Genomic and functional characterization of a novel halophilic bacteriophage targeting carbapenem-resistant Klebsiella pneumoniae.

Carbapenem-resistant Klebsiella pneumoniae (CRKP) is a multidrug-resistant (MDR) pathogen causing severe infections in immunocompromised patients, prompting the exploration of alternative therapies like bacteriophage therapy. In this study, we isolated and characterized a novel halophilic lytic bacteriophage, Halo KS-7, targeting K. pneumoniae, and used an AI-driven annotation pipeline in Python to analyze its genome and therapeutic potential. Bacteriophages were isolated from Hospital wastewater, purified through plaque isolation, and confirmed using the double-layer agar method. Morphologica

RanDeL-Seq: a High-Throughput Method to Map Viral cis- and trans-Acting Elements.

It has long been known that noncoding genomic regions can be obligate cis elements acted upon in trans by gene products. In viruses, cis elements regulate gene expression, encapsidation, and other maturation processes, but mapping these elements relies on targeted iterative deletion or laborious prospecting for rare spontaneously occurring mutants. Here, we introduce a method to comprehensively map viral cis and trans elements at single-nucleotide resolution by high-throughput random deletion. Variable-size deletions are randomly generated by transposon integration, excision, and exonuclease c

Final report of the safety assessment of methacrylic acid.

Methacrylic Acid is an organic acid used at concentrations between 50 and 88 percent to pretreat the nail and maximize the adhesion between the nail and artificial nail extender. Methacrylic Acid is readily absorbed through mucous membranes of the lungs, the gastrointestinal tract, and the skin; and is distributed to all major tissues. Oral LD50 values for rats ranged from 277 to 2260 mg/kg; acute toxicity symptoms included severe gastric irritation, gasping, labored respiration, prostration and hematuria. In a short-term inhalation study, rats exposed to Methacrylic Acid at 1300 ppm showed no

Comparison between Go-GutDx, a novel diagnostic stool test kit with potential impact in low-income countries, and BioFire test.

INTRODUCTION: Infectious diarrheal diseases are one of the leading causes of worldwide morbidity and mortality. The incidence of diarrhea is higher in Low-Middle-Income Countries (LMIC), where more than 90% of deaths from diarrheal diseases occur. Diagnostic tests for infectious diarrhea are not readily available in Low-Middle-Income Countries. Our study evaluates a novel, cost-effective, easy-to-use DNA stool testing kit for infectious diarrhea that can easily be rolled out in low-resource settings and has comparable performance to current testing modalities in the USA in terms of diagnostic

Clinical and molecular prognostic factors in operable laryngeal cancer.

Many factors affect the prognosis in operable laryngeal squamous cell carcinoma (LSCC). Many clinical factors have been implicated in tumor recurrence and poor survival of the patients. The aim of the present study is to investigate the demographic, clinical and histological characteristics as prognostic factors. Moreover, our aim is to analyze the role of modern molecular biomarkers in the prognosis of patients with LSCC. One hundred patients with operable laryngeal carcinoma underwent surgery as primary treatment between April 1999 and April 2002. Ninety-four of them were men and 6 women, wi

Potential of SERINC2 as a biomarker in tumor prognosis and immunology: analysis from pan-cancer studies.

Serine incorporator (SERINC) is a family of transmembrane protein involved in lipid synthesis. Among its family members, SERINC2 has been implicated in tumor pathogenesis. Pancreatic carcinoma (PAAD), a highly malignant tumor characterized by an extremely poor prognosis, lacks effective biomarkers. To date, the association between SERINC2 and pan-cancer or tumor immunity remains unreported in the literature. Consequently, investigating the utility of SERINC2 for prognostic prediction in PAAD lays the groundwork for developing diagnostic biomarkers and SERINC2-targeted immunotherapy strategies.

Comprehensive computational analysis of differentially expressed miRNAs and their influence on transcriptomic signatures in prostate cancer.

Prostate cancer presents a major health issue, with its progression influenced by intricate molecular factors. Notably, the interplay between miRNAs and changes in transcriptomic patterns is not fully understood. Our study seeks to bridge this knowledge gap, employing computational techniques to explore how miRNAs and transcriptomic alterations jointly regulate the development of prostate cancer. The study involved retrieving miRNA expression data from the GEO database specific to prostate cancer. Identification of DEMs was conducted using the 'limma' package in R. Integration of these DEMs wi

Bacterial outer membrane vesicles bound to bacteriophages modulate neutrophil responses to bacterial infection.

Pseudomonas aeruginosa is a major human pathogen, particularly effective at colonizing the airways of patients with cystic fibrosis. Bacteriophages are highly abundant at infection sites, but their impact on mammalian immunity remains unclear. We previously showed that Pf4, a temperate filamentous bacteriophage produced by P. aeruginosa, modifies the innate immune response to P. aeruginosa infections via TLR3 signaling, but the underlying mechanisms remained unclear. Notably, Pf4 is a single-stranded DNA and lysogenic phage, and its production does not typically result in lysis of its bacteria
