Adjuvant effects of bacillus Calmette-Guerin DNA or CpG-oligonucleotide in the immune response to Taenia solium cysticercosis vaccine in porcine.

The immune stimulation properties of CpG-oligonucleotides (CpG-ODN) containing a central unmethylated CpG motif could be useful for vaccination against parasite infection. However, the high cost of synthetic CpG-ODN has limited its use in veterinary vaccines. In this study, we investigated whether genomic DNA derived from Mycobacterium bovis bacillus Calmette-Guerin (BCG-DNA) could be used as an effective adjuvant to enhance the immunogenicity and the protective capacity of recombinant cC1 antigen (rcC1) against pig cysticercosis. Pigs were vaccinated with rcC1 plus CpG-containing DNA adjuvant

Alveolar echinococcosis: characterization of diagnostic antigen Em18 and serological evaluation of recombinant Em18.

The Echinococcus multilocularis protein Em18 is one of the most promising antigens for use in serodiagnosis of alveolar echinococcosis in human patients. Here we identify an antigenic relationship between Em18 and a 65-kDa immunodominant E. multilocularis surface protein previously identified as either EM10 or EmII/3. The NH(2)-terminal sequence of native Em18 was determined, revealing it to be a fragment of EM10. Experiments were undertaken to investigate the effect of proteinase inhibitors on the degradation of EM10 in crude extracts of E. multilocularis protoscoleces. Em18 was found to be t

Identification of human intestinal parasites affecting an asymptomatic peri-urban Argentinian population using multi-parallel quantitative real-time polymerase chain reaction.

BACKGROUND: In resource-limited countries, stool microscopy is the diagnostic test of choice for intestinal parasites (soil-transmitted helminths and/or intestinal protozoa). However, sensitivity and specificity is low. Improved diagnosis of intestinal parasites is especially important for accurate measurements of prevalence and intensity of infections in endemic areas. METHODS: The study was carried out in Orán, Argentina. A total of 99 stool samples from a local surveillance campaign were analyzed by concentration microscopy and McMaster egg counting technique compared to the analysis by mu

Molecular vaccines against parasites.

Prophylactic vaccines can be expected to be one of the major practical outputs of parasitology research. Various groups within Australia have pursued the vaccine objective for several years, with particular emphasis on blood-stage falciparum malaria in man, intestinal helminths of sheep and cattle, cutaneous myiasis (blowfly strike) in sheep, cysticercosis in sheep and cattle, bovine babesiosis, and cattle ticks. Other vaccine programmes are concerned with giardiasis, filariasis, toxoplasmosis, fascioliasis, coccidiosis in poultry, cutaneous leishmaniasis and schistosomiasis japonica. For many

Complete rDNA of Diplodiscus japonicus and Diplodiscus mehrai reveals IGS as a promising marker for intraspecific genetic variation.

Diplodiscus japonicus (Yamaguti, 1936) and Diplodiscus mehrai (Pande, 1937) (Trematoda: Diplodiscidae) are parasitic trematodes infecting wood frogs (Anura: Ranidae) in Heilongjiang Province, China. In this study, we amplify the complete rDNA of five D. japonicus and five D. mehrai. The assembled sequences were processed using DNASTAR and MEGA X to evaluate the sequence length and genetic divergence of the 18S, ITS1, 5.8S, ITS2, 28S rRNA, and IGS regions. To quantitatively assess the intraspecific genetic polymorphism, the nucleotide diversity (π) and haplotype diversity (Hd) for each rDNA reg

Molecular characterization of Fasciola jacksoni from wild elephants (Elephas maximus maximus) of Sri Lanka: a taxonomic evaluation.

Fasciola jacksoni is a significant contributor to the health and mortality of Asian elephants, particularly those in Sri Lanka. Despite the impact of fascioliasis on elephant populations, it is a neglected veterinary disease with limited taxonomic understanding. Molecular characterization and phylogenetic analysis of F. jacksoni were carried out to evaluate its suggested basal position in the Fasciolidae. Adult worms were collected during post-mortem of elephants, and eggs were collected from living elephants in National parks across Sri Lanka. Using the mitochondrial genes nicotinamide dehydr

Human ocular onchocerciasis caused by Onchocerca lupi (Spirurida, Onchocercidae) in Iran.

Cases of canine onchocerciasis caused by Onchocerca lupi are increasingly reported from Europe and the western United States of America. The zoonotic role of this parasite had already been suspected in Europe as the clinical signs and histopathology seen in two ocular cases from Albania and the Crimean region were very similar to those of canine ocular onchocerciasis. In the most recent reports of human onchocerciasis, O. lupi has been morphologically and molecularly identified as the causative agent of ocular infestation in two patients from Turkey, and one patient from Tunisia. Here, we repo

Complex species structure of Lecithaster salmonis (Digenea: Lecithasteridae), a fish parasite in the Arctic and Pacific Northwest.

Studying complexes of cryptic or pseudocryptic species opens new horizons for the understanding of speciation processes, an important yet vague issue for the digeneans. We investigated a hemiuroidean trematode Lecithaster salmonis across a wide geographic range including the northern European seas (White, Barents, and Pechora), East Siberian Sea, and the Pacific Northwest (Sea of Okhotsk and Sea of Japan). The goals were to explore the genetic diversity within L. salmonis through mitochondrial (cox1 and nad5 genes) and ribosomal (ITS1, ITS2, 28S rDNA) marker sequences, to study morphometry of

Population genetics of the filarial worm wuchereria bancrofti in a post-treatment region of Papua New Guinea: insights into diversity and life history.

BACKGROUND: Wuchereria bancrofti (Wb) is the primary causative agent of lymphatic filariasis (LF). Our studies of LF in Papua New Guinea (PNG) have shown that it is possible to reduce the prevalence of Wb in humans and mosquitoes through mass drug administration (MDA; diethylcarbamazine with/without ivermectin). While MDAs in the Dreikikir region through 1998 significantly reduced prevalence of Wb infection, parasites continue to be transmitted in the area. METHODS: We sequenced the Wb mitochondrial Cytochrome Oxidase 1 (CO1) gene from 16 people infected with Wb. Patients were selected from 7

Molecular studies of Onchocerca volvulus isolates from Mexico.

DNA from Onchocerca volvulus from Oaxaca and Chiapas, Mexico were used as templates to amplify members of the O-150 Onchocerca specific repeat sequence family. The resulting PCR amplicons all hybridized with OVS2, an oligonucleotide that has been previously shown to recognize amplicons derived from O. volvulus with 100% sensitivity. However, when PCR products amplified from the O. volvulus specific plasmid pOVS134 were used as a probe, most samples did not hybridize. Similarly, when PCR products amplified from DNA isolated from adult O. volvulus from Oaxaca were used as a probe, amplicons from

Lamellodiscus louiseuzeti n. sp. (Monopisthocotyla: Diplectanidae: Lamellodiscinae): a parasite of Chaetodon hoefleri (Teleostei, Chaetodontidae) with distinct attachment organ morphology.

During a study on representatives of Diplectanidae (Monopisthocotyla) infecting Chaetodon hoefleri, a marine teleost from Senegal, we collected individuals belonging to a species of Lamellodiscinae, which showed a unique arrangement of the haptoral lamellodiscs, so far unreported in the literature, with the first lamella of the ventral lamellodisc open anteriorly, unlike the dorsal lamellodisc, which is closed. Due to this arrangement, we describe the species Lamellodiscus louiseuzeti n. sp.. Apart from the morphology of the lamellodiscs, the new species differs from its congeners by having a

New Contracaecum species (Nematoda, Anisakidae) parasitizing Ardea cocoi Linnaeus (Aves, Ardeidae) in a mixed infection: morphological and molecular studies in Argentina.

This study provides the description of a new species of Anisakidae, Contracaecum cocoi sp. nov., as well as the record of Contracaecum jorgei, both species parasitizing the Cocoi heron Ardea cocoi (Ardeidae) in a locality from the Buenos Aires Province, Argentina. An integrative taxonomic approach was used, involving phylogenetic analyses and the examination of diagnostic morphological features in the studied specimens. Among other features, the new species can be morphologically distinguished by the papillae arrangement on the male tail: while C. jorgei exhibits a simple morphotype A, C. coco

A new species of the genus Monoecocestus (Cestoda: Anoplocephalidae) from Lagidium viscacia (Rodentia: Chinchillidae) from the high-altitudes of Peru.

Anoplocephalidae (Cestoda: Cyclophyllidea) is a diverse family, with the subfamily Anoplocephalinae comprising 59 genera that primarily parasitize the digestive tract of rodents and birds. Monoecocestus threlkeldi was originally described from the northern vizcacha, Lagidium peruanum, in Junin, Peru, and subsequently reported in cricetids (Holochilus spp.) in Argentina and Bolivia. In the present study, we describe a new species of the genus Monoecocestus parasitizing the southern vizcacha, Lagidium viscacia, based on specimens collected in Puno, Peru, at an elevation of 3932 m a. s. l. Assessing the diversity of freshwater fish trematodes from Laguna Escondida, Los Tuxtlas tropical rainforest, Mexico, using morphology and 28S rDNA sequences as barcodes.

Despite a great effort made for almost 90 years, the diversity of freshwater fish trematodes in Mexico is still far from being fully known. The addition of molecular data to the description of trematode diversity in the last two decades added the potential to establish more robust species limits and a more accurate biodiversity estimation, but also led in some instances to the recognition of cryptic species complexes. Here, we used sequences of the large subunit of the nuclear ribosomal gene (28S rRNA) as barcodes, and morphological data, to assess the diversity of freshwater fish trematodes f

A case of asymptomatic infection of Ascaris suum identified by PCR-restriction fragment length polymorphism and DNA sequence analysis.

Human ascariasis is a soil-transmitted helminthiasis and remains a neglected tropical disease. Ascaris suum has the potential to cause cross-infections between humans and pigs. In this study, we present a rare case of a patient with asymptomatic infection by Ascaris suum. A 66-year-old male underwent colonoscopy, and a white linear worm body was found in the hepatic curvature. The worm was collected by aspiration and submitted to the laboratory for parasite identification. The patient had no symptoms related to parasitic infection. The worm was highly suspected to be of the genus Ascaris. Beca

Genetic evidence for hybridisation between Haemonchus contortus and Haemonchus placei in natural field populations and its implications for interspecies transmission of anthelmintic resistance.

Genetic hybridisation between parasitic nematode species has potentially important consequences. It could lead to the introgression of genes between species including those involved in pathogenicity, host specificity, transmission and drug resistance. It could also complicate diagnosis and control. However, there are few compelling examples of its occurrence in parasites in the field. Haemonchus contortus and Haemonchus placei are two closely related parasitic nematode species that predominantly infect small ruminants and cattle, respectively. They are capable of experimental hybridisation whe

PCR-based differentiation of Fasciola species (Trematoda: Fasciolidae), using primers based on RAPD-derived sequences.

The zoonotic liver flukes Fasciola hepatica and F. gigantica co-exist in parts of Africa and Asia. The two species have similar life-cycles but different transmission characteristics. Although the identification of adult Fasciola to species level is traditionally based on differences in size and shape, recent studies have demonstrated this method to be unreliable. Species of Fasciola can be distinguished by staining and comparing the morpho-anatomy of the gut and ovaries or by iso-enzyme analysis but such approaches are time-consuming and require specialist skills. Two primer sets, based on RA
