Pathogenic mitochondrial DNA mutations inhibit melanoma metastasis.

Mitochondrial DNA (mtDNA) mutations are frequent in cancer, yet their precise role in cancer progression remains debated. To functionally evaluate the impact of mtDNA variants on tumor growth and metastasis, we developed an enhanced cytoplasmic hybrid (cybrid) generation protocol and established isogenic human melanoma cybrid lines with wild-type mtDNA or pathogenic mtDNA mutations with partial or complete loss of mitochondrial oxidative function. Cybrids with homoplasmic levels of pathogenic mtDNA reliably established tumors despite dysfunctional oxidative phosphorylation. However, these mtDN

Liquid Biopsies for Early Response Evaluation of Radium-223 in Metastatic Prostate Cancer.

PURPOSE: Reliable biomarkers for response monitoring during radium-223 treatment in patients with metastatic castration-resistant prostate cancer (mCRPC) are lacking. Circulating tumor cells (CTCs) and circulating tumor DNA (ctDNA), obtained from liquid biopsies, are shown to have prognostic value in mCRPC. The aim of this study was to determine the value of CTCs and ctDNA for response evaluation of radium-223. METHODS: In this prospective multicenter study, longitudinal blood draws and imaging were performed in 28 patients with mCRPC and predominantly bone disease, who were treated with radi…

Tumor cell-based liquid biopsy using high-throughput microfluidic enrichment of entire leukapheresis product.

Circulating Tumor Cells (CTCs) in blood encompass DNA, RNA, and protein biomarkers, but clinical utility is limited by their rarity. To enable tumor epitope-agnostic interrogation of large blood volumes, we developed a high-throughput microfluidic device, depleting hematopoietic cells through high-flow channels and force-amplifying magnetic lenses. Here, we apply this technology to analyze patient-derived leukapheresis products, interrogating a mean blood volume of 5.83 liters from seven patients with metastatic cancer. High CTC yields (mean 10,057 CTCs per patient; range 100 to 58,125) reveal

Frequent hypomethylation of PTGS2 gene promoter in human term placenta.

BACKGROUND: Gene expression profiles of several tumor suppressor genes are regulated by the methylation and demethylation of their promoters. Here, we aim to identify and quantify the methylation status of four tumor suppressor genes from placentas at term and compare them with the maternal white-blood-cells. METHODS: In order to achieve this objective, DNA enriched from twenty placentas at term and maternal white blood cells was bisulfite-converted and amplified using quantitative real-time methyl-light polymerase chain reaction for the four-genes studied (RASSF1A, APC, RAR-beta, and PTGS2).

Metal accumulation in paired colon cancer and adjacent tissues and its relationship with genotoxic and epigenetic biomarkers.

BACKGROUND: Environmental exposure to toxic and trace metals has been implicated in colorectal carcinogenesis through oxidative DNA damage and epigenetic dysregulation. However, human evidence linking tissue-level metal accumulation with epigenetic and genotoxic biomarkers remains limited, particularly in studies using paired tumor and adjacent non-cancerous colon tissues. OBJECTIVES: This study aimed to (i) characterize concentrations of selected toxic and trace metals in paired cancerous and non-cancerous colon tissues, (ii) examine associations between tissue metal concentrations and globa

Biomarkers in Breast Cancer: Where Are We and Where Are We Going?

Biomarkers play an important role in the detection and management of patients with breast cancer. Thus, BRCA1/2 mutation testing is used for risk assessment in families with a high prevalence of breast and ovarian cancer. Following a diagnosis of breast cancer, measurement of multi-analyte profiles such as uPA/PAI-1 or Oncotype DX may be used for determining prognosis and identifying lymph node-negative patients who may be spared from having to receive adjuvant chemotherapy. Other -gene tests such as the PAM50 ROR, Breast Cancer Index, and EndoPredict have been reported to predict the developm

Utility of DNA methylation markers for diagnosing cancer.

DNA methylation occurs at the CpG residues and serves as a powerful epigenetic mechanism that negatively regulates gene expression. This process is catalyzed by DNA methyltransferases and occurs within "CpG islands" found in the promoter regions of >70% of human genes. Given the important role of DNA methylation in regulating gene expression, un-programmed changes in methylation patterns are expected to either silence or activate transcription of tumor suppressor genes (via hypermethylation) or oncogenes (via demethylation), respectively, and by doing so promote a disease state. In light of th

Clinical validation of an HPV whole-genome sequencing assay for MRD detection in patients with HPV+ head and neck cancer treated with surgery.

Surgery is a standard treatment for early-stage human papillomavirus-associated head and neck cancer (HPV+HNC), yet adjuvant therapy decisions rely on clinicopathologic features with limited prognostic accuracy. Circulating tumor HPV DNA is a promising biomarker, but current assays lack the sensitivity needed for detecting minimal residual disease after surgery. This study evaluated the prognostic value of an HPV whole-genome sequencing assay, HPV-DeepSeek, and compared this with existing methods and clinical standards. One hundred three patients with stage I to IV HPV+HNC treated with surgery

Genomic profiling for copy number changes in plasma of ovarian cancer patients - a new era for cancer diagnostics?

A blood test that can detect human malignancy with high clinical sensitivity and specificity is highly desirable. To achieve this, a tumor marker is needed that correlates with tumor burden and that can be measured with high analytical sensitivity and specificity. Over the past decades, a number of different types of tumor markers have emerged, including proteins such as enzymes, glycoproteins, and oncofetal antigens. Besides proteins, genetic abnormalities such as mutations, amplifications, and circulating tumor DNA have served as tumor markers. Despite the diversity of such biomarkers, their…

NSCLC: Five trials to watch.

Despite advances in targeted therapy and immunotherapy, non-small cell lung cancer (NSCLC) has yet to be managed as a chronic disease. The emergence of circulating tumor DNA (ctDNA)/molecular residual disease (MRD)-guided adaptive treatment and novel agents, including KRAS G12D inhibitors, trophoblast cell surface antigen 2 (TROP2)-antibody-drug conjugates (ADCs), and PD-1/VEGF bispecific antibodies, is creating progress toward this goal. Five relevant clinical trials may reshape clinical practice, advance precision treatment, and bring this vision closer.
