Comparative analysis of different artificial neural networks for predicting and optimizing in vitro seed germination and sterilization of petunia.

The process of optimizing in vitro seed sterilization and germination is a complicated task since this process is influenced by interactions of many factors (e.g., genotype, disinfectants, pH of the media, temperature, light, immersion time). This study investigated the role of various types and concentrations of disinfectants (i.e., NaOCl, Ca(ClO)2, HgCl2, H2O2, NWCN-Fe, MWCNT) as well as immersion time in successful in vitro seed sterilization and germination of petunia. Also, the utility of three artificial neural networks (ANNs) (e.g., multilayer perceptron (MLP), radial basis function (RB

Imputation methods to improve inference in SNP association studies.

Missing single nucleotide polymorphisms (SNPs) are quite common in genetic association studies. Subjects with missing SNPs are often discarded in analyses, which may seriously undermine the inference of SNP-disease association. In this article, we develop two haplotype-based imputation approaches and one tree-based imputation approach for association studies. The emphasis is to evaluate the impact of imputation on parameter estimation, compared to the standard practice of ignoring missing data. Haplotype-based approaches build on haplotype reconstruction by the expectation-maximization (EM) al

An algorithm-enhanced stool DNA system improves the differential diagnosis of colorectal cancer versus Crohn's disease in high-risk symptomatic patients.

BACKGROUND AND OBJECTIVE: Crohn's disease (CD) and colorectal cancer (CRC) share many clinical symptoms, making non-invasive differential diagnosis difficult. FIT-sDNA is sensitive for CRC screening in average-risk populations but often gives false positives in CD patients due to inflammation-induced mucosal turnover. This study aimed to develop and validate an algorithm-enhanced system (FIT-sDNA-CA) to improve the specificity of CRC triage using current DNA tests. METHODS: The study enrolled 312 subjects, comprising a training cohort of 234 confirmed patients and a prospective validation coh

Interplay of PRMTs and Identification of Biomarkers Through Machine Learning Algorithms in Pan-Cancer, Highlighting PRMT3 as a Biomarker in Pancreatic Cancer.

Protein arginine methylation was a common post-translational modification, playing a key role in many biological processes and disease. But the regulatory mechanisms of protein arginine methyltransferases (PRMTs) in cancer were not well understood. This study aimed to examine the influence of PRMTs across various cancers and identify potential PRMTs-related prognostic biomarkers. Three machine learning algorithms were employed to identify potential PRMTs biomarkers across cancers. Additionally, drug prediction and molecular docking studies were carried out to discover new therapeutic targets.

Deep generative spatiotemporal learning for integrating fracture mechanics in composite materials: inverse design, discovery, and optimization.

The trade-off between strength and toughness presents a fundamental challenge in engineering material design. Composite materials (CMs) can strategically arrange different materials to enhance both strength and toughness by optimizing the distribution of loads and increasing resistance to crack propagation. However, current data-driven computational modeling approaches for CM configuration optimization suffer from limitations of "substantial computational cost" and "poor predictive power over extrapolation spaces", making it difficult to integrate with global optimization algorithms, and ultim

Effectiveness of mesh hernioplasty in incarcerated inguinal hernias.

OBJECTIVES: Psoriatic arthritis (PsA) arises in roughly 20-30% of individuals with psoriasis (PsO); notably, >80% of PsA cases are preceded by PsO after a clinically silent interval of immunogenetic priming and subclinical synovio-entheseal inflammation. Identification of this at-risk and subclinical window is hindered by absent staging criteria, validated biomarkers, and standardised surveillance, limiting opportunities for early intervention to prevent irreversible joint damage. METHODS: We conducted a structured narrative review of PubMed (through June 2025) targeting studies on: 1. immuno

Detection of giant cytoplasmic inclusions in a pediatric patient with recurrent infections: a case report.

OBJECTIVES: Giant inclusions in leukocytes is a common feature that can be observed in some infections but can be also related to rare genetic disorders such as Chédiak-Higashi syndrome (CHS). A differential diagnosis between these groups of diseases has to be performed using specific genetic tests. Clinical and pathological history is relevant for a diagnostic orientation due to the difficulty and specificity of the diagnostic confirmation. CASE PRESENATION: We present the case of a 3-years-old male patient with recurrent respiratory infections. It is important to highlight the presence of a

Arterial tortuosity in genetic arteriopathies.

PURPOSE OF REVIEW: Arterial tortuosity is emerging as a common feature in genetically mediated thoracic aortic disease that may be prognostic. This review will summarize recent literature on arterial tortuosity in the setting of genetic arteriopathies. RECENT FINDINGS: Although arterial tortuosity has been primarily described in Loeys-Dietz syndrome due to TGFBR1 and TGFBR2 mutations and in arterial tortuosity syndrome due to SLC210A mutations, recent studies that use quantitative measures of tortuosity suggest that tortuosity is present in many other genetic conditions associated with aortic

Diagnostic algorithm in men suspected with nonobstructive azoospermia.

This review focuses on the diagnostic algorithm for nonobstructive azoospermia (NOA), a significant male factor contributing to infertility. NOA, characterized by the absence of sperm in the ejaculate, requires a systematic diagnostic approach to identify reversible conditions, genetic factors, and prognosis for achieving pregnancy. The diagnostic pathway involves semen analysis and a comprehensive evaluation for hormonal deficiencies, anatomical abnormalities, and genetic factors. The importance of medical history, physical examination, endocrine evaluation, imaging, and genetic testing is em

[Embryological and genetic mechanisms of cardiac great arteries malformations].

Developmental genetics of congenital heart diseases have evolved from analysis of embryonic hearts towards molecular genetics of cardiac morphogenesis with a dynamic view of cardiac development. Ablation techniques, transgenic animal models and clonal analysis of the developing heart led to identification of different cardiac lineages and their respective roles. The mechanistic approach for great arteries anomalies has led to emerging concepts such as common embryological origin of anatomically different cardiac defects, phenotypic continuum of left heart obstructive defects, or developmental

Monte Carlo estimation of variance component models for large complex pedigrees.

Variance component models are widely used in animal and plant breeding. In human genetics, they can be used to identify, among other traits associated with the definition of disease, those that have a significant genetic component in their aetiology. In addition, they can be used in genetic counselling. Most of the methods currently proposed for estimating variance component models often involve repeated inversion of large matrices, resulting in intensive computations, large storage requirements, and numerical instability. Consequently, these methods are restricted to data on nuclear families,

A hybrid deep learning approach to solve optimal power flow problem in hybrid renewable energy systems.

The reliable operation of power systems while integrating renewable energy systems depends on Optimal Power Flow (OPF). Power systems meet the operational demands by efficiently managing the OPF. Identifying the optimal solution for the OPF problem is essential to ensure voltage stability, and minimize power loss and fuel cost when the power system is integrated with renewable energy resources. The traditional procedure to find the optimal solution utilizes nature-inspired metaheuristic optimization algorithms which exhibit performance drop in terms of high convergence rate and local optimal s

In Silico Prediction of BRCA1 and BRCA2 Variants with Conflicting Clinical Interpretation in a Cohort of Breast Cancer Patients.

Germline BRCA1/2 alteration has been linked to an increased risk of hereditary breast and ovarian cancer syndromes. As a result, genetic testing, based on NGS, allows us to identify a high number of variants of uncertain significance (VUS) or conflicting interpretation of pathogenicity (CIP) variants. The identification of CIP/VUS is often considered inconclusive and clinically not actionable for the patients' and unaffected carriers' management. In this context, their assessment and classification remain a significant challenge. The aim of the study was to investigate whether the in silico pr

Machine Learning to Predict Outcomes of Fetal Cardiac Disease: A Pilot Study.

Prediction of outcomes following a prenatal diagnosis of congenital heart disease (CHD) is challenging. Machine learning (ML) algorithms may be used to reduce clinical uncertainty and improve prognostic accuracy. We performed a pilot study to train ML algorithms to predict postnatal outcomes based on clinical data. Specific objectives were to predict (1) in utero or neonatal death, (2) high-acuity neonatal care and (3) favorable outcomes. We included all fetuses with cardiac disease at Sunnybrook Health Sciences Centre, Toronto, Canada, from 2012 to 2021. Prediction models were created using t

DNA barcoding as a valuable tool for delimiting mollusk species of the genus Biomphalaria Preston, 1910 (Gastropoda: Planorbidae).

INTRODUCTION: The genus Biomphalaria in Brazil includes 11 species and one subspecies, three of which are intermediate hosts of Schistosoma mansoni. Due to the recent evolution of this group, some species are difficult to identify based on morphological characters, making the use of genetic markers necessary for species identification. This study aimed to evaluate the use of partial sequences of the cytochrome c oxidase I (coi) gene for the identification of Biomphalaria species using phylogenetic reconstruction and species delimitation algorithms. The study tested the use of DNA barcoding tec

Identification of Transdiagnostic Childhood Externalizing Pathology Within an Electronic Medical Records Database and Application to the Analysis of Rare Copy Number Variation.

Externalizing traits and behaviors are broadly defined by impairments in self-regulation and impulse control that typically begin in childhood and adolescence. Externalizing behaviors, traits, and symptoms span a range of traditional psychiatric diagnostic categories. In this study, we sought to generate an algorithm that could reliably identify transdiagnostic childhood-onset externalizing cases and controls within a university hospital electronic health record (EHR) database. Within the Vanderbilt University Medical Center (VUMC) EHR, our algorithm identified cases with a clinician-validated
