Transcription factor activity divergence across muscle atrophy conditions: A comparative analysis of spaceflight, aging, and disuse using decoupleR.

Skeletal muscle atrophy arises from heterogeneous insults including spaceflight, aging, and immobilization. Prior studies have catalogued differentially expressed genes (DEGs) in each context, but the transcription factors (TFs) coordinating these programs remain inconsistently characterized across conditions, and gene-level overlap between conditions is small. Three mouse transcriptomic datasets were analyzed: NASA OSD-576 (Rodent Research-23) tibialis anterior muscle from spaceflight versus ground control animals; GSE145480 gastrocnemius muscle from 28-month-old versus 8-month-old mice as a

Integrating Single-Sample Networks and Genetic Algorithm to Estimate Radiation Dose in Multiple Tissues of Mice under Space Environment.

Exposure to space radiation poses various health risks, so accurately estimating radiation dose in space is crucial. Herein, we integrated 301 transcriptomic profiles from 30 spaceflight datasets and developed radiation-dose estimation models for the space environment using a genetic algorithm. Two models were constructed in this work: one using gene expression fold changes as input (fold change model) and the other using gene degrees as input (degree model). Of note, we initially constructed a single sample network (SSN) for each spaceflight sample, respectively, and the degrees that represen

Treatment with a superoxide dismutase mimetic for joint preservation during 35 and 75 days in orbit aboard the international space station, and after 120 days recovery on Earth.

Reduced weight-bearing during spaceflight has been associated with musculoskeletal degradation that risks astronaut health and performance in transit and upon reaching deep space destinations. Previous rodent experiments aboard the international space station (ISS) have identified that the spaceflight-induced molecular arthritic phenotype was characterized with an increase in oxidative stress. This study evaluated if treatment with a superoxide dismutase (SOD) mimetic on orbit could prevent spaceflight-induced damage to the knee and hip articular cartilage, and the menisci in rodents. Impact of Spaceflight and Artificial Gravity on the Mouse Retina: Biochemical and Proteomic Analysis.

Astronauts are reported to have experienced some impairment in visual acuity during their mission on the International Space Station (ISS) and after they returned to Earth. There is emerging evidence that changes in vision may involve alterations in ocular structure and function. To investigate possible mechanisms, changes in protein expression profiles and oxidative stress-associated apoptosis were examined in mouse ocular tissue after spaceflight. Nine-week-old male C57BL/6 mice (n = 12) were launched from the Kennedy Space Center on a SpaceX rocket to the ISS for a 35-day mission. The anima

NASA GeneLab Project: Bridging Space Radiation Omics with Ground Studies.

Accurate assessment of risks of long-term space missions is critical for human space exploration. It is essential to have a detailed understanding of the biological effects on humans living and working in deep space. Ionizing radiation from galactic cosmic rays (GCR) is a major health risk factor for astronauts on extended missions outside the protective effects of the Earth's magnetic field. Currently, there are gaps in our knowledge of the health risks associated with chronic low-dose, low-dose-rate ionizing radiation, specifically ions associated with high (H) atomic number (Z) and energy (

Stem cell health and tissue regeneration in microgravity.

Exposure to microgravity causes significant mechanical unloading of mammalian tissues, resulting in rapid alterations of their physiology, which poses a significant risk for long-duration manned spaceflight. The immediate degenerative effects of spaceflight we understand best are those studied during short-term low-Earth-orbit experiments, and include rapid microgravity-adaptive bone and muscle loss, loss of cardiovascular capacity, defects in wound and bone fracture healing, and impaired immune function. Over the long-term, exposure to microgravity may cause severe deficits in mammalian stem

Spaceflight-induced alterations in cerebral artery vasoconstrictor, mechanical, and structural properties: implications for elevated cerebral perfusion and intracranial pressure.

Evidence indicates that cerebral blood flow is both increased and diminished in astronauts on return to Earth. Data from ground-based animal models simulating the effects of microgravity have shown that decrements in cerebral perfusion are associated with enhanced vasoconstriction and structural remodeling of cerebral arteries. Based on these results, the purpose of this study was to test the hypothesis that 13 d of spaceflight [Space Transportation System (STS)-135 shuttle mission] enhances myogenic vasoconstriction, increases medial wall thickness, and elicits no change in the mechanical pro

Changes in gene expression and signal transduction in microgravity.

Studies from space flights over the past three decades have demonstrated that basic physiological changes occur in humans during space flight. These changes include cephalic fluid shifts, loss of fluid and electrolytes, loss of muscle mass, space motion sickness, anemia, reduced immune response, and loss of calcium and mineralized bone. The cause of most of these manifestations is not known and until recently, the general approach was to investigate general systemic changes, not basic cellular responses to microgravity. This laboratory has recently studied gene growth and activation of normal

A direct approach to the study of the effect of gravity on axis formation in birds.

A system has been developed to enable the normal development of aborted very early uterine avian embryos, outside the female's uterus. The shell-less aborted egg was put into a foster shell of a sister egg, previously laid by the same female. The empty space between the shell and aborted egg was filled with artificial uterine fluid. The reconstructed eggs were incubated at 42 degrees C for 30 hours in a vertical position. The atmosphere contained a high concentration of CO2 (8-10%). At the termination of the 30 h the eggs were transferred to incubation at 37 degrees C in normal atmospheric con

Effects of orthostatic and anti-orthostatic stress on patent and stenotic coronary arteries in swine.

Head-up tilt (HUT) followed by head-down tilt (HDT) has been used to simulate the acute phase of adaptation to microgravity. This study evaluates the effects of HUT and HDT on the coronary circulation before and during coronary stenosis. Seven pigs were placed in the prone position and exposed to the following orientations for 20 min each: 1) 0 degrees horizontal (HZ); 2) +70 degrees HUT; and 3) -15 degrees HDT. The swine were then placed in the HZ position for 20 min while hemodynamics returned to baseline. The diameter of the left anterior descending (LAD) coronary artery was reduced to a po

Carriage of antimicrobial-resistant E. coli in urban stray cats (Felis catus).

Antimicrobial resistance (AMR) in Escherichia coli is a growing concern in both human and veterinary medicine, driven by the inappropriate use of antimicrobials. Companion animals may act as community reservoirs and sentinels for antimicrobial-resistant bacteria due to their close contact with humans and other animals. However, resistance carriage in urban stray cats is poorly defined, particularly as many have limited direct antimicrobial exposure. This population provides an opportunity to assess baseline antimicrobial resistance carriage in companion animals with relatively low selection pr

Integrating multi-wet laboratory diagnostics to study staphylococci in animals in Uganda.

BACKGROUND: Several diagnostic environments in Uganda lack real-time, robust and high-throughput technologies for comprehensive typing of microbes, which is a setback to infectious disease surveillance. This study combined various wet laboratory diagnostics to understand the epidemiology of pathogenic staphylococci isolated from animals in Uganda and the implications for global health security priorities. METHODS: A retrospective study was conducted employing records and pathogenic staphylococci (from animals) archived at the Central Diagnostic Laboratory (CDL), Makerere University, Uganda, b

Synergism of IP3R and Parkin mutants identifies mitochondrial stress as an early feature of Parkinson's disease.

Our understanding of mechanisms underlying familial Parkinson's disease (PD) have benefitted from studies in Drosophila models of PD. However, in a majority of patients with PD, the disease occurs sporadically, and cellular phenotypes that arise early in sporadic PD are not yet fully understood. A genetic predisposition, arising from variants in pathways that impact dopaminergic neuron health could be one cause of sporadic PD. Here, we studied Drosophila with single-copy mutation of the recessive IP3R-encoding gene (itpr) in combination with a recessive null mutation of the parkin gene. Wherea

Female-specific flightless phenotype for mosquito control.

Dengue and dengue hemorrhagic fever are increasing public health problems with an estimated 50-100 million new infections each year. Aedes aegypti is the major vector of dengue viruses in its range and control of this mosquito would reduce significantly human morbidity and mortality. Present mosquito control methods are not sufficiently effective and new approaches are needed urgently. A "sterile-male-release" strategy based on the release of mosquitoes carrying a conditional dominant lethal gene is an attractive new control methodology. Transgenic strains of Aedes aegypti were engineered to h

Establishment of Circadian Rhythm Experiments using Bengalese Finches.

Marked increases in sleep deprivation and circadian rhythm disruption adversely affect human health and the quality of life. In particular, the expansion of nighttime lighting associated with a 24-h society and digital technology has promoted late-night activity in mothers and children. Studies have examined the effects of circadian disruption on brain development in chickens, which are diurnal animals with genomic similarities to humans; however, adult chickens lack post-hatching parental care, which limits their utility in analyses of parent-offspring interactions. In this study, we investig

Interaction of compass sensing and object-motion detection in the locust central complex.

Goal-directed behavior is often complicated by unpredictable events, such as the appearance of a predator during directed locomotion. This situation requires adaptive responses like evasive maneuvers followed by subsequent reorientation and course correction. Here we study the possible neural underpinnings of such a situation in an insect, the desert locust. As in other insects, its sense of spatial orientation strongly relies on the central complex, a group of midline brain neuropils. The central complex houses sky compass cells that signal the polarization plane of skylight and thus indicate

Psychomotor impairments and therapeutic implications revealed by a mutation associated with infantile Parkinsonism-Dystonia.

Parkinson disease (PD) is a progressive, neurodegenerative disorder affecting over 6.1 million people worldwide. Although the cause of PD remains unclear, studies of highly penetrant mutations identified in early-onset familial parkinsonism have contributed to our understanding of the molecular mechanisms underlying disease pathology. Dopamine (DA) transporter (DAT) deficiency syndrome (DTDS) is a distinct type of infantile parkinsonism-dystonia that shares key clinical features with PD, including motor deficits (progressive bradykinesia, tremor, hypomimia) and altered DA neurotransmission.
