Long non‑coding RNA LINC01960‑201 hinders decidualization of endometrial stromal cell in endometriosis: Relevance to endometrial receptivity.

INTRODUCTION: Significant health problems encountered in adulthood often have their roots in health behaviours initiated during adolescence. In order to reverse this trend, school and health personnel, as well as parents and other community members working with high school students, need to be aware of the health-related beliefs and choices that guide the behaviours of teenagers. Although a wide variety of research has been conducted on this topic among urban adolescents, less is known about the health beliefs and behaviors of adolescents residing in rural areas, particularly in Canada. In gen

ADAM9: an updated view of its biology and pathology.

A disintegrin and metalloprotease 9 (ADAM9) is a catalytically active member of the ADAM family, a group of transmembrane and secreted proteins that regulate adhesion, proteolysis, migration, and cell signaling. ADAM9 exists in two isoforms, ADAM9-L (membrane bound form) and ADAM9-S (secreted form), which differ in structure and have opposing roles in cell migration. Its multiple domains support ectodomain shedding, integrin binding, intracellular signaling, and interactions with SH3-containing proteins. The activity of ADAM9 is regulated at different levels through transcriptional, post-trans

Developmental landmarks and cellular transitions during extravillous trophoblast cell differentiation.

Human trophoblast stem (TS) can be captured, maintained in vitro under specific conditions, and differentiated into extravillous trophoblast (EVT) cells. The regulatory mechanisms that govern the self-renewal and differentiation of human TS cells into EVT cells are largely unknown. In this study, bulk RNA-sequencing (RNA-seq) and single cell RNA-seq (scRNA-seq) were performed on human TS cells maintained in the stem state and on cells progressing from the stem state into EVT cells (differentiation days 3, 6, and 8). Distinct bulk and single cell transcript profiles were identified for each day

Inhibition of the Metalloproteinase ADAMTS5 Suppresses Colorectal Cancer Metastasis via the PEDF/Wnt/β-Catenin Pathway.

At present, colorectal cancer (CRC) ranks as the third most prevalent cancer globally and is the second most common cause of mortality associated with cancer. The expression of A Disintegrin and Metalloproteinase with Thrombospondin Motifs 5 (ADAMTS5) is upregulated in CRC, and high ADAMTS5 expression strongly correlates with an unfavorable prognosis. However, the function of ADAMTS5 in CRC remains unknown. This study aimed to investigate the role of ADAMTS5 in CRC and its potential mechanisms of action. The results showed that ADAMTS5 expression was higher in CRC tissues than in paracancerous

ADAMTS-7 Increase Contributes to Worsened Neurological Outcome in Mice on High Fat Diet.

BACKGROUND: Obesity and associated metabolic disturbance are associated with worse outcomes after brain ischemia. Cerebrovascular remodeling mediated by matrix metalloproteinase 9 (MMP-9) may contribute to this poor outcome. A disintegrin and metalloproteinase-7 (ADAMTS-7) has been shown to regulate vascular remodeling in peripheral vessels. This study was designed to determine the role of ADAMTS-7 in regulating MMP-9 activity, cerebrovascular remodeling and neurological outcomes in mice. METHODS: Six-week-old CD-1 male mice were fed regular diet (RD) or high fat diet (HFD) for 10 weeks befor

Homeostatic imbalance between von Willebrand factor and ADAMTS13 in hemorrhagic shock-induced endotheliopathy.

Endothelial dysfunction in patients after severe injury with hemorrhagic shock is associated with excessive release of ultralarge von Willebrand factor (VWF) without a corresponding increase in ADAMTS13 (A disintegrin and metalloprotease thrombospondin type 1 motifs, member 13), resulting in acquired ADAMTS13 deficiency and VWF hyperadhesive activity. We hypothesized that the VWF to ADAMTS13 imbalance is exacerbated by increased VWF oxidation, rendering it resistant to cleavage by ADAMTS13. Plasma indices of endotheliopathy and coagulopathy in 100 hemorrhagic shock patients were obtained, ADAM

PLGA Nanoparticle-Mediated Sustained Release of Fisetin for Intra-Articular Therapy of Temporomandibular Joint Osteoarthritis.

Temporomandibular joint osteoarthritis (TMJOA) is a degenerative maxillofacial disorder marked by progressive cartilage degradation and subchondral bone resorption, severely compromising patients' quality of life. Intra-articular injection (IA), a standard route for conservative therapy, offers clinical advantages in safety and efficacy; however, outcomes remain limited due to short drug retention, poor tissue penetration, and variable agent efficacy, necessitating repeated administration. To overcome these limitations, fisetin-loaded poly (lactic-co-glycolic acid) nanoparticles (FST-PNP) were

ADAMTS4 elicits myeloid-derived immune cell recruitment and liver fibrogenesis in metabolic dysfunction-associated steatotic liver disease.

A disintegrin and metalloproteinase with thrombospondin motifs 4 (ADAMTS4) has been implicated in arthritis and lung fibroblast activation; however, its role in liver homeostasis and fibrogenesis remains largely unexplored. Here, we investigated the functional significance of ADAMTS4 in liver fibrosis. We found that hepatic ADAMTS4 mRNA expression was significantly elevated in patients with fibrotic steatohepatitis. In mouse models of liver fibrosis, genetic deletion of ADAMTS4 protected against liver fibrogenesis, accompanied by a marked reduction in the recruitment of myeloid-derived infiltr

EphA2 Silencing Attenuates IL-1β-Induced Chondrocyte Ferroptosis and Extracellular Matrix Degradation In Vitro.

This study investigated the role of erythropoietin-producing hepatocellular carcinoma receptor A2 (EphA2) in interleukin-1β (IL-1β)-induced chondrocyte injury and ferroptosis-associated alterations in osteoarthritis (OA). Primary rat chondrocytes were stimulated with IL-1β to establish an in vitro OA-like model. EphA2 was silenced using siRNA. Cell viability and apoptosis were assessed using MTT and TUNEL assays, respectively. Extracellular matrix (ECM) metabolism was evaluated by immunofluorescence and RT-qPCR. Ferroptosis-associated changes were assessed by measuring mitochondrial ferrous ir

FGF8 accelerates osteoarthritic chondrocyte hypertrophy by JunB-mediated cytokine perturbation.

Temporomandibular joint osteoarthritis (TMJOA) is recognized as one of the most important oral-maxillofacial degenerative diseases, and affects a large group of the population worldwide. The progression of TMJOA is accompanied by an imbalance of cytokines in the joint cavity and a slow but long-lasting degradation of the joint tissues. FGF8, an important member of the fibroblast growth factor family, has been shown to be up-regulated in the cavities of osteoarthritis joints. However, its role in TMJOA progression remains unclear. Here, we established a mouse TMJOA model by using unilateral ant

SIRPα Cleavage Is Associated with Inflammatory Response, Corticosteroid Nonresponse, and Worse Prognosis in Clinical and Preclinical Models of Alcohol-Associated Hepatitis.

Severe alcohol-associated hepatitis (sAH) has been associated with a high 30-day mortality rate. Corticosteroids are used in patients with sAH with limitations. There are no biomarkers measured upon hospitalization that predict responsiveness to corticosteroids. The role of signal regulatory protein (SIRP)-α, a receptor that regulates inflammation, has not been studied in patients with AH. In prospective test and validation cohorts of sAH patients treated with corticosteroids, plasma cytokines and SIRPα levels were measured. Studies in Sirpa-/- mice and in vitro studies were performed to chara

Acitretin-inspired medicinal chemistry for Alzheimer's disease: A biomarker-gated central nervous system (CNS) discovery framework.

Alzheimer's disease (AD) still lacks scalable oral strategies for early intervention. This article examines acitretin, a systemic retinoid used for severe psoriasis, as a clinically characterized scaffold for central nervous system (CNS) medicinal chemistry-not as a parent drug for direct repurposing or as a predefined candidate. Retinoic acid receptor/retinoid X receptor signaling can induce a disintegrin and metalloprotease 10 (ADAM10), the principal neuronal α-secretase of amyloid precursor protein, thereby encouraging non-amyloidogenic cleavage and soluble amyloid precursor protein-α (sAPP

Refining therapeutic targeting of interleukin-6 by signalling mode selectivity.

Interleukin-6 (IL-6) is a multifunctional cytokine that regulates immunity, tissue repair and metabolic homeostasis, while also driving chronic inflammation in a broad range of diseases. IL-6 exerts its effects through distinct signalling modes, including classic signalling via its membrane-bound receptor (IL-6R) and trans-signalling via the soluble IL-6R (sIL-6R), which expands IL-6 responsiveness to most if not all cell types. Therapeutic targeting of the IL-6 pathway - primarily through monoclonal antibodies neutralizing IL-6 or IL-6R and Janus kinase (JAK) inhibitors targeting downstream s

Efficacy of Plasma Exchange in Secondary Thrombotic Microangiopathy: A Case Report and Literature Review.

Thrombotic microangiopathy (TMA) is a severe syndrome characterized by thrombocytopenia, microangiopathic hemolytic anemia, and organ damage. It includes primary forms such as thrombotic thrombocytopenic purpura (TTP) and atypical hemolytic uremic syndrome, as well as secondary TMA, which can result from conditions such as autoimmune diseases, infections, and malignancies. While plasma exchange (PE) is a well-established treatment for thrombocytopenic purpura, its effectiveness against secondary TMA is debated, primarily because of the varying etiologies and pathomechanisms. There is a lack of

Thrombotic Microangiopathy Presenting With Multisystem Involvement and Acute Neurologic Deficits in a 61-Year-Old Female: A Case of Acquired Thrombotic Thrombocytopenic Purpura.

Thrombotic microangiopathy (TMA) is a rare, life-threatening hematologic syndrome characterized by microangiopathic hemolytic anemia, thrombocytopenia, and end-organ dysfunction. Among TMAs, thrombotic thrombocytopenic purpura (TTP), an emergent subtype caused by severe deficiency of a disintegrin and metalloproteinase with thrombospondin type 1 motif, member 13 (ADAMTS13), requires urgent recognition and prompt treatment. We present a 61-year-old woman who initially presented with abdominal pain and hematuria and was treated for a presumed urinary tract infection. She later developed chest pa

Photogenetic-Like Liposomes Disrupt Neuroligin-3 Dependency to Enhance Glioma Treatment.

Neuronal activity is shown to potentiate glioma initiation, progression, and/or metastasis. A key mechanism in neural regulation of brain cancer involves the activity-dependent cleavage and release of the synaptic adhesion molecule neuroligin-3 (NLGN3). Here, this report describes the preparation of optogenetics-like liposome Lip-CuRA, which is used to regulate the content of NLGN3 in neurons and mediate phototherapy in cancer cells. Lip-CuRA contains upconversion nanoparticles encapsulating CuS (CuS@PUCNPs), a visible light-activated neurotransmitter prodrug RuBi-GABA, and a disintegrin and m

Targeting Microglial CD49a Inhibits Neuroinflammation and Demonstrates Therapeutic Potential for Parkinson's Disease.

Persistent microglial activation drives chronic neuroinflammation, a characteristic pathological hallmark of neurodegenerative disorders, including Parkinson's disease (PD). Although integrin receptor CD49a (Itga1 gene) serves as a canonical biomarker of tissue-resident immune populations, its microglial expression patterns, functions, and signaling pathways have not been elucidated. In this study, we aim to investigate the impact of CD49a in hyperactivated microglia on PD pathogenesis and elucidate downstream signaling pathways. Specifically, we demonstrate microglia-enriched CD49a expression

Proteome-wide mendelian randomization identifies causal plasma proteins in interstitial lung disease.

Interstitial lung disease (ILD) has shown limited treatment advancements, with minimal exploration of circulating protein biomarkers causally linked to ILD and its subtypes beyond idiopathic pulmonary fibrosis (IPF). In this study, we aimed to identify potential drug targets and circulating protein biomarkers for ILD and its subtypes. We utilized the most recent large-scale plasma protein quantitative trait loci (pQTL) data detected from the antibody-based method and ILD and its subtypes' GWAS data from the updated FinnGen database for Mendelian randomization analysis.
