Impaired pentose phosphate shunt function in sickle cell disease: a potential mechanism for increased Heinz body formation and membrane lipid peroxidation.

The red cells' antioxidant defense mechanisms were compared between individuals with sickle cell disease and those with hemolytic anemia and reticulocytosis. In sickle cell disease, there was a significant increase in incubated Heinz body formation (p less than .001), a decrease in reduced glutathione concentration (p less than .01), an increase in glucose-6-phosphate dehydrogenase activity (p less than .01), and a decrease in glutathione reductase activity (p less than .005). The patients with sickle cell disease hd an absolute increase in the activity of the pentose shunt in the intact red c

Flow cytometric enumeration of parasitaemia and haematologic changes in trypanosoma-infected mice.

African trypanosomiasis is a severe parasitic disease affecting both man and livestock. It is crucial to expand our fundamental knowledge of the intimate interactions between trypanosomes and their vertebrate hosts in order to develop new and efficient control strategies. The mouse model of trypanosomiasis is the most popular for research purposes because of all the logistic advantages of using this species. Studies of any aspect of trypanosomiases in the mouse systematically require the quantification of some phenotypic traits which translate its degree of resistance/susceptibility to the dis

Hematologic data of selenium-deficient and selenium-supplemented rats.

Effect of dietary selenium as sodium selenite on in vivo hematological parameters of Sprague-Dawley rats was examined over a 7-month period. Dietary selenium did not alter total hemoglobin, hematocrits, erythrocyte counts, or the osmotic fragility pattern of rat blood. Selenium-excessive (1.0 ppm) rats showed slightly lower but not significantly lower methemoglobin levels than selenium-adequate (0.1 ppm) or selenium-deficient rats. Platelet counts tended to be higher in selenium-excessive rats and lower in selenium-deficient rats than in selenium-adequate rats, but the differences were not sta…

Laboratory Diagnosis of sickling hemoglobinopathies.

Sickle cell trait is present in about 8% of black Americans, and clinically significant sickling disorders are common in this population. These disorders can be accurately defined by combinations of quantitative hemoglobin electrophoresis at alkaline pH, citrate agar electrophoresis, solubility tests for sickle hemoglobin, fetal hemoglobin measurements, blood counts, erythrocyte indices and family studies. Unusual types of sickling hemoglobinopathies may require more extensive, specialized study. An unquestioned diagnosis should be prerequisite for any subsequent genetic counseling.

The clinical course of mesangial IgA associated nephropathy in adults.

Clinical data in 244 patients with IgA nephropathy and biopsy findings in 519 biopsies (107 patients had at least two biopsies) were analysed. Males predominated (73 per cent) and had more severe disease and a worse prognosis than females. The most frequent symptom was macroscopic haematuria, often with associated loin pain; however, this was typical only in young males. Hypertension was the major presenting feature in 23 per cent of patients. Urinary erythrocyte counts correlated with the presence of crescents on biopsy (p less than 0.0001). Serum IgA levels wer elevated in only 21 per cent, …

Erythropoietin and Hematological Responses in Recreational Scuba Divers: Insight Into the Hyperoxic-Hypoxic Paradox.

PURPOSE: During scuba diving, breathing under elevated pressure leads to hyperoxia, which may induce oxidative stress and erythrocyte damage. Hyperoxia may also influence erythropoietin (EPO) production, a phenomenon referred to as the hyperoxic-hypoxic paradox. This study investigated changes in EPO levels and red blood cell (RBC) parameters after 1) a single dive, 2) three successive dives over 2 d, 3) the summer diving season. METHODS: 1) Divers ( N = 23) completed a single 30-min dive to 30 m. Blood samples were collected immediately pre- and post-dive, and 3 h post-dive. 2) Divers ( N =

The effects of blood transfusion on red blood cell distribution width in critically ill patients: a pilot study.

BACKGROUND: Red blood cell distribution width (RDW) is a measure of anisocytosis, generally used in the differential diagnosis of anemia. Recently, RDW was associated with increased mortality in critically ill patients. Red blood cell (RBC) transfusions are potential confounders on RDW values interpretation. The aim of this study was to analyze the changes in RDW after RBC transfusion in intensive care unit (ICU) patients. STUDY DESIGN AND METHODS: This was a prospective, observational study including patients admitted to ICU requiring 1 RBC unit. We analyzed RDW values of the patients at fou

Minimizing red blood cell contamination while isolating mononuclear cells from whole blood: the next step for the treatment of severe hemolytic disease of the fetus/newborn.

OBJECTIVE: We examined whether mononuclear cell purification of human blood could be done while minimizing contamination with RhD-positive red blood cells to treat hemolytic disease of the fetus/newborn that was caused by rhesus disease. STUDY DESIGN: Whole blood from 16 individuals who tested rhesus positive was diluted and centrifuged over a Ficoll gradient. The cell pellet was incubated with red blood cell lysis buffer, divided into three samples, and analyzed for cell count, mononuclear cell yield, and RhD-positive red cell contamination by flow cytometry. RESULTS: Mean RhD-positive red

Clinical signs of anemia in vitamin A-deficient rats.

Young rats weighing 150 g (initial weight) were fed diets sufficient or deficient in vitamin A. Postweaning rats were used in order to retard the rapid onset of vitamin A deficiency. The effects of the deficiency were studied with respect to impairment of hematopoietic function and anemia. Values for hemoglobin and hematocrit provided evidence of anemia before the signs of severe vitamin A deficiency became apparent. These included alopecia, ocular lesions, and low levels of retinol in plasma and liver. At the point where liver stores of vitamin A were virtually depleted, however, estimates fo

Red cell life span after splenectomy in hereditary spherocytosis.

Despite the persistence of spherocytosis after splenectomy in hereditary spherocytosis, it has usually been assumed that red cell life span returns completely to normal after this treatment. Diisopropyl fluorophosphate. DF(32)P, a noneluting red cell label, was given intravenously to 11 patients in five unrelated families 2-27 yr after splenectomy for typical hereditary spherocytosis. Hemoglobin ranged from 14.0 to 19.8 g/100 ml in this group and reticulocytes from 1.1 to 2.9%, showing the excellent clinical response to splenectomy. Loss of red cell radioactivity corrected for radiophosphorus

CSF from a puppy with a cerebral vascular hamartoma.

A 9-week-old puppy with refractory seizures and a dome-shaped head presented to the Mississippi State College of Veterinary Medicine Specialty Center for suspected hydrocephalus. Computerized tomography (CT) findings included transtentorial herniation and an intra-axial mass with dystrophic mineralization. Cerebrospinal fluid analysis revealed an increased nucleated cell count of 1100/μl (RI < 5/μl), erythrocyte count of 2.2 × 106 /μl, and markedly increased microprotein of 1939 mg/dl (RI < 30 mg/dl). On cytologic examination of the CSF, numerous erythrophagocytic, and hemosiderin-laden macrop…

Effect of starving and feeding on some haematological and physiological responses of the Nile catfish, Clarias gariepinus exposed to copper at extreme seasons.

The lethal concentration for 50% of fish for 96h (96h LC(50)) of copper (Cu(2+)) was estimated for the Nile catfish (Clarias gariepinus) in extreme seasons, winter and summer, 4.31 and 4.79 mg/l, respectively. The Nile catfish was exposed to 96h LC(50) of copper for 7 days in extreme winter and summer. The body indices, haematological parameters as well as some plasma and liver enzyme activities and metabolite level were significantly differed in fish exposed to copper over than those of the control fish. Most of the tested parameters were not significantly different between the control fish o

Reticulocytes in peripheral blood of mice infected with Trypanosoma brucei: dramatic decrease of reticulocytes during primary wave of parasitemia.

Infection of mice with rapidly proliferating trypanosomes that killed the mice in the first or second wave of parasitemia led to a dramatic decrease of immature erythrocytes in the peripheral blood during the primary wave of parasitemia. Trypanosome variants producing lower first waves of parasitemia, and/or less rapid deaths, caused a variable, on the average small, decrease of immature erythrocytes. Trypanosome variants causing rapidly lethal infections killed the mice before a decrease of immature erythrocytes became apparent.
