First detection of Nyssorhynchus rondoniensis (Diptera: Culicidae) in southern Venezuela and its infection with Plasmodium falciparum.

Ongoing efforts to control vectors and eliminate malaria in the Neotropics are hampered by challenges that include accurate mosquito identification and acquisition of relevant local ecological data. In Bolivar state, southern Venezuela, 88 specimens of a potential malaria vector, Nyssorhynchus rondoniensis, were detected in several villages, identified by COI barcode sequences, and one mosquito was found infected with Plasmodium falciparum by real-time PCR, for an infection rate of 1.14 %. Increasingly, deforestation for a range of anthropogenic activities, especially mining, urbanization, and

DNA Barcoding for Species Identification and Conservation of Caimans (Crocodilia: Alligatoridae).

Systematics has become an essential aspect of managing and conserving species from the order Crocodilia. All members of the group are listed in CITES appendices, and to control illegal traffic of animals and subproducts, we must be able to correctly assess the specific identity of the samples. Genetic DNA barcoding is a very efficient tool for species identification in the animal kingdom, based on the sequencing of a region of a mitochondrial gene, the Cytochrome oxidase subunit I (COI). Our principal aims were to design specific primers that allow obtaining this sequence from the genetic mate

Molecular diversity of cox1 and LSU rDNA sequences of Sarcocystis bertrami (syn. S. fayeri) (Apicomplexa: Eucoccidiorida: Sarcocystidae) in horses.

Food poisoning caused by consuming raw horsemeat contaminated with Sarcocystis is a significant public health concern. Two morphotypes of sarcocysts in horsemeat, characterized by upright and folded villar protrusions, are typically identified as Sarcocystis fayeri and S. bertrami, respectively. However, recent molecular studies focusing on the ribosomal RNA gene (rDNA) and mitochondrial cytochrome c oxidase subunit I gene (cox1) have indicated a conspecific relationship between these two morphotypes using a limited number of specimens. To explore further genetic diversity in equid sarcocysts,

Genetic variation and population structure of Haemonchus contortus: an in-silico analysis.

Haemonchus contortus is a parasitic nematode that causes significant economic losses in ruminant livestock worldwide. In this study, we assessed the global genetic diversity and population structure of H. contortus using mitochondrial COX1 and ribosomal ITS2 sequences retrieved from the NCBI GenBank database. In total, 324 haplotypes of the COX1 and 72 haplotypes of the ITS2 were identified. The haplotype diversity values were all higher than 0.5, and the nucleotide diversity values were higher than 0.005. The Tajima's D value for COX1 (-1.65634) was higher than that for ITS2 (-2.60400). OXA1L deficiency causes mitochondrial myopathy via reactive oxygen species regulated nuclear factor kappa B signalling pathway.

BACKGROUND: OXA1L is crucial for mitochondrial protein insertion and assembly into the inner mitochondrial membrane, and its variants have been recently linked to mitochondrial encephalopathy. However, the definitive pathogenic link between OXA1L variants and mitochondrial diseases as well as the underlying pathogenesis remains elusive. METHODS: In this study, we identified bi-allelic variants of c.620G>T, p.(Cys207Phe) and c.1163_1164del, p.(Val388Alafs*15) in OXA1L gene in a mitochondrial myopathy patient using whole exome sequencing. Retrospective study of spirocercosis in coyotes (Canis latrans) in Costa Rica: epidemiology, pathological findings, and molecular characterization of Spirocerca lupi.

BACKGROUND: Spirocerca lupi is a nematode that infects domestic dogs and wild carnivores. METHODS: This study retrospectively analyzed postmortem records between 1989 and 2024 to assess S. lupi-associated lesions in coyote necropsies. In addition, it conducted molecular characterization of 18S rRNA and cytochrome oxidase subunit 1 (cox1) gene fragments of larvae found at necropsies. Fecal samples from free-ranging coyotes were molecularly examined for S. lupi 18S DNA. RESULTS: Of the 39 coyote cases, 33 (84. 6%) presented S. lupi-associated lesions. A significant association was observed bet

A novel real-time PCR for New World screwworm fly (Cochliomyia hominivorax) and its application in a non-destructive multiplex for efficient detection of screwworm flies.

Surveillance and diagnostics are critical for the early detection, containment and eradication of exotic pests. For the screwworm fly, this is usually via targeted surveillance and exclusion testing of trap-caught flies, as well as the identification of larvae associated with myiasis wounds. We present a specific and sensitive real-time polymerase chain reaction (PCR) assay for the detection of the New World screwworm fly, Cochliomyia hominivorax Coquerel (Diptera: Calliphoridae). The assay targets the cytochrome oxidase subunit 1 (cox1) gene from adult flies or larvae and retains high analyti

Demographic history and population structure of Phlebotomus argentipes (Diptera: Psychodidae) complex, the leishmaniasis vector in Sri Lanka.

Phlebotomus argentipes sensu lato Annandale & Brunetti, 1908 is the primary vector of Leishmania donovani MON 37, the causative agent of cutaneous leishmaniasis (CL) in Sri Lanka. Effective vector control is essential for managing leishmaniasis. Although numerous taxonomic studies have been conducted on P. argentipes s.l., population genetics remains insufficiently explored. This study investigated the demographic history and population genetic structure of P. argentipes s.l. in Sri Lanka using sequence data obtained from the earlier investigation of two mitochondrial markers, Cytochrome c oxi

The heme A synthase Cox15, as a target of redox-active 3-benzylmenadiones with antiparasitic activity.

Chagas disease, caused by Trypanosoma cruzi, is a neglected parasitic infection. The very limited arsenal of anti-T. cruzi treatments calls for the development of new drugs. Recently, a library of 3-benzylmenadione derivatives was synthesized, with cruzidione being the most efficient and specific compound against the parasite. To decipher its mode of action, we used the yeast Saccharomyces cerevisiae as a model. Evidence pinpointed at the heme A synthase Cox15 as a primary target of cruzidione: (i) a mutation in Cox15 (i.e., S429F) renders the yeast cells highly sensitive to the drug, (ii) tre

High temperature induces the upward shift of the thermal neutral zone and decreases metabolic capacity in zebra finches.

The thermal neutral zone (TNZ) represents a fundamental concept in the thermal physiology of homeothermic organisms. TNZ is characterized as a specific range of environmental temperatures within which the metabolic rate remains at its basal level. The ambient temperature is regarded as a critical environmental factor that affects an animal's thermoregulation and propels the development of various morphological, physiological, and behavioral adaptations. In the present investigation, we assessed the influence of environmental temperature on various physiological parameters, including body mass,

Air-liquid interface cultures trigger a metabolic shift in intestinal epithelial cells (IPEC-1).

An improved oxygen availability in air-liquid interface (ALI) cultures of enterocytes of the small intestine seems to be primarily responsible for morphological, metabolic, and functional changes. Intestinal porcine epithelial cells 1 (IPEC-1) are less investigated and are rarely used as model for intestinal barrier but showed a profound change of cell shape during ALI cultivation. We aim to answer the following question: Are the observed morphological effects accompanied by changes in metabolic function? A microarray analysis of submerged culture (SMC) and ALI cultures identified 830 signific

Molecular differentiation of Sarcocystis miescheriana and Sarcocystis suihominis using a new multiplex PCR targeting the mtDNA cox1 gene in wild boars in southern Italy.

The increase of wild boar populations density and their meat consumption across Europe could expose humans to a plethora of foodborne diseases as sarcocystosis, caused by the zoonotic protozoan Sarcocystis suihominis. Humans become infected by eating raw or undercooked pig (Sus scrofa domesticus) containing S. suihominis sarcocysts. Despite this, to date very few data are available on the risk of infection by this parasite to wild boar (Sus scrofa) meat consumers. Thus, the present study aimed to assess the occurrence of Sarcocystis spp. in wild boars from southern Italy, applying both histolo

A biallelic variant in COX18 cause isolated Complex IV deficiency associated with neonatal encephalo-cardio-myopathy and axonal sensory neuropathy.

Pathogenic variants impacting upon assembly of mitochondrial respiratory chain Complex IV (Cytochrome c Oxidase or COX) predominantly result in early onset mitochondrial disorders often leading to CNS, skeletal and cardiac muscle manifestations. The aim of this study is to describe a molecular defect in the COX assembly factor gene COX18 as the likely cause of a neonatal form of mitochondrial encephalo-cardio-myopathy and axonal sensory neuropathy. The proband is a 19-months old female displaying hypertrophic cardiomyopathy at birth and myopathy with axonal sensory neuropathy and failure to th

Genetic Diversity of Anopheles coustani (Diptera: Culicidae) in Malaria Transmission Foci in Southern and Central Africa.

Despite ongoing malaria control efforts implemented throughout sub-Saharan Africa, malaria remains an enormous public health concern. Current interventions such as indoor residual spraying with insecticides and use of insecticide-treated bed nets are aimed at targeting the key malaria vectors that are primarily endophagic and endophilic. Anopheles coustani s.l., an understudied vector of malaria, is a species previously thought to exhibit mostly zoophilic behavior. Like many of these understudied species, An. coustani has greater anthropophilic tendencies than previously appreciated, is often

How parasite exposure and time interact to determine Australapatemon burti (Trematoda: Digenea) infections in second intermediate hosts (Erpobdella microstoma) (Hirudinea: Erpodellidae).

Australapatemon spp. are cosmopolitan trematodes that infect freshwater snails, aquatic leeches, and birds. Despite their broad geographic distribution, relatively little is known about interactions between Australapatemon spp. and their leech hosts, particularly under experimental conditions and in natural settings. We used experimental exposures to determine how Australapatemon burti cercariae dosage (number administered to leech hosts, Erpobdella microstoma) affected infection success (fraction to encyst as metacercariae), infection abundance, host survival, and host size over the 100 days

Infestation of the Parasitic Isopod Mothocya parvostis on Juveniles of the Black Sea Bream Acanthopagrus schlegelii as an Optional Intermediate Host in Hiroshima Bay.

In Hiroshima Bay, parasitic isopods of the genus Mothocya infest the black sea bream Acanthopagrus schlegelii (Bleeker, 1854) and the Japanese halfbeak Hyporhamphus sajori (Temminck and Schlegel, 1846), two fish species that are abundant and commercially important in the Seto Inland Sea of Japan. Immature and mature Mothocya individuals can infect both juveniles and adults of H. sajori, while immature Mothocya are known to parasitize juveniles of A. schlegelii; i.e., no Mothocya parasites are found in adult A. schlegelii. The identification of the immature Mothocya parasitizing juveniles of A.

Molecular diversity of Bulinus species in Madziwa area, Shamva district in Zimbabwe: implications for urogenital schistosomiasis transmission.

BACKGROUND: Bulinus species are freshwater snails that transmit the parasitic trematode Schistosoma haematobium. Despite their importance, the diversity of these intermediate host snails and their evolutionary history is still unclear in Zimbabwe. Bulinus globosus and B. truncatus collected from a urogenital schistosomiasis endemic region in the Madziwa area of Zimbabwe were characterized using molecular methods. METHODS: Malacological survey sites were mapped and snails were collected from water contact sites in four communities in the Madziwa area, Shamva district for a period of one year,

Uncovering diversity and metabolic spectrum of animals in dead zone sediments.

Ocean deoxygenation driven by global warming and eutrophication is a primary concern for marine life. Resistant animals may be present in dead zone sediments, however there is lack of information on their diversity and metabolism. Here we combined geochemistry, microscopy, and RNA-seq for estimating taxonomy and functionality of micrometazoans along an oxygen gradient in the largest dead zone in the world. Nematodes are metabolically active at oxygen concentrations below 1.8 µmol L-1, and their diversity and community structure are different between low oxygen areas. This is likely due to toxi

DNA analysis of elasmobranch products originating from Bangladesh reveals unregulated elasmobranch fishery and trade on species of global conservation concern.

Trade involving elasmobranch products in Bangladesh is a four-decade-long practice in large scale and there is little understanding of its impact on species composition, population, and subsequent conservation. Capacity for monitoring and identification is lacking in landing and shark processing centres. A rapid survey and collection of tissue samples were performed in three landings and nine shark processing centres between 2016 and 2017 in the south-eastern coastal region of Bangladesh.
