str

[Changes in the juvenile pelvis in congenital systemic diseases of the skeleton (author's transl)].

Constitutional disorders of the skeleton show a characteristic appearance in the radiogram of the juvenile pelvis. These radiologic changes are presented in tables and are discussed demonstrating individual cases. Because of the limited reactive possibility of the skeleton not every disorder shows a typical feature of differential diagnosis and radiologic pattern. To differentiate from the congenital dysplasia of the hip and Legg-Calvé-Perthes disease the distribution of the skeletal disorders and their course therefore have to be known. Accordingly the therapeutic consequences have to be draw

Unravelling the Intricate Roles of FAM111A and FAM111B: From Protease-Mediated Cellular Processes to Disease Implications.

Proteases are critical enzymes in cellular processes which regulate intricate events like cellular proliferation, differentiation and apoptosis. This review highlights the multifaceted roles of the serine proteases FAM111A and FAM111B, exploring their impact on cellular functions and diseases. FAM111A is implicated in DNA replication and replication fork protection, thereby maintaining genome integrity. Additionally, FAM111A functions as an antiviral factor against DNA and RNA viruses. Apart from being involved in DNA repair, FAM111B, a paralog of FAM111A, participates in cell cycle regulation

ADAMTSL2 mutations determine the phenotypic severity in geleophysic dysplasia.

Geleophysic dysplasia-1 (GD1) is an autosomal recessive disorder caused by ADAMTS-like 2 (ADAMTSL2) variants. It is characterized by distinctive facial features, limited joint mobility, short stature, brachydactyly, and life-threatening cardiorespiratory complications. The clinical spectrum spans from perinatal lethality to milder adult phenotypes. We developed and characterized cellular and mouse models, to replicate the genetic profile of a patient who is compound heterozygous for 2 ADAMTSL2 variants, namely p.R61H and p.A165T. The impairment of ADAMTSL2 secretion was observed in both varian
