[Pulmonary megakaryocytes (MK) in experimental hemorrhagic shock].

Quantification of MK in blood and pulmonary sections was performed in rabbits with induced hemorrhagic shock (group I) and in control rabbits (group II). In electron transmission microscopy we studied the ultrastructure of pulmonary capillaries. Animals in group I were exsanguinated to achieve systemic blood pressure level of 40 mm Hg. This level was maintained for 1.5 hours and then the rabbits from both groups were sacrificed. Pulmonary arterial and venous blood was sampled in the amount of 10 ml and two sections from each pulmonary lobe were taken. The blood samples were then treated with S

HCG treatment increases intratesticular pressure in the abdominal testis of unilaterally cryptorchid rats.

Adult, unilaterally cryptorchid rats were given a single subcutaneous injection of hCG. HCG treatment of 100 I.U. (but not 10 I.U.) resulted in a marked increase in intratesticular pressure (approximately 40 mm Hg) in the abdominal testis that was maximal 24 hours after treatment. This increase in pressure is caused by increased vascular permeability coupled with insufficient lymph drainage. In the scrotal testis, hCG treatment resulted in increased vascular permeability and lymph flow, but this did not result in a marked increase in testicular pressure. No morphologic signs of hCG-induced dam

Extravascular lung water.

Extravascular lung water (idQw1) is measured in vivo from the difference in mean transit times, computed by extrapolating the dilution curves, of two indicators, one freely diffusible, the other confined to the intravascular space. Using 3H2O it has been shown that idQw1 is smaller than the amount of extravascular water obtained from the difference between wet and dry lung weight (Qw1). Extrapolation allows one to use dilution curves for a short time, i.e., up to onset of obvious recirculation. Clearing the dilution curves or recirculation by deconvolution extends the observation time, which t

[The alpha-adrenergic effect of thymoxamine on the vessels of the ciliary body].

The effects of epinephrine on the permeability of the blood-aqueous barrier, stimulated by paracentesis of the anterior chamber, were studied in rabbits following alpha-adrenergic blocking by means of topical or systemic thymoxamine. The alpha-adrenergic block was tested by continuous recording of the systemic arterial blood pressure. The reversal of the vascular effects of epinephrine after administration of thymoxamine was demonstrated at the bloodaqueous barrier. The miotic property of thymoxamine is believed to have had no influence on our results.

Measurement of water filtration in skeletal muscle in man by an osmotic transient method.

Water filtration in the human forearm was determined with a new method using a hyperoncotic transient of albumin solution infused into the brachial artery. Baseline dilution of labelled albumin in deep forearm vein plasma in excess of the contribution from arterial blood and from infusate was assumed to originate from extravascular water filtered into the blood by the transient. The filtration coefficient (Fc) was determined as the ratio between filtered water and increase in colloid osmotic pressure in the blood samples, and gives the filtrative water permeability in the exchange areas of the

Effect of serotonin, histamine and bradykinin on outflow facility following ciliary muscle retrodisplacement in the cynomolgus monkey.

Cynomolgus monkeys with surgically untouched, aniridic, or ciliary muscle disinserted eyes underwent two-level constant pressure perfusion of the anterior chamber to determine total outflow facility before and after intracameral infusion of bradykinin triacetate (0.1, 1, 10 micrograms), histamine dihydrochloride (1, 10, 100 micrograms), or serotonin creatinine sulfate (1, 10, 100 micrograms). Bradykinin had no effect on facility in non-disinserted eyes, but significantly decreased facility in disinserted eyes at all three dosages. Histamine tended to decrease facility in non-disinserted and di

A physiological study of elastic compression stockings in venous disorders of the leg.

The physiological effects of elastic compression stockings on venous disorders of the leg have been studied in three groups of 10 patients. These patients were divided by examination and ascending venography into those with (a) a normal venous system in the leg, (b) superficial varicose veins and (c) a post-phlebitic limb. In a randomised cross-over trial each group wore Eesiness NHS two-way stretch or Sigvaris medium compression below-knee stockings for three weeks. In the post-phlebitic limb group a further comparison was made with Sigvaris strong compression below-knee stockings after a 6-w

Transcapillary refill in hemorrhage and shock.

Moderate or major hemorrhage leads to diminished cardiac output and to increased total peripheral resistance. These cannot be corrected fully until blood volume is restored, which, in the absence of therapy, requires the movement of fluid and of protein from the interstitium into the capillaries. This movement appears mediated entirely by changes in the Starling forces, dominated in the first phase by a fall in capillary hydrostatic pressure, which promotes a rapid shift of protein-free fluid from the interstitium into the capillaries. The second phase, temporally overlapping the initial phase

[State of the microcirculation following total blood replacement with a model perfluorotributylamine-based blood substitute in rats].

Microcirculation was studied in experiments on rats whose blood had been completely replaced by perfluorotributylamin emulsion. After blood replacement the microcirculatory channel was actively perfused with the blood substitute. It was shown that the emulsion circulation rate exceeded the initial rate of the blood flow, that high level of nutritive vessels perfusion was maintained and the physiological parameters of reactivity and functional motility of microvessels remained unchanged. It is suggested that under these circumstances there exist favourable conditions for transcapillary exchange

Increased sheep lung vascular permeability caused by pseudomonas bacteremia.

In awake sheep, we compared the responses of lung lymph flow and lymph and plasma protein concentrations to steady state elevations of pulmonary vascular pressures made by inflating a left atrial balloon with those after an intravenous infusion of 10(5)-10(10)Pseudomonas aeruginosa. Lymph flow increased when pressure was increased, but lymph-plasma protein concentration ratios always fell and lymph protein flow (lymph flow x lymph protein concentration) increased only slightly. After Pseudomonas, sheep had transient chills, fever, leukopenia, hypoxemia, increased pulmonary artery pressure and

HIF-1α Enhances Vascular Endothelial Cell Permeability Through Degradation and Translocation of Vascular Endothelial Cadherin and Claudin-5 in Rats With Burn Injury.

The mechanism underlying burn injury-induced enhanced vascular endothelial permeability and consequent body fluid extravasation is unclear. Here, the rat aortic endothelial cells (RAECs) were treated with the serum derived from rats with burn injury to elucidate the mechanism. Sprague-Dawley (SD) rats were grouped as follows (10 rats/group): control, 2, 4, 8, 12, and 24 hours postburn groups. The heart, liver, kidney, lung, jejunum, and ileum of rats injected with 2% Evans blue (EB) through the tail vein were excised to detect the EB level in each organ. The serum levels of hypoxia-inducible f

Increased Accumulation and Retention of rhFVIIa (eptacog beta) in Knee Joints of Hemophilia A Mice Compared to Wild-Type Mice.

Our earlier studies showed that recombinant human factor VIIa (rhFVIIa) administered intravascularly in mice disappeared rapidly from the circulation. However, a small fraction of rhFVIIa that entered extravascular remained functionally active for an extended period. The present study aims to investigate the dose-dependency of rhFVIIa accumulation and retention in mouse knee joints and test whether the hemophilic condition affects rhFVIIa sequestration in joints. Wild-type and FVIII-/- mice were injected with three doses of rhFVIIa (eptacog beta, 90, 250, and 500 μg/kg) via the tail vein. At v
