The syncytial slime mold Physarum polycephalum exhibits cytosolic streaming, cell migration, and cell network remodeling within a single cell. It is unclear how cytoskeletal elements are spatiotemporally organized to mediate these processes. Here, we combine live and fixed imaging of Physarum F-actin to uncover organizational and dynamic signatures of the actin cytoskeleton across cell structures. We find circumferential actin filaments constrict and relax to drive peristaltic contractions in feeding tubes, while migratory fans are characterized by actin patches and comets. Our work presents u

It is widely accepted that giant cell arteritis (GCA) is a disease of the carotid branches. There are very few cases describing disseminated disease or visceral involvement. Isolated renal GCA presenting with renal failure is even more unusual. A 76-year-old male with no known medical history presented to the emergency department with three weeks of weakness, swelling, oliguria, and shortness of breath. He was started on hemodialysis for renal failure. After microscopic urinalysis revealed red blood cell casts, a kidney biopsy was performed and showed granulomatous arteritis suggestive of GCA

TRPV1 (transient receptor potential vanilloid 1) is a non-selective cation channel with high permeability to Ca2+ and is best known for its roles in sensory signaling. However, its function in immune cell biology, particularly in macrophage fusion, remains unknown. Cell fusion is a critical process in both physiological and pathological contexts, including development, tissue remodeling, and the foreign body response (FBR) to implanted biomaterials. During FBR, macrophages undergo fusion to form multinucleated foreign body giant cells (FBGCs), which contribute to implant degradation and fibrot…

Central giant cell granuloma (CGCG) is an uncommon benign intraosseous lesion of the jaws that typically affects the anterior mandible in yo … rathyroidism. Histopathological examination confirmed the diagnosis by demonstrating multinucleated giant cells within a fibrocellular stroma with hemorrhagic areas and woven bone formation. The lesion was managed by surgical enucleation, curettage, peripheral osteoplasty, and adjunctive cryotherapy. The postoperative period was uneventful, although mild ging …

Giant cell tumour of bone (GCTB) is viewed as a benign, locally aggressive primary bone tumour with metastatic potential. Current management is surgery with bone curettage or resection and systemic therapy with denosumab. Diagnosis is confirmed histologically prior to surgery, with staging for pulmonary disease, as pulmonary metastases (PM) reportedly occur in <8%. This study aimed to assess incidence, surveillance and management of PM in patients with GCTB, with histopathological review. A retrospective audit of the Oxford bone tumour registry was performed from January 2014 - October 2023. I

Synovial chondromatosis is a benign locally aggressive neoplasm that rarely affects the temporomandibular joint (TMJ). This report describes a case of synovial chondromatosis in a 34-year-old woman presenting TMJ pain and mandibular deviation with limited mouth opening. Imaging examinations revealed multiple calcified masses within the TMJ. During surgery, cartilage fragments were easily detached. On histological examination, multiple coalescent nodules of hyaline cartilage with areas of endochondral ossification and mild cell pleomorphism were observed, together with a significant giant cell

Giant cell tumors are neoplasms that usually occur in the long bones of young adults. They can rarely present in the soft tissue and may display malignant behavior. Giant cell malignancies have previously been reported as tumors primary of the uterus but are exceptionally rare. Although uncommon, it is important to consider when a neoplasm with osteoclast-like giant cells is encountered. In this report, we describe a 70-year-old female patient with an unremarkable past history who presented with an enlarged uterus. Transvaginal ultrasound demonstrated multiple fibroids and a thickened endometr…

OBJECTIVES: The objectives of this study were to evaluate the frequency and timing of sustained drug-free remission (SDFR) in patients with GCA and to identify potential predictive factors of this outcome.
METHODS: A retrospective review of all patients included in the large Spanish multicentre registry for GCA (ARTESER) with at least 2 years of follow-up was undertaken. SDFR was defined as the absence of typical signs, symptoms, or other features of active GCA for ≥12 months after discontinuation of treatment.
RESULTS: We included 872 patients. Forty-seven percent had received concomitant t

Giant cell arteritis is a systemic vasculitis that causes inflammation in medium- and large-sized blood vessels. The condition can lead to irreversible blindness if not recognised and treated promptly with high-dose steroids. Clinical manifestations typically include headache, jaw pain, fever, and fatigue. However, unusual manifestations of the disease have been reported, including pulmonary nodules, uveitis, pericarditis, and stroke. We report a case of biopsy-confirmed giant cell arteritis in a patient found to have renal cell carcinoma, exhibiting these unusual manifestations simultaneously

INTRODUCTION: Tenosynovial giant cell tumor (TGCT) is a rare soft tissue neoplasm with aggressive local growth. The disease is driven by excessive CSF1 expression in tumor cells, leading to increased recruitment of monocytes and macrophages, cytokine production, and tumor development. Targeted therapy against CSF1R is an effective treatment approach for unresectable, symptomatic TGCT. Vimseltinib, a novel, small-molecule tyrosine kinase inhibitor of CSF1R, has shown clinical efficacy in patients with TGCT.
AREAS COVERED: This paper outlines the pathogenesis and therapeutic options for TGCT, a

Tenosynovial giant cell tumors (TGCTs) are a spectrum of benign growths that can occur in both intra-articular and extra-articular locations. The pattern of involvement also varies from nodular, typically small-volume disease to extensive and diffuse synovial infiltration. Surgical resection remains the treatment of choice for most patients and resection techniques include arthroscopic, open, or a combined approach. However, TGCT can be locally aggressive and exhibit high recurrence rates even after adequate surgical removal. Improved understanding of the complex genetic and environmental fact

Tenosynovial giant cell tumors (TSGCT) are uncommon soft tissue neoplasms that can become debilitating if not diagnosed and treated promptly, particularly in advanced stages. Because of their nonspecific clinical manifestations, they are often misdiagnosed or overlooked. This case series highlights the varied clinical presentations and characteristic imaging findings of patients with TSGCT. By addressing key diagnostic challenges and pitfalls, the study aims to improve awareness and enhance diagnostic accuracy, thereby supporting timely and effective management in clinical practice.
