Refinement of a Methodology for Untargeted Detection of Serum Albumin Adducts in Human Populations.

Covalently modified blood proteins (e.g., serum albumin adducts) are increasingly being viewed as potential biomarkers via which the environmental causes of human diseases may be understood. The notion that some (perhaps many) modifications have yet to be discovered has led to the development of untargeted adductomics methods, which attempt to capture entire populations of adducts. One such method is fixed-step selected reaction monitoring (FS-SRM), which analyses distributions of serum albumin adducts via shifts in the mass of a tryptic peptide [Li et al. (2011) Mol. Cell. Proteomics 10, M110

Environmental factors and their interactions with risk genotypes in MS susceptibility.

PURPOSE OF REVIEW: The area of multiple sclerosis (MS) epidemiology has expanded during the last few years. Established lifestyle and environmental factors influencing MS risk are Epstein-Barr virus infection, sun exposure/vitamin D, and smoking. We review these factors and a series of other potential candidates implicated in the pathogenesis of MS and how environmental factors interact with genetic susceptibility with regard to disease risk. RECENT FINDINGS: On top of established MS-associated factors, there is now strong evidence for influence of adolescent obesity, exposure to organic solv

Optimization and head-to-head comparison of MISSR-PCR, ERIC-PCR, RAPD and 16S rRNA evolutionary clock for the genotyping of Vibrio cholerae isolated in China.

PURPOSE: To establish a new genotyping method for Vibrio cholerae and compare it with other methods. MATERIALS AND METHODS: In the current study, a modified inter simple sequence repeat-polymerase chain reaction (MISSR-PCR) system was developed via several rounds of optimisation. Comparison study was then conducted between MISSR-PCR and three other methods, including enterobacterial repetitive intergenic consensus sequences-based PCR (ERIC-PCR), randomly amplified polymorphic DNA (RAPD) and 16S rRNA evolutionary clock, for the detection and genetic tracing of Vibrio cholerae isolated from sea

Recognition and management of rodent-borne infectious disease outbreaks after heavy rainfall and flooding.

Climatic events, especially heavy rains and flooding following periods of relative drought, have precipitated both arthropod-borne and rodent-borne infectious disease outbreaks. Heavy rainfall encourages excessive wild grass seed production that supports increased outdoor rodent populations, and flooding forces rodents from their burrows near water sources into the built environment and closer to humans. The objectives of this review are to alert clinicians to the climatic conditions common to hurricane-prone regions, such as Louisiana, that can precipitate outbreaks of the two rodent-borne di

Positional plagiocephaly: an analysis of the literature on the effectiveness of current guidelines.

OBJECT: Positional plagiocephaly (PP) has been on the rise in recent years. In this review, the authors' aim was to assess the effectiveness of current recommendations to parents on this exceedingly common problem through a comprehensive literature search. Additionally, the current treatment options and the most recent studies on PP are reviewed. METHODS: A search of the existing literature was conducted to obtain all relevant studies on guidelines, recommendations, parental and clinician practices, and epidemiological aspects. RESULTS: Although the incidence and risk factors for PP have bee

Quantifying sources of environmental contamination with Toxocara spp. eggs.

A rich body of work has reported levels of infection with Toxocara species in definitive hosts, and the frequency of eggs in the environment, in many different regions and situations. These have greatly increased our understanding of the relationship between egg excretion from companion and wild animals and the risk of human infection by inadvertent ingestion of eggs from soil and other environmental reservoirs. Nevertheless, it is difficult to compare studies directly because of vagaries in sampling and laboratory methods, a preponderance of prevalence rather than abundance data, and a lack o

Closing the praziquantel treatment gap: new steps in epidemiological monitoring and control of schistosomiasis in African infants and preschool-aged children.

Where very young children come into contact with water containing schistosome cercariae, infections occur and schistosomiasis can be found. In high transmission environments, where mothers daily bathe their children with environmentally drawn water, many infants and preschool-aged children have schistosomiasis. This 'new' burden, inclusive of co-infections with Schistosoma haematobium and Schistosoma mansoni, is being formally explored as infected children are not presently targeted to receive praziquantel (PZQ) within current preventive chemotherapy campaigns. Thus an important PZQ treatment

[MicroRNAs in bipolar disorder: diagnostic and therapeutic applications].

INTRODUCTION: Bipolar disorder is a severe mental disease of unknown etiology that affects about 1% of the population. It is characterized by mood changes, alternating episodes of mania with depression. The current experimental and epidemiological data suggest that bipolar disorder represents a group of disorders with similar symptoms caused by the combination of genetic and environmental factors. Genetic analysis has identified several genes whose dysfunction might predispose to the disorder, although most of the results have not been confirmed in other studies. DEVELOPMENT: The aim of this

A review of African American-white differences in risk factors for cancer: prostate cancer.

OBJECTIVE: African American men have higher prostate cancer incidence rates than White men, for reasons not completely understood. This review summarizes the existing literature of race-specific associations between risk factors and prostate cancer in order to examine whether associations differ. METHODS: We reviewed epidemiologic studies published between January 1970 and December 2008 that reported race-specific effect estimates. We focused mainly on modifiable risk factors related to lifestyle and health. A total of 37 articles from 21 study populations met our inclusion criteria. RESULTS

Th1 and th2 cytokine expression in nasopharyngeal secretions during acute bronchiolitis in children younger than two years old.

OBJECTIVES: We characterised the T helper cytokine profiles on the surface of nasal mucosa of children with acute bronchiolitis caused by Respiratory Syncytial Virus, Parainfluenza Virus, Influenza Virus, Adenovirus, or without any viral identification, in order to examine whether these viral types modified cytokine responses. As an additional objective we sought to determine if T helper polarisation was associated with other demographic and environmental factors. METHODS: A prospective study of children with acute bronchiolitis was performed. Patients were recruited from the emergency depart

Consensus recommendations for managing asymptomatic persistent non-virus non-alcohol related elevation of aminotransferase levels: suggestions for diagnostic procedures and monitoring.

A persistent increase in non-virus non-alcohol related aminotransferase levels can have multiple causes, which differ in terms of prevalence and clinical importance. In the general population, the most frequent cause is non-alcoholic hepatic steatosis, which can evolve into steato-hepatitis and cirrhosis. The treatment for steatosis and non-alcoholic steato-hepatitis consists of modifying lifestyles, whereas the effectiveness of drug treatment remains to be determined. Other much less frequent (yet not rare) causes of persistent non-virus non-alcohol related elevations in aminotransferase leve

Gender differences in psychological effect of the August 1999 earthquake in Turkey.

Disasters traumatically expose normal populations to severe threats to life, deaths of relatives and massive environmental destruction. Epidemiological studies found that women would be more vulnerable to disaster than men. In this study, we examined gender differences in short-term psychological effect of the 1999 earthquake in Turkey on adult survivors. A total of 184 subjects (79 males and 105 females) who used a psychiatric service were interviewed within 6-10 weeks after the earthquake. All subjects were assessed using a modified version of the Mississippi Scale for Posttraumatic Stress D

[Prenatal exposure to androgens as a factor of fetal programming].

Both epidemiological and clinical evidence suggest a relationship between the prenatal environment and the risk of developing diseases during adulthood. The first observations about this relationship showed that prenatal growth retardation or stress conditions during fetal life were associated to cardiovascular, metabolic and other diseases in later life. However, not only those conditions may have lasting effects after birth. Growing evidence suggests that prenatal exposure to steroids (either of fetal or maternal origin) could be another source of prenatal programming with detrimental conseq

Therapeutic potential of adenosine A(2A) receptor antagonists in Parkinson's disease.

In the pursuit of improved treatments for Parkinson's disease (PD), the adenosine A(2A) receptor has emerged as an attractive nondopaminergic target. Based on the compelling behavioral pharmacology and selective basal ganglia expression of this G-protein-coupled receptor, its antagonists are now crossing the threshold of clinical development as adjunctive symptomatic treatment for relatively advanced PD. The antiparkinsonian potential of A(2A) antagonism has been boosted further by recent preclinical evidence that A(2A) antagonists might favorably alter the course as well as the symptoms of th

Biomarkers for low-level exposure causing epigenetic responses in stem cells.

Our current understanding of experimental in vitro and in vivo studies, as well as of epidemiological data, suggests that carcinogenesis is the result of many endogenous and exogenous factors interacting during a multi-step, multi-mechanism process. No single factor "causes" cancer. Carcinogenesis is known to involve both mutagenic and nonmutagenic processes. Therefore, the objective of this review is to examine how low dose exposure to ionizing radiation might contribute to the carcinogenic process and if measurable "biomarkers" might be used to monitor the susceptibility, exposure, biologica

Approach to the patient with prolonged granulocytopenia.

In most centers treating cancer patients, significant progress has been made in permitting patients to survive even prolonged courses of neutropenia. This has resulted from a better understanding of the epidemiology of infection and the points during the clinical course when they pose a risk for the patient with prolonged neutropenia. Considerable benefit has been derived from the availability of more potent broad-spectrum antimicrobial agents and from organized strategies for when they should be initiated, how and when they should be modified, and for how long they should be continued. [Therapy and prevention of reflux esophagitis. Results of a multicenter study with cimetidine. I: Epidemiology and results of acute therapy].

In a sequential observational study efficacy and safety of two dosage regimens of cimetidine for the treatment of reflux-oesophagitis (RE) were examined. 22 office-based specialists took part in the trial. 187 patients received 1600 mg cimetidine (400 mg q.i.d.) daily and 136 patients received 800 mg cimetidine (400 mg b.i.d.) daily, over twelve weeks. The two dosages proved to be equally effective. In the group of patients with RE of stage I at the beginning of the trial, the healing rates were 82% under 1600 mg cimetidine/die and 86% under 800 mg cimetidine/die. In the group of those patient

Natural history of the cervical precancerous lesions and their evolutions.

The numerous epidemiologic studies confirm that precancerous cervical lesions and C.I.S. are dependent on multiple mutagenic factors (chemical, virus and other nucleo-proteins, radiations and co-carcinogens, hormones, infections). These studies demonstrate that cervical cancer is a "sexually transmissible" disease. The C.I.N. have a monomorphic mechanism. Their point of departure is the undifferentiated stem cells. According to the number of mitosis, their epidermoid differentiation and their capacity to mature, the morphology of these lesions will vary (C.I.N. 1, 2, 3, dysplasias: slight, mod

Impact of the Recanalization Level and the First-Pass Effect on Functional Outcomes in Patients After M2 MCA Occlusion Thrombectomy.

Objective: Acute ischemic stroke (AIS) remains one of the most common causes of death and disability in the world. Mechanical thrombectomy (MT) is the modality of choice in the treatment of AIS and large vessel occlusion (LVO). The endovascular treatment of medium and distal vessel occlusions (DMVO) is currently under intensive scientific investigation. The aim of our study was to prove the feasibility, effectiveness and safety of MT in patients with a primary, isolated occlusion of the M2 segment of the middle cerebral artery (MCA), with a focus on the recanalization level and the first-pass
