Mammillary body regulates state-dependent fear by alternating cortical oscillations.

State-dependent memory describes a phenomenon that memory will be efficiently retrieved only when the brain state during retrieval matches the state during encoding. While a variety of psychoactive drugs, such as ethanol, cocaine, morphine and NMDA receptor antagonists, are able to induce state-dependent memory, the biological hallmark of brain state and neural mechanism of its regulation are still unknown. In this study, we found that MK-801 enhanced delta oscillations in awake mice, representing a drug-induced brain state, in which fear memory could only be successfully retrieved when the sa…

Instilling fear makes good business sense: unwarranted hysterectomies in Karnataka.

A four-parameter Boltzmann empirical model was evaluated as an alternative to the classical Logistic-Luedeking-Piret (L-LP) kinetic framework for describing batch fermentation of poly-γ-glutamic acid (γ-PGA) by Bacillus subtilis natto. To characterise the classical model's failure to reproduce the post-peak decline in γ-PGA concentration observed under carbon-depleted conditions, a first-order product degradation term was incorporated into the L-LP framework as a diagnostic extension. Across 36 h of batch fermentation, bacterial growth followed a typical sigmoidal curve, reaching a maximum spe

Glucocorticoid Administration Improves Aberrant Fear-Processing Networks in Spider Phobia.

Glucocorticoids reduce phobic fear in patients with anxiety disorders. Previous studies have shown that fear-related activation of the amygdala can be mediated through the visual cortical pathway, which includes the fusiform gyrus, or through other pathways. However, it is not clear which of the pathways that activate the amygdala is responsible for the pathophysiology of a specific phobia and how glucocorticoid treatment alleviates fear processing in these neural networks. We recorded the brain activity with functional magnetic resonance imaging in patients with spider phobia, who received ei…

Fear avoidance and pain catastrophizing among women with sexual pain.

AIM: Among women under 30 years of age, around 20% report recurrent sexual pain. Although the amount of studies investigating sexual pain has increased, little is known about the role of psychosocial factors. The present study used the fear-avoidance model from musculoskeletal pain as a theoretical frame of reference, and examined fear-avoidance beliefs, avoidance behaviors, pain catastrophizing and symptoms of anxiety and depression among women (18-35 years) reporting sexual pain. METHODS: The study was cross-sectional, based on a sample of 133 women, where 37 women (27.8%) reported sexual p…

Factors associated with fear-avoidance beliefs about low back pain.

BACKGROUND: Low back pain (LBP) is a common major health problem. Fear-avoidance beliefs are thought to be an important risk factor for chronic LBP. This study investigated factors associated with the Fear-Avoidance Beliefs Questionnaire (FABQ) scores of Japanese adults who experienced LBP. We focused on the association of FABQ scores with medical advice to rest and the experience of seeing family members and/or significant others with disabling LBP. METHODS: In February 2011, 1,063,083 adults aged 20-79 years who had registered as internet-based research volunteers were randomly selected and…

Dopaminergic mechanisms underlying catalepsy, fear and anxiety: do they interact?

Haloperidol is a dopamine D2 receptor antagonist that induces catalepsy when systemically administered to rodents. The haloperidol-induced catalepsy is a state of akinesia and rigidity very similar to that seen in Parkinson's disease. There exists great interest in knowing whether or not some degree of emotionality underlies catalepsy. If so, what kind of emotional distress would permeate such motor disturbance? This study is an attempt to shed some light on this issue through an analysis of ultrasound vocalizations (USVs) of 22 kHz, open-field test, and contextual conditioned fear in rats wit…

Neuropeptide S receptor gene: fear-specific modulations of prefrontal activation.

Since central administration of neuropeptide S (NPS) has been shown to exert anxiolytic effects on rodent behavior in a number of studies, genetic variants of its cognate G-protein coupled receptor (NPSR1) became the focus of several recent human studies on anxiety and anxiety disorders. The T allele of rs324981, which goes along with enhanced receptor function, was associated with panic disorder, increased anxiety sensitivity in healthy subjects, attenuated prefrontal brain activation and elevated amygdala responses to fear-relevant stimuli. To investigate whether prefrontal attenuations in r…

Neural mechanisms of impaired fear inhibition in posttraumatic stress disorder.

Posttraumatic stress disorder (PTSD) can develop in some individuals who are exposed to an event that causes extreme fear, horror, or helplessness (APA, 1994). PTSD is a complex and heterogeneous disorder, which is often co-morbid with depression, substance abuse, and anxiety disorders such as panic or social phobia. Given this complexity, progress in the field can be greatly enhanced by focusing on phenotypes that are more proximal to the neurobiology of the disorder. Such neurobiological intermediate phenotypes can provide investigative tools to increase our understanding of the roots of the…

NMDA receptors and fear extinction: implications for cognitive behavioral therapy.

Based primarily on studies that employ Pavlovian fear conditioning, extinction of conditioned fear has been found to be mediated by N-methyi-D-aspartate (NMDA) receptors in the amygdala and medial prefrontal cortex. This led to the discovery that an NMDA partial agonist, D-cycloserine, could facilitate fear extinction when given systemically or locally into the amygdala. Because many forms of cognitive behavioral therapy depend on fear extinction, this led to the successful use of D-cycloserine as an adjunct to psychotherapy in patients with so-called simple phobias (fear of heights), social p…

Learning to fear obstructed breathing: comparing interoceptive and exteroceptive cues.

The present study investigated interoceptive fear conditioning (IFC) to an interoceptive and exteroceptive conditional stimulus (CS) with a severe respiratory load applied for 30s as the unconditional stimulus (US). CSs were another, weak respiratory load in the intero-IFC study (N=74), and a neutral picture in the extero-IFC study (N=42). CSs preceded the US in the paired groups, whereas the unpaired groups received the same number of unpaired CSs and USs. We measured startle blink EMG, self-reported fear and respiration. In the intero-IFC study, the CS-load was associated with larger startle…

Temporal factors control hippocampal contributions to fear renewal after extinction.

Fear can be extinguished by repeated exposure to a cue that signals threat. However, extinction does not erase fear, as an extinguished cue presented in a context distinct from that of extinction results in renewed fear of that cue. The hippocampus, which is involved in the formation of contextual representations, is a natural candidate structure for investigations into the neural circuitry underlying fear renewal. Thus far, studies examining the necessity of the hippocampus for fear renewal have produced mixed results. We isolated the conditions under which the hippocampus may be required for…

Policing drug users in Russia: risk, fear, and structural violence.

We undertook qualitative interviews with 209 injecting drug users (IDUs) (primarily heroin) in three Russian cities: Moscow, Barnaul, and Volgograd. We explored IDU's accounts of HIV and health risk. Policing practices and how these violate health and self, emerged as a primary theme. Findings show that policing practices violate health and rights directly, but also indirectly, through the reproduction of social suffering. Extrajudicial policing practices produce fear and terror in the day-to-day lives of drug injectors, and ranged from the mundane (arrest without legal justification; the plan…

Contextual fear conditioning differs for infant, adolescent, and adult rats.

Contextual fear conditioning was tested in infant, adolescent, and adult rats in terms of Pavlovian-conditioned suppression. When a discrete auditory-conditioned stimulus (CS) was paired with footshock (unconditioned stimulus, US) within the largely olfactory context, infants and adolescents conditioned to the context with substantial effectiveness, but adult rats did not. When unpaired presentations of the CS and US occurred within the context, contextual fear conditioning was strong for adults, weak for infants, but about as strong for adolescents as when pairings of CS and US occurred in th…

Magnesium deficiency reduces fear-induced conditional lick suppression in mice.

The consequences of broad-scale alterations in magnesium (Mg2+) levels on learning and memory are poorly understood. We have recently demonstrated that adult male mice maintained on an Mg2+-deficient diet exhibit reduced conditional freezing behavior. The purpose of the present study was to determine if the detrimental effect of Mg2+ deficiency in mice extended to another measure of conditional fear, conditioned lick suppression (CLS), as well as to another form of learning, spatial learning in the swim maze task. Adult male C57Bl/6J mice were provided with a normal or Mg2+-deficient diet and …
