A novel liquefied gas based oral controlled release drug delivery system for liquid drug formulations.

A novel liquefied gas based drug delivery system for the oral delivery of liquid and semi-solid drug formulations is presented. The capsule-shaped system is equipped with a capillary as an element controlling the release rate. The delivery mechanism is based on a constant vapor pressure produced by isopentane as a low-boiling liquefied gas. The liquid drug valproic acid (VA) was used as a model compound. The viscosity was increased by the addition of povidone (PVP). The VA-PVP gel exhibited pseudoplastic rheological properties, the shear rate was above 0.1s(-1), similar to a Newtonian liquid.

From nature to nanoscale: advances, challenges, and preclinical translation of nanofiber drug delivery systems containing plant-derived active ingredients.

Since the early 1960s, nanotechnology has been a critical area of science, allowing for the development of sophisticated nanomaterials. Nanofibers, one of the most widely used nanotechnological drug delivery systems, have emerged as a highly versatile platform within modern pharmaceutical sciences. By combining various polymers with active herbal ingredients, these systems mimic the natural extracellular matrix and provide improved functions such as a high surface area-to-volume ratio, drug targeting, and controlled drug release. Preclinical studies demonstrate that phytochemical-containing na…

About the impact of medium chain fatty acids on the permeation enhancing effects of self-emulsifying drug delivery systems.

Self-emulsifying drug delivery systems (SEDDS) are widely utilized to enhance oral drug absorption; however, limited epithelial permeability remains a major barrier, particularly for hydrophilic drugs and macromolecules. Medium-chain fatty acids are promising permeation enhancers (PEs) due to their transient effects on epithelial barriers. In this study, caprylic acid (C8) and capric acid (C10) were systematically evaluated as PEs in SEDDS, with emphasis on the influence of ionization state and mode of presentation within the formulation. SEDDS were modified by incorporating C8 or C10 either a…

Research Advances on Organelle-Targeted Drug Delivery Systems for the Treatment of Brain Tumors and Central Nervous System Inflammation.

Central nervous system (CNS) inflammation and brain tumor treatment are constrained by the heterogeneity of the blood-brain barrier (BBB) and blood-brain tumor barrier (BBTB), as well as by the sequential barriers to drug delivery across lesions, target cells and subcellular organelles. Simply increasing brain exposure does not ensure that drugs reach their actual sites of action. This review systematically examines the pathological roles and therapeutic rationales of mitochondrial, lysosomal, nuclear, endoplasmic reticulum and Golgi apparatus dysfunction in neuroinflammation and glioblastoma

Microfluidic engineering of polymer-based drug delivery systems: From particle synthesis to organ-on-a-chip validation and application.

The convergence of microfluidic engineering and organ-on-a-chip (OoC) technology is redefining the development and preclinical validation of polymer-based drug delivery systems and biomedical preparations. This review presents a bidirectional framework: first, how microfluidics enables precision synthesis of polymer particles with controlled size, morphology and multifunctionality; second, how OoC platforms serve as physiologically relevant testbeds to evaluate these particles under dynamic, human-mimetic conditions. We examine droplet microfluidics principles for generating monodisperse polym…

Collagenase-mediated extracellular matrix targeting for enhanced drug penetration and therapeutic efficacy in nanoscale delivery systems for cancer therapy.

The highly fibrotic extracellular matrix (ECM) constitutes a major barrier that restricts the penetration and delivery efficiency of anticancer drugs, particularly in fibrotic tumors such as pancreatic cancer, in which cancer-associated fibroblasts (CAFs) secrete excess collagen and cross-linking enzymes to establish a mechanically resistant and poorly compressible tumor microenvironment (TME). This review summarized the pathological mechanisms underlying collagen enrichment in the ECM and its pivotal role in tumor drug resistance, with a systematic overview on recent advances in collagenase-b

Validated Stability-Indicating RP-HPLC Method for Nirmatrelvir in Self-Emulsifying Drug Delivery Systems: Formulation Characterization and Permeability Enhancement.

This study presents a novel analytical and formulation strategy to enhance the oral delivery and quality assessment of Nirmatrelvir, a poorly water-soluble antiviral agent. A self-emulsifying drug delivery system (SEDDS) was developed using Labrafac MC 60, ethanol, and Transcutol HP (55:25:20), resulting in a nanoemulsion with a droplet size of 145.23 ± 3.23 nm, low polydispersity index (0.189 ± 0.023), and high transmittance (98.97 ± 0.25%). The formulation exhibited rapid emulsification (< 90 s) and significantly improved permeability, achieving a fivefold increase (Papp: 4.20 × 10-6 cm/s) a

[Development of Tumor-targeting Drug Delivery Systems Based on an Understanding of Polymer Characteristics and the Tumor-specific Environment].

Tumor-specific active drug release from macromolecular antitumor drugs after tumor delivery is critical to achieve efficient cellular uptake of the active drug, thereby ensuring therapeutic efficacy. Upon reaching the tumor tissue, protease-facilitated depegylation of pegylated zinc protoporphyrin with ester bonds between PEG and ZnPP (esPEG-ZnPP) occurs, leading to a faster cellular uptake and superior antitumor efficacy compared to PEG-ZnPP with ether bonds (etPEG-ZnPP). This finding provides a viable strategy for achieving efficient tumor-specific drug release by utilizing an ester linkage

Triamcinolone-loaded self nano-emulsifying drug delivery systems for ocular use: An alternative to invasive ocular surgeries and injections.

Inflammation of the posterior segment of the eye is a severe condition and hard to cure as delivery of drugs to the inflammation site is inefficient. Currently, the primary treatment approach is ocular surgery or invasive ocular injections. Herein, we designed and developed a topically self nano-emulsifying drug delivery system (SNEDDs) to deliver triamcinolone acetonide (TCA) to the posterior segment of the eye. A screening based on TCA solubility was conducted on each excipient followed by preparation of various formulations using different ratios of the selected excipients. Vesicles of opti

Traditional Chinese medicine combined with hepatic targeted drug delivery systems: A new strategy for the treatment of liver diseases.

Liver diseases are clinically common and present a substantial public health issue. Many of the currently available drugs for the treatment of liver diseases suffer from limitations that include low hepatic distribution, lack of target effects, poor in vivo stability and adverse effects on other organs. Consequently, conventional treatment of hepatic diseases is ineffective. TCM is commonly used in the treatment of liver diseases worldwide, particularly in China, and has advantages over conventional therapy. HTDDS can be designed to enhance clinical efficacy in the treatment of liver diseases.

Impact of different emulsifiers on biocompatibility and inflammatory potential of Perfluorohexyloctane (F6H8) emulsions for new intravenous drug delivery systems.

BACKGROUND: Emulsions on the basis of Perfluorohexyloctane (F6H8), a semifluorinated alkane (SFA), have shown to dissolve and transport highly lipophilic compounds. It is unknown how F6H8-containing emulsions (F6H8-cEM) interact with compartment blood, the reticuloendothelial system (RES), or influence injured organs in vivo. The current study was conducted to investigate the in vitro biocompatibility of F6H8-cEM and their drug delivery properties. Afterward, an in vivo study was performed as a proof-of-concept study in a rat model of acute kidney injury (AKI), which focused on the potential i

Longevity and cost of implantable intrathecal drug delivery systems for chronic pain management: a retrospective analysis of 365 patients.

OBJECTIVES: Intrathecal drug delivery systems represent an important component of interventional strategies for refractory chronic pain syndromes. Continuous intrathecal administration of opioids results in higher subarachnoid drug concentrations, improved pain scores, and less frequent side effects when compared with systemic opioid administration. Substantial costs arise at the time of surgical implantation and at revision for battery depletion or treatment of a complication. Despite current widespread use, the real-world longevity and cost of implanted intrathecal pumps (ITP) has not been f…
