Effects of Dietary Quality on Vaginal Microbiome Composition Throughout Pregnancy in a Multi-Ethnic Cohort.

Background: Vaginal Lactobacillus predominance is associated with improved vaginal health and reduced pregnancy complications. Little is known about how dietary quality may improve vaginal microbial composition or about dietary interventions that may promote Lactobacillus abundance. To understand the host factors affecting vaginal microbiota during pregnancy in a multi-ethnic cohort in Hawai`i. We hypothesize that better diet quality improves vaginal microbial composition, as represented by Lactobacillus abundance and depletion of anaerobic organisms. Methods: We compared comprehensive diet qu

Downregulation of VvPLL3 enhances grapevine resistance to Erysiphe necator via defense activation and cell wall remodeling.

Grapevine powdery mildew (PM) is a major cause of yield loss worldwide, highlighting the need for durable resistance strategies. Recessive resistance against PM has been previously reported by silencing a pectate lyase-like (PLL) susceptibility gene in the model plant Arabidopsis thaliana (AtPMR6), but the function of this susceptibility gene is unknown in crop plants. Grapevine has multiple copies of PLL genes, but their precise role in PM resistance is not fully understood. Here we report using an artificial microRNA-based gene silencing approach in PM-susceptible Vitis vinifera cv. Chardonn

Anaerostipes stercorihominis sp. nov., isolated from human faeces.

The genus Anaerostipes is one of the major butyrate-producing genera in the gut microbiota of healthy human adults. During a culturomics study of the human gut, strain 17EGHSPOS18-1T was isolated from the faeces of a healthy adult. Here, we conducted a taxonomic study of the strain. Phylogenetic analysis based on 16S rRNA gene sequences placed strain 17EGHSPOS18-1ᵀ in the Anaerostipes cluster. This placement was supported by a core gene-based phylogenetic analysis. Strain 17EGHSPOS18-1T shared average nucleotide identity (ANI) values of <80% and digital DNA-DNA hybridization (dDDH) values of <

Metagenome-Based Functional Differentiation of Gut Microbiota and Ecological Adaptation Among Geographically Distinct Populations of Przewalski's Gazelle (Procapra przewalskii).

Przewalski's gazelle (Procapra przewalskii) is an endangered ungulate endemic to the Qinghai-Tibet Plateau, with a small population size and exposure to multiple ecological pressures. Its gut microbiota may play a crucial role in host environmental adaptation. To investigate the functional divergence of gut microbial communities, we performed high-throughput metagenomic sequencing on 105 wild fecal samples collected from 10 geographic regions around Qinghai Lake. The results revealed significant regional differentiation in key functional modules related to metabolism, antibiotic resistance mec

Puf4 -mediated oxidative stress virulence attenuation in Cryptococcus neoformans.

INTRODUCTION: Cryptococcus neoformans is a ubiquitous environmental fungal pathogen whose pathogenicity is closely linked to its ability to adapt to host environments. The RNA-binding protein Puf4 plays a key role in regulating the cell wall of C. neoformans. However, the specific mechanisms by which Puf4 regulates metabolism and virulence remain unclear. METHODS: In this study, we systematically investigated the role of Puf4 in the metabolism and virulence of C. neoformans using RNA sequencing (RNA-seq), biochemical analysis, and in vivo animal experiments. RESULTS: Colony-spotting assay as

Bisphenol analogues induced metabolic effects through eliciting intestinal cell heterogeneous response.

The metabolic effects of endocrine-disrupting chemicals, such as bisphenol analogues, have drawn increasing attention. Bisphenol A (BPA) usage is associated with the occurrence of many metabolic diseases. With the restricted use of BPA, alternatives like bisphenol F (BPF) and bisphenol AF (BPAF) have been greatly introduced for industrial manufacture, and brings new hazard to public health. To understand how bisphenol analogues induced metabolic effects, zebrafish are continuous exposed to environmental level (0.5 μg/L) of BPA, BPF and BPAF since embryonic stage, and identified hepatic steatos

Comprehensive analysis of pharmaceutical products using simultaneous mixed-mode (ion-exchange/reversed-phase) and hydrophilic interaction liquid chromatography.

Liquid chromatographic assays were developed using a mixed-mode column coupled in sequence with a hydrophilic interaction liquid chromatography column to allow the simultaneous comprehensive analysis of inorganic/organic anions and cations, active pharmaceutical ingredients, and excipients (carbohydrates). The approach utilized dual sample injection and valve-mediated column switching and was based upon a single high-performance liquid chromatography gradient pump. The separation consisted of three distinct sequential separation mechanisms, namely, (i) ion-exchange, (ii) mixed-mode interaction

Properties and serum levels of pregnancy-associated variant of human transcortin.

The amino acid composition, N- and C-terminal amino acid sequences, and the basic physicochemical and immunochemical properties of the recently discovered pregnancy-associated molecular variant of human transcortin (Strel'chyonok, O.A., Avvakumov, G.V. and Akhrem, A.A. (1984) Carbohydr. Res. 134, 133-140) have been found to be identical to those of transcortin from normal donor serum. This suggests the identity of polypeptide moieties of the two glycoproteins. The transcortin variant has a lower isoelectric point (3.5-4.1) than normal transcortin (3.6-4.2), and different electrophoretic mobili

The molecular genetic basis of muscle phosphoglycerate mutase (PGAM) deficiency.

The glycolytic enzyme phosphoglycerate mutase (PGAM) is a dimer, and mature human skeletal muscle contains almost exclusively the MM form of the enzyme, PGAM-M. In 1981, we identified a patient with PGAM-M deficiency, and three additional patients have since been described. All presented with exercise intolerance, cramps, and myoglobinuria. We report two new patients with PGAM-M deficiency and describe the molecular lesions in five patients--four African-Americans and one Caucasian. Three patients were homozygous for an identical G-to-A transition converting an encoded Trp to an in-frame stop

MUC6 apomucin shows a distinct normal tissue distribution that correlates with Lewis antigen expression in the human stomach.

BACKGROUND & AIMS: Among the human mucin complementary DNAs thus far identified, two (MUC5AC and MUC6) were cloned from stomach libraries. This study examines the distribution of MUC6 in normal tissues and compares it with that of MUC5AC as well as with the expression of Lewis blood group antigens. METHODS: Affinity-purified rabbit antibodies detecting epitopes within the repetitive sequence of MUC5AC and MUC6 were used in enzyme-linked immunosorbent assays and immunohistochemical assays. RNA expression was analyzed by in situ hybridization. Double-labeling immunofluorescence was used to stud

Molecular characterization of the O-acetyl transferase gene of converting bacteriophage SF6 that adds group antigen 6 to Shigella flexneri.

Bacteriophage SF6 antigenically converts Shigella flexneri serotype Y strains (-;3,4) to type 3b carrying group antigen 6,3,4 by means of an O-acetylation of the O-antigenic polysaccharide chain. The gene for O-acetyl transferase of bacteriophage SF6 has been cloned, identified and sequenced. The predicted O-acetyl transferase protein encoded by this gene was found to consist of 333 amino acids, (37,185 daltons) and to have some similarity with the galactose-1-phosphate uridylyltransferase protein of Escherichia coli. The gene has been shown to function in a live vaccine strain of S. flexneri

The Cell-Surface N-Glycome of Human Embryonic Stem Cells and Differentiated Hepatic Cells thereof.

Human embryonic stem cells (hESCs) are pluripotent stem cells that offer a wide range of applications in regenerative medicine. In addition, they have been proposed as an appropriate alternative source of hepatocytes. In this work, hESCs were differentiated into definitive endodermal cells (DECs), followed by maturation into hepatocyte-like cells (HLCs). Their cell-surface N-glycome was profiled and also compared with that of primary human hepatocytes (PHHs). Undifferentiated hESCs contained large amounts of high-mannose N-glycans. In contrast, complex-type N-glycans such as asialylated or mon

Geographic differentiation of the gut microbiota in Lissorhoptrus oryzophilus (Coleoptera: Curculionidae): associations with environmental gradients and predicted functional potential.

The rice water weevil, Lissorhoptrus oryzophilus (Kuschel), is a major invasive pest native to rice that has expanded far beyond its native North American range to establish widespread populations across Asia and Europe. However, how altitudinal gradients shape this microbial ecosystem remains unclear. We performed shotgun metagenomics to profile the gut microbiota of 3 geographically distinct populations-Pingba District (PB), Yibin City (SC), and Mudanjiang (MDJ) City-of L. oryzophilus across elevational gradients in China. Though low-abundance archaeal, fungal, and viral sequences were detec

Paired oral clinical specimens reveal the underlying ecology supporting the emergence of inflammophilic microbiome communities.

BACKGROUND: Inflammatory oral diseases are associated with reproducible shifts from commensal-dominated microbiota toward pathobiont-enriched communities, yet the ecological mechanisms underlying the emergence of inflammophiles remain poorly understood. This study aims to investigate if host-derived inflammatory environments act as selective pressures that restructure microbial metabolism and community organization during disease progression. METHODS: We performed 16S rRNA gene sequencing of patient-matched pediatric dental plaque and odontogenic abscess specimens to capture microbial communi

Fructophilic lactic acid bacteria as a window into multi-scale convergent evolution.

Fructophilic lactic acid bacteria (FLAB) are a group of lactic acid bacteria with unique growth characteristics, that is, poor growth on glucose. Their growth is enhanced in the presence of fructose or external electron acceptors. These organisms inhabit fructose-rich environments such as flowers, fruits, and pollinating insects, particularly honey bees. Apilactobacillus spp. and Fructobacillus spp. are representatives of FLAB, although they belong to phylogenetically distant clades. These organisms commonly possess markedly small genomes with a low number of coding DNA sequences. Furthermore,
