Cytochrome c peroxidase regulates intracellular reactive oxygen species and methylglyoxal via enzyme activities of erythroascorbate peroxidase and glutathione-related enzymes in Candida albicans.

D-erythroascorbate peroxidase (EAPX1) deficiency causes glutathione deprivation, leading to the accumulation of methylglyoxal and reactive oxygen species (ROS), and especially, induction of cytochrome c peroxidase (Ccp1) in Candida albicans. Nevertheless, reciprocal effects between changes in Ccp1 activity and the antioxidative D-erythroascorbic acid- and glutathione-dependent redox status, which reflects methylglyoxal biosynthesis altering pathophysiology are unclear in eukaryotes. To elucidate the effect of CCP1 expression on EAPX1 and glutathione reductase (Glr1) activity-mediated D-erythro

Mutant alpha-galactosidase A enzymes identified in Fabry disease patients with residual enzyme activity: biochemical characterization and restoration of normal intracellular processing by 1-deoxygalactonojirimycin.

Fabry disease is a lysosomal storage disorder caused by the deficiency of alpha-Gal A (alpha-galactosidase A) activity. In order to understand the molecular mechanism underlying alpha-Gal A deficiency in Fabry disease patients with residual enzyme activity, enzymes with different missense mutations were purified from transfected COS-7 cells and the biochemical properties were characterized. The mutant enzymes detected in variant patients (A20P, E66Q, M72V, I91T, R112H, F113L, N215S, Q279E, M296I, M296V and R301Q), and those found mostly in mild classic patients (A97V, A156V, L166V and R356W) a

Dual control mechanism for heme oxygenase: tin(IV)-protoporphyrin potently inhibits enzyme activity while markedly increasing content of enzyme protein in liver.

Tin(IV)-protoporphyrin (Sn-protoporphyrin) potently inhibits heme degradation to bile pigments in vitro and in vivo, a property that confers upon this synthetic compound the ability to suppress a variety of experimentally induced and naturally occurring forms of jaundice in animals and humans. Utilizing rat liver heme oxygenase purified to homogeneity together with appropriate immunoquantitation techniques, we have demonstrated that Sn-protoporphyrin possesses the additional property of potently inducing the synthesis of heme oxygenase protein in liver cells while, concurrently, completely inh

Interaction of biochar with multiple amendment combinations affects plant biomass and soil enzyme activities.

Biochar is an important restoration amendment that is often applied together with other amendments, such as microbial inoculum, or organic and inorganic amendments. However, the mechanisms underlying biochar interactions with other restoration amendment combinations on soil functions and plant growth often remain unclear. Here we investigate the interaction of biochar with up to six additional restoration amendments using a novel experimental design. We test 6 distinct restoration amendments (amorphous silica, vermiculite, bentonite, AM fungi, potassium dissolving bacteria, and rhizobium) and

Cayman enables large-scale analysis of gut microbiome carbohydrate-active enzyme repertoires.

Carbohydrate-active enzymes (CAZymes) are crucial for digesting glycans, but tools for CAZyme profiling and interpretation of substrate preferences in microbiome data are lacking. Here we develop a CAZyme profiler called Cayman (Carbohydrate Active Enzymes Profiling of Metagenomes) and a hierarchical substrate annotation scheme for use with genomic or shotgun metagenomic datasets. Using these tools, we systematically surveyed CAZymes in human gut microorganisms (n = 107,683 genomes) and identified several putative mucin-foraging bacteria, including Hungatella and Eisenbergiella species, which

Accelerated discovery of highly active enzyme nanohybrids with parallelized Bayesian optimization in hybrid space.

Artificial intelligence (AI) has significantly advanced protein engineering, enabling rapid enzyme evolution for diverse applications. However, the fragile nature of biomacromolecules requires enzyme immobilization to preserve catalytic activity under harsh industrial conditions, which often restricts substrate diffusion and reduces enzymatic activity. This challenge demands extensive trial-and-error experiments to optimize immobilized carriers with high activity for different enzymes. Here we show a machine-learning-guided workflow along with an algorithm named parallelized hybrid-space Bayes

Analysis of the influence of rs1801265 polymorphism on DPD enzyme activity and contributing factors in the Chinese population.

INTRODUCTION: Dihydropyrimidine dehydrogenase (DPD), encoded by DPYD, is essential for 5-fluorouracil (5-FU) metabolism, yet functional variants remain understudied in Chinese populations. METHODS: We enrolled 300 healthy Chinese subjects and measured DPD activity as the HPLC-based 5-FU degradation rate in PBMCs. Five DPYD SNPs selected from NCBI and PharmGKB were genotyped by SNaPshot; genotype-degradation-rate associations were assessed by univariate and multivariable regression with FDR correction, and functional consequences predicted using PolyPhen-2, SIFT, SWISS-MODEL and molecular dock

Pressure-Regulated Enzyme-Equivalent Strontium Cobalt Oxide "Perovzymes" with Phase-Dependent Activity.

Enzymes exhibit remarkable structural adaptability, often exhibiting structure-dependent catalytic performance. Despite the emergence of perovskite-based inorganic oxides as promising enzyme-like catalysts, the effect of crystal structure modulation on their catalytic activity remains unknown. In this context, we demonstrate the SrCoO3-δ (δ = 0.3-0.5) perovskite system as an enzyme mimic that possesses a rich phase landscape spanning hexagonal to cubic structures. Herein, we investigated the effect of pressure-controlled phase transformations between the oxygen-deficient orthorhombic phase (SC

Development of Indole-3-yl-methylene-thiobarbital Derivatives as Inhibitors of HDAC8 Enzyme Activity.

Histone deacetylase 8 (HDAC8), a zinc-dependent class I enzyme, plays key roles in regulating gene expression and is implicated in various malignancies. Although hydroxamic acid-based inhibitors, like PCI-34051, have demonstrated potent HDAC8 inhibition, limitations including poor selectivity, instability, and off-target effects underscore the need for novel scaffolds. Here, we designed and synthesized a series of indole-based (thio)barbiturate derivatives to identify selective HDAC8 inhibitors. Compound 3d showed promising potency (IC50 = 2.9 μM) and selectivity for HDAC8 over other isoforms.

Activity-Based Ubiquitin-α-amidomethyl Acrylate Probe for Profiling of Ubiquitinating Enzymes and Deubiquitinases.

Activity-based ubiquitin probes have emerged as powerful tools for dissecting protein ubiquitination. In this work, we discovered that an α-amidomethyl-substituted acrylate electrophile exhibits markedly enhanced reactivity. Based on this finding, we designed and synthesized a new activity-based probe, ubiquitin-α-amidomethyl acrylate (Ub-AMA). In vitro biochemical assays demonstrated that Ub-AMA effectively crosslinked with deubiquitinases, E1 and E3s. In addition, compared to the classic Ub-VME and Ub-PA probes, Ub-AMA achieves improved labeling efficiency for E2 enzymes. Finally, proteomic

Design, Synthesis, and Evaluation of Alkoxyamine Anticancer Prodrugs Bearing Galactoside as an Enzyme-Activated Trigger.

Enzyme-induced cytotoxicity can be considered the main challenge in modern cancer therapy that provides unique opportunities for targeted apoptosis. To address this challenge, we implemented enzyme-triggered activation of alkoxyamine-galactose conjugates with the formation of active carbon-centered radical species for the apoptosis of cancer cells. The comparative analysis of two alkoxyamine derivatives, bearing stable and self-immolative linkers between the radical precursor and galactose moiety, revealed the crucial role of molecular structure in anticancer activity. The study of cytotoxicit

Enzyme-Loaded Liposomal Edible Hydrogel Films to Enhance Lactase Activity in Perline Mozzarella.

Lactase enzyme-based products experience challenges including residual lactose that result in lactose intolerance. The purpose of this study was to develop polyelectrolyte polysaccharide-enriched lactase-encapsulated liposomal hydrogel films as an edible coating of Perline Mozzarella cheese that delivers enzymes along with the product on the side of absorption in the small intestine. Coatings were investigated for shelf-life enhancement and in vitro enzyme release behaviour. Two different polymeric hydrogel film formulations were evaluated: lactase-encapsulated liposome-enriched chitosan (PCLL

Platinum nanozymes system with tunable enzyme activity for on-demand supply therapy of infected wounds.

The rapid healing of bacterially infected wounds remains a major clinical challenge. The pathological microenvironment-characterized by bacterial infection, reactive oxygen species (ROS) accumulation, persistent inflammation, and impaired tissue repair-severely impedes this process. In this work, we constructed a photothermally triggered, microenvironmental pH-regulating platinum nanozyme system, ACC@LPDAPt nanoparticles (NPs), leveraging the pH-dependent peroxidase-like and catalase-like activities of platinum nanozymes. This system achieves enzyme-like activity conversion by modulating the w
