[Influence of anesthesia on the localization of Schistosoma mansoni. Intestinal schistosomiasis in white mice].

General anesthetics or hypnotics (ketamine, propanidid, thiopental, althesin, halothane, enflurane, ether), were tested on white mice infested with Schistosoma mansoni to study possible changes in the migration of this trematode from the mesentery to the liver. With this in mind, populations of 25 mice of the same sex and age were chosen at random; they were divided into a control group and a group submitted to an anaesthetic agent. The mice were perfused after dissection and catheterization of the abdominal aorta. A vacuum-pump was used to collect separately the parasites from the mesenteric

Metabolic alterations in the erythrocyte during blood-stage development of the malaria parasite.

BACKGROUND: Human blood cells (erythrocytes) serve as hosts for the malaria parasite Plasmodium falciparum during its 48-h intraerythrocytic developmental cycle (IDC). Established in vitro protocols allow for the study of host-parasite interactions during this phase and, in particular, high-resolution metabolomics can provide a window into host-parasite interactions that support parasite development. METHODS: Uninfected and parasite-infected erythrocyte cultures were maintained at 2% haematocrit for the duration of the IDC, while parasitaemia was maintained at 7% in the infected cultures. The…

Elucidating the functional role of the novel BdP50 protein and extracellular vesicles in the human erythrocyte infection by Babesia divergens.

Babesia divergens is a blood-borne parasite that invades, replicates within and destroys red blood cells (RBCs) during its asexual life cycle, causing babesiosis in humans and cattle. This study focuses on BdP50, a putative B. divergens glycosylphosphatidylinositol-anchored protein involved in the parasite life cycle. BdP50 is found on the surface of B. divergens invasive parasites (merozoites) as well as on extracellular vesicles (Bd-derived EVs). These EVs are secreted by parasites cultured in fresh human RBCs and, in addition to BdP50, are enriched in human and parasite proteins, including

Apicomplexan parasite, Eimeria falciformis, co-opts host tryptophan catabolism for life cycle progression in mouse.

The obligate intracellular apicomplexan parasites, e.g. Toxoplasma gondii and Plasmodium species, induce an IFNγ-driven induction of host indoleamine 2,3-dioxygenase (IDO), the first and rate-limiting enzyme of tryptophan catabolism in the kynurenine pathway. Induction of IDO1 supposedly depletes cellular levels of tryptophan in host cells, which is proposed to inhibit the in vitro growth of auxotrophic pathogens. In vivo function of IDO during infections, however, is not clear, let alone controversial. We show that Eimeria falciformis, an apicomplexan parasite infecting the mouse caecum, indu

Using scid mice to study parasitic diseases.

The interaction between parasites and their animal or human hosts is one of the most interesting and important biologic phenomena. Understanding the host-parasite interplay is essential to understanding the pathogenesis of parasitic diseases and for the development of new vaccines and chemotherapy. One approach to identifying key components of the host immune response to parasites is using immunodeficient host models. Scid mice are one of the most useful models of an immunodeficient host. Herein, we describe the use of scid mice to identify the steps in granuloma formation to parasite eggs, de

Haemoparasite infection risk in multi-host avian system: an integrated analysis.

Avian blood parasites play a crucial role in wildlife health and ecosystem dynamics, exhibiting heterogeneous spatial distribution influenced by various factors. Although factors underlying heterogeneity in infection with blood parasites have been explored in many avian hosts, their importance in the context of host species and the parasite taxon remains poorly understood, particularly in cohabiting host species. Using next-generation sequencing for parasite screening, we investigate the association between Haemoproteus, Plasmodium and Trypanosoma infections in relation to individual parameter

Unique Endomembrane Systems and Virulence in Pathogenic Protozoa.

Virulence in pathogenic protozoa is often tied to secretory processes such as the expression of adhesins on parasite surfaces or the secretion of proteases to assisted in tissue invasion and other proteins to avoid the immune system. This review is a broad overview of the endomembrane systems of pathogenic protozoa with a focus on Giardia, Trichomonas, Entamoeba, kinetoplastids, and apicomplexans. The focus is on unique features of these protozoa and how these features relate to virulence. In general, the basic elements of the endocytic and exocytic pathways are present in all protozoa. Parasites and Microbiota: Dual Interactions and Therapeutic Perspectives.

The human gut hosts a diverse and active community of bacteria that symbiotically support the physiology, metabolism, and immunity of the intestinal lining. Nevertheless, a dynamic community of parasites (helminths and protozoa) may share a habitat with gut-dwelling microbiota. Both microbiota and parasites can significantly change the physical and immunological environment of the gut, thus generating several mechanisms of interaction. Studying this field is crucial for understanding the pathogenesis of parasitic diseases. Additionally, intestinal microbiota and gut-dwelling parasites may inte

Comprehensive Analysis of Non-coding RNA Profiles of Exosome-Like Vesicles From the Protoscoleces and Hydatid Cyst Fluid of Echinococcus granulosus.

Cystic echinococcosis is a worldwide chronic zoonotic disease that threatens human health and animal husbandry. Exosome-like vesicles (ELVs) have emerged recently as mediators in the parasite-parasite intercommunication and parasite-host interactions. Exosome-like vesicles from parasites can transfer non-coding RNAs (ncRNAs) into host cells to regulate their gene expression; however, the ncRNAs profiles of the ELVs from Echinococcus granulosus remain unknown. Here, we isolated protoscolece (PSC)-ELVs and hydatid fluid (HF)-ELVs from the culture medium for E. granulosus PSCs in vitro and the HF

Influence of DNA methylation and chromatin accessibility on regulation of gene expression during Trichomonas vaginalis-host cell interaction.

UNLABELLED: Trichomonas vaginalis is a common sexually transmitted parasite that colonizes the human urogenital tract, causing infections that range from asymptomatic to highly inflammatory. As an extracellular pathogen, adherence to host epithelial cells is an important step to colonize the human host. Hence, understanding how the process of attachment to host cells is regulated remains an important goal in parasitology research and human health. T. vaginalis-host interaction is regulated by changes in gene expression, but it is still largely unknown how these changes in transcriptional profi

Abundance patterns of two Oropsylla (Ceratophyllidae: Siphonaptera) species on black-tailed prairie dog (Cynomys ludovicianus) hosts.

Behavioral, genetic, and immune variation within a host population may lead to aggregation of parasites whereby a small proportion of hosts harbor a majority of parasites. In situations where two or more parasite species infect the same host population there is the potential for interaction among parasites that could potentially influence patterns of aggregation through either competition or facilitation. We studied the occurrence and abundance patterns of two congeneric flea species on black-tailed prairie dog (Cynomys ludovicianus) hosts to test for interactions among parasite species. We li

Defining host range: host-parasite compatibility during the non-infective phase of the parasite also matters.

Host range and parasite specificity determine key epidemiological, ecological and evolutionary aspects of host-parasite interactions. Parasites are usually classified as generalists or specialists based on the number of hosts they feed on. Yet, the requirements of the various stages of a parasite may influence the suitability of a given host species. Here, we investigate the generalist nature of three common ectoparasites (the dipteran Carnus hemapterus and two species of louse flies, Pseudolynchia canariensis and Ornithophila metallica), exploiting two avian host species (the European roller …

Evaluation of a novel vaccine candidate derived from newly excysted juveniles of Fasciola hepatica in sheep.

Vaccine approaches for controlling Fasciola hepatica present a promising avenue, particularly considering increasing resistance to anthelmintic treatments and concerns over chemical residues. Targeting vaccine candidates that are expressed and secreted during the early infective stage of F. hepatica could offer an effective alternative. This approach aims to inhibit the invasion and migration of juvenile parasites, which have not yet fully developed their immune evasion mechanisms, thereby preventing parasite establishment and development in the host. In this study, we evaluated the host immun

Population dynamics and diversity of trematode infections in Bithynia siamensis goniomphalos in an irrigated area in northeast Thailand.

Several trematodes including Opisthorchis viverrini utilize Bithynia siamensis goniomphalos as a snail intermediate host in their life cycles. In order to capture a comprehensive range of host–parasite interactions and their transmission dynamic patterns, B. s. goniomphalos were sampled monthly over 4 consecutive years in an irrigated paddy-field habitat in northeast Thailand. Using a standard cercarial shedding method, a high diversity of trematodes (17 types) was recovered. Virgulate xiphidiocercariae were the most prevalent (7.84%) followed by O. viverrini (0.71%). In addition to seasonal a

Individual subject meta-analysis of parameters for Giardia duodenalis shedding in animal experimental models.

Giardia duodenalis is a zoonotic protozoan parasite with public health importance worldwide. While articles about animal model infectivity have been published for G. duodenalis, the studies have used diverse protocols and parameters to evaluate the infectivity of this protozoan parasite. Hence, the objectives of this study were to (1) conduct a meta-analysis of published literature for cyst shedding and diarrhea outcomes in animal models and (2) develop recommendations to help standardize experimental dose response studies. Results showed that, for the outcome of cyst shedding in faeces, the c

Parasite transmission in complex communities: predators and alternative hosts alter pathogenic infections in amphibians.

While often studied in isolation, host-parasite interactions are typically embedded within complex communities. Other community members, including predators and alternative hosts, can therefore alter parasite transmission (e.g., the dilution effect), yet few studies have experimentally evaluated more than one such mechanism. Here, we used data from natural wetlands to design experiments investigating how alternative hosts and predators of parasites mediate trematode (Ribeiroia ondatrae) infection in a focal amphibian host (Pseudacris regilla). In short-term predation bioassays involving mollus…

Eavesdropping Kairomone Markers of Interspecific Communication in Predatory and Parasitic Arthropods: A Review.

Interspecific chemical communication in insects is quintessential in mediating ecological interactions across trophic levels. Kairomones are semiochemicals emitted by one species (the emitter) and are inadvertently beneficial to another species (the receiver). These chemical markers are particularly critical in prey-predator or host-parasitoid relationships, such as host/prey location and selection. Host/prey bodies and their by-products emit pheromones, volatile and non-volatile cuticular compounds, and induce plant volatiles by oviposition, zoophytophagy, and herbivory, which are copious sou

Can't see the forest for the trees? Statistical considerations for disease macroecology.

Disease macroecology relies on large, complex datasets to understand the biotic and abiotic factors shaping parasite distributions and emerging infectious disease risk. These datasets span local to global host-parasite interactions and often integrate diverse host or parasite traits across evolutionary histories. Selecting appropriate statistical approaches requires first asking key questions about the study system: How well is the system understood? How important is predictor accuracy? How much bias is present in the data? We outline three broad analytical pathways commonly used in disease ma…

Uncovering host diversity and global distribution of Forcipomyia Meigen (Diptera: Ceratopogonidae) ectoparasitism on Lepidoptera larvae using citizen science records.

Parasitic interactions play a central role in shaping insect diversity; however, many host-parasite associations remain poorly documented, particularly among Diptera. Species of Forcipomyia (Microhelea) are known ectoparasites of other insects, yet information on their host associations is still fragmentary. The Fuliginosa species-group is known to be ectoparasite of larvae of Lepidoptera and Hymenoptera. In this study, we present the first global synthesis of ectoparasitism by Forcipomyia (Microhelea), Fuliginosa species-group, on caterpillars, integrating literature records with data obtaine
