Improved Detection Sensitivity of Spring Viremia of Carp Virus by Substituting a Two-Step with a One-Step Nested Reverse Transcription Polymerase Chain Reaction Method.

Spring viremia of carp (SVC) is a highly contagious disease that affects cyprinids, resulting in systemic hemorrhage, abdominal distension, exophthalmia, and high mortality in juveniles. This can lead to significant losses in the aquaculture industry. The World Organization for Animal Health (WOAH) recommends a two-step semi-nested reverse transcription polymerase chain reaction (RT-PCR) method for diagnosis. However, this method is labor-intensive, requires large reagent volumes, and is prone to carry-over contamination. Here, we evaluated the detection sensitivity of one-step semi-nested RT-

Functional divergence of fabp genes in common carp and the effects of high-fat diets supplemented with linalool on growth, fatty acid composition and fabp gene expressions.

The fatty acid-binding protein (fabp) genes involved in β-oxidation were identified and characterized in the common carp (Cyprinus carpio) genome. Fish were then fed high-fat diets supplemented with various amounts of linalool, a natural peroxisome proliferator-activated receptor α ligand, for 9 weeks. We determined that C. rapio genome contains 19 fabp genes. The mRNA expression of 4R-whole genome duplication copies of fabp genes was differently regulated by the diets, except for fabp3a.1/fabp3a.2 and fabp7a.1/fabp7a.2. These findings indicate that the transcriptional regulatory mechanisms go

Transcriptomic analysis reveals that selenium-enriched Lactobacillus plantarum alleviates high -salinity stress in common carp through lipid metabolism and ferroptosis signalling pathways.

Inland water resource salinisation is a global problem. High salinity leads to increased pressure on the survival of aquatic animals, severely limiting the development of the aquaculture industry. Salinity stress disrupts antioxidant defences in aquatic animals and results in oxidative stress, damaging body functions. As an important constituent of glutathione, selenium has been shown to play an important role in improving antioxidant defences and regulating metabolism. Biological methods for synthesising selenium nanoparticles (Bio-SeNPs) using Lactobacillus plantarum are safe, inexpensive, a

Cell-free hemoglobin released from hemolysis induces programmed cell death through iron overload and oxidative stress in grass carp (Ctenopharyngodon idella).

Intravascular hemolysis releases hemoglobin (Hb) from red blood cells under specific conditions, yet the effect of hemolysis in aquaculture systems remain poorly understood. In this study, a continuous hemolysis model for grass carp was established by injection of phenylhydrazine (PHZ) to investigate the mechanistic impacts of sustained hemolysis. PHZ-induced hemolysis altered liver color, and subsequent hematoxylin and eosin staining revealed substantial Hb accumulation in the head kidney, accompanied by inflammatory cell infiltration and vacuolization in liver tissue. Quantitative real-time

Comprehensive Evaluation of Growth Performance, Hematological Parameters, Antioxidant Capacity, Innate Immunity, and Disease Resistance in Crucian Carp (Carassius auratus) Lacking Intermuscular Bones.

The presence of intermuscular bones severely affects the edibility and value-added processing of crucian carp (Carassius auratus), becoming a constraint to the high-quality development of its industry. Our previous study identified bmp6 as the key osteogenic regulator and successfully developed a new crucian carp strain lacking intermuscular bones (WUCI) using CRISPR/Cas9 technology. To accelerate its industrialization, we comprehensively assessed WUCI's growth performance, hematological parameters, antioxidant capacity, innate immunity, and disease resistance. The results demonstrated that th

Bioaccumulation of lead, arsenic, and mercury in vital organs of common carp (Cyprinus carpio L.): Assessment of pathological effects and possible hazards associated with human consumption.

BACKGROUND: Pollution of aquatic environments with heavy metals causes severe adverse effects on fish, invertebrates, and human. The importance of this study lies in the fact that long-term ingestion of heavy metal-contaminated fish can result in the accumulation of harmful metals in numerous organs and pose a major risk to human health. AIM: The current study was designed to investigate the concentrations of toxic arsenic (As), lead (Pb), and mercury (Hg) in the liver, gills, and muscles of highly consumed aqua cultured common carp (Cyprinus carpio L.) in Baqubah city, to evaluate the toxico

Vitamin D ameliorates Aeromonas hydrophila-induced iron-dependent oxidative damage of grass carp splenic macrophages by manipulating Nrf2-mediated antioxidant pathway.

Aeromonas hydrophila (A. hydrophila) is one of major pathogenic bacteria in aquaculture and potentially virulent to grass carp (Ctenopharyngodon idella). As an essential nutrient for fish, vitamin D3 (VD3) has been reported to play a role against oxidative stress, but the exact mechanism remains to be elusive. In this study, we found that A. hydrophila induced ferrugination and macrophage aggregation in the spleen of grass carp. Along this line, using the splenic macrophages as the model, the effects of VD3 on A. hydrophila-caused iron deposition and subsequent injuries were determined. In the

Histopathological evaluation of the infection elicited by three species of Myxobolus (Cnidaria: Myxosporea), with identification of Myxobolusxiaoganensis n. sp. isolated from the silver carp Hypophthalmichthys molitrix.

Several parasites infecting the Asian carp species have been broadly spread along with the global fish trade. However, the diversity of specific parasite groups and their pattern of parasitism remain insufficiently elucidated even in their native regions. Here, we conducted a holistic identification and histological analysis of three Myxobolus species. The oblate myxospores of the first isolates were found infecting the spleen of silver carp Hypophthalmichthys molitrix, measuring 6.4 ± 0.4 (5.4-7.1) μm in length, 8.2 ± 0.4 (7.5-9.0) in width, and 5.9 ± 0.3 (5.2-6.2) in thickness. Hepatotoxicity in carp (Cyprinus carpio) exposed to environmental levels of norfloxacin (NOR): Some latest evidences from transcriptomics analysis, biochemical parameters and histopathological changes.

Recently, the residues of quinolones have received widespread attention. However, toxicological studies on aquatic organisms are relatively scarce, especially on the liver metabolism and immune effects of these aquatic organisms. In this study, we investigated the toxic effects of carp exposed to 0, 100 ng/L, and 1 mg/L norfloxacin (NOR) at environmental concentrations for 42 days. In this study, through transcriptomics analysis, we found that some genes involved in lipid metabolism, immune response, and cytochrome P450 metabolism, especially genes accounting for the metabolism-related disorde

Genomic polymorphisms at the crhr2 locus improve feed conversion efficiency through alleviation of hypothalamus-pituitary-interrenal axis activity in gibel carp (Carassius gibelio).

Improvement in fish feed conversion efficiency (FCE) is beneficial for sustaining global food fish supplies. Here, we show that a set of polymorphisms at locus of the corticotropin releasing hormone receptor 2 (crhr2), which is involved in hypothalamus-pituitary-interrenal (HPI) axis signaling, is associated with improved FCE in farmed allogynogenetic gibel carp strain CAS III compared with that in the wild gibel carp strain Dongting (DT). This set of polymorphisms downregulates the expression levels of crhr2 mRNA in the brain and pituitary tissues in gibel carp strain CAS III compared with th

Molecular characterization and functional analysis of interferon regulatory factor 9 (irf9) in common carp Cyprinus carpio: a pivotal molecule in the Ifn response against pathogens.

In the present study, interferon (IFN) regulatory factor (IRF) 9 gene (irf9) was identified and characterized in common carp Cyprinus carpio. The predicted protein sequence of Irf9 contains a DNA binding domain (DBD) that possess five tryptophans, an IRF association domain (IAD) and two nuclear localisation signals (NLS). Alignment of Irf9 of C. carpio with the corresponding Irf9 proteins of other species showed that the DBD is more highly conserved than the IAD. The putative Irf9 protein sequence of C. carpio shares higher identities with teleosts (53.8-82.3%) and lower identities with mammal

Enhanced protective immunity against spring viremia of carp virus infection can be induced by recombinant subunit vaccine conjugated to single-walled carbon nanotubes.

As a successful prevention strategy for controlling the highly contagious and pathogenic disease of spring viremia of carp (SVC), DNA vaccines reported in recent years could trigger protective responses against SVC with the means of injection. However, there remains many concerns and uncertainties related to DNA vaccination as well as injection is labor intensive, costly and not suitable to vaccinate large numbers of fish. Therefore, more efficient and safe prophylactic measures should be urgently investigated. In this research, single-walled carbon nanotubes (SWCNTs) as the candidate SWCNTs-p

Expression profile of carp IFN correlate with the up-regulation of interferon regulatory factor-1 (IRF-1) in vivo and in vitro: the pivotal molecules in antiviral defense.

Interferon regulatory factors (IRFs) are a family of transcription factors that mediate the transcriptional regulation of interferon (IFN) genes and IFN-inducible genes. In this study, IRF-1 gene is cloned from the common carp, Cyprinus carpio L., named CcIRF-1. The full-length cDNA of CcIRF-1 is 1427 bp, including an open reading frame of 861 bp encoding a protein of 286 amino acids. The putative CcIRF-1 is characterized by a conserved DNA-binding domain and includes a signature of six conserved tryptophan residues. The genomic sequence of CcIRF-1 is described, which consists of 9 exons and 8

Effects induced by feeding organochlorine-contaminated carp from Saginaw Bay, Lake Huron, to laying White Leghorn hens. II. Embryotoxic and teratogenic effects.

Carp from Saginaw Bay, Lake Huron, MI, was fed to White Leghorn chickens for a period of 8 wk. The diets contained 0.3 (control; 0% carp), 0.8 (3.4% carp), and 6.6 (35% carp) mg polychlorinated biphenyls (PCBs)/kg diet, by wet weight (ww). These concentrations corresponded to 3.3, 26, and 59 pg 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) equivalents/g diet ww, respectively. Though the diets were not acutely toxic to the adult laying hens, dose- and time-dependent responses were observed in the embryos and chicks. Toxicity was manifested as a dose-dependent increase in embryo mortality and decre
