SARS-CoV-2 Error Catastrophe Under Molnupiravir: Mutagenic Enhancement Enables Viral Persistence with Impaired Fitness.

Molnupiravir induces mutations that render severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) replication-competent through error catastrophe mechanisms. Previous studies showed no resistant virus emergence during prolonged molnupiravir treatment, with no resistant variants reported. However, these approaches were limited by genetic uniformity at passage initiation. To investigate viral population dynamics under enhanced genetic diversity, we employed mutagenic pre-treatment using 5-fluorouracil (5-FU) and favipiravir to generate diverse quasi-species populations before molnupiravir

Effectiveness of nucleoside analogs against Wetland virus infection.

Wetland virus (WELV), a newly identified Orthonairovirus phylogenetically related to the Crimean-Congo hemorrhagic fever virus (CCHFV), has recently been shown to cause human infections. A portion of patients infected with WELV usually present with febrile diseases, accompanied by hemorrhagic and neurological symptoms. Currently, there are no reports demonstrating effective therapeutic drugs for the treatment of WELV. In this study, we evaluated the anti-WELV efficacy of five nucleoside analogs: four clinically approved drugs-ribavirin, remdesivir, molnupiravir, and sofosbuvir; and a clinical

Efficacy and safety of arimoclomol in Niemann-Pick disease type C: Results from a double-blind, randomised, placebo-controlled, multinational phase 2/3 trial of a novel treatment.

Niemann-Pick disease type C (NPC) is a rare, genetic, progressive neurodegenerative disorder with high unmet medical need. We investigated the safety and efficacy of arimoclomol, which amplifies the heat shock response to target NPC protein misfolding and improve lysosomal function, in patients with NPC. In a 12-month, prospective, randomised, double-blind, placebo-controlled, phase 2/3 trial (ClinicalTrials.gov identifier: NCT02612129), patients (2-18 years) were randomised 2:1 to arimoclomol:placebo, stratified by miglustat use. Routine clinical care was maintained. Arimoclomol was administe

Therapeutically administered ribonucleoside analogue MK-4482/EIDD-2801 blocks SARS-CoV-2 transmission in ferrets.

The coronavirus disease 2019 (COVID-19) pandemic is having a catastrophic impact on human health1. Widespread community transmission has triggered stringent distancing measures with severe socio-economic consequences. Gaining control of the pandemic will depend on the interruption of transmission chains until vaccine-induced or naturally acquired protective herd immunity arises. However, approved antiviral treatments such as remdesivir and reconvalescent serum cannot be delivered orally2,3, making them poorly suitable for transmission control. We previously reported the development of an orall

Degradation characteristics and mechanisms of ammonium and phenol by the direct ammonia oxidation bacterium Alcaligenes faecalis AP3-14.

Direct ammonia oxidation (Dirammox) is an emerging nitrogen removal process that exhibits strong environmental adaptability, a simple flow, and can directly convert ammonium into N₂. However, no Dirammox bacterium has been reported for efficient treatment of antibiotic pharmaceutical wastewater co-contaminated with phenol and nitrogen. In this study, a simultaneous nitrogen- and phenol-removing strain, Alcaligenes faecalis AP3-14, was isolated from activated sludge. The strain harbors the key Dirammox gene cluster dnfABC, suggesting its direct ammonia oxidation potential. With sodium acetate a

Islet NO-Synthases, extracellular NO and glucose-stimulated insulin secretion: Possible impact of neuronal NO-Synthase on the pentose phosphate pathway.

The impact of islet neuronal nitric oxide synthase (nNOS) on glucose-stimulated insulin secretion (GSIS) is less understood. We investigated this issue by performing simultaneous measurements of the activity of nNOS versus inducible NOS (iNOS) in GSIS using isolated murine islets. Additionally, the significance of extracellular NO on GSIS was studied. Islets incubated at basal glucose showed modest nNOS but no iNOS activity. Glucose-induced concentration-response studies revealed an increase in both NOS activities in relation to secreted insulin. Culturing at high glucose increased both nNOS a

Transcriptional and metabolomic responses of Methylococcus capsulatus Bath to nitrogen source and temperature downshift.

Methanotrophs play a significant role in methane oxidation, because they are the only biological methane sink present in nature. The methane monooxygenase enzyme oxidizes methane or ammonia into methanol or hydroxylamine, respectively. While much is known about central carbon metabolism in methanotrophs, far less is known about nitrogen metabolism. In this study, we investigated how Methylococcus capsulatus Bath, a methane-oxidizing bacterium, responds to nitrogen source and temperature. Batch culture experiments were conducted using nitrate or ammonium as nitrogen sources at both 37°C and 42°

Human trophoblast-derived hydrogen sulfide stimulates placental artery endothelial cell angiogenesis.

Endogenous hydrogen sulfide (H2S), mainly synthesized by cystathionine β-synthase (CBS) and cystathionine γ-lyase (CTH), has been implicated in regulating placental angiogenesis; however, the underlying mechanisms are unknown. This study was to test a hypothesis that trophoblasts synthesize H2S to promote placental angiogenesis. Human choriocarcinoma-derived BeWo cells expressed both CBS and CTH proteins, while the first trimester villous trophoblast-originated HTR-8/SVneo cells expressed CTH protein only. The H2S producing ability of BeWo cells was significantly inhibited by either inhibitors

A kinetic study on the reactivity of azanone (HNO) toward its selected scavengers: Insight into its chemistry and detection.

Recently, azanone (HNO), which is the protonated one-electron reduction product of ·NO, has gained considerable attention due to its unique pharmacological effects. Although there has been much progress in understanding HNO biology and chemistry, it remains the most elusive reactive nitrogen species. Herein, we applied the competition kinetics method, based on two parallel HNO reactions with the different scavengers and molecular oxygen (kO2 = (1.8 ± 0.3) × 104 M-1 s-1), to determine the rate constants for the reactions of HNO with its selected co-reactants. The rate constants for the reaction

Role of hydrogen sulfide in the pain processing of non-diabetic and diabetic rats.

Hydrogen sulfide (H2S) is a gasotransmitter endogenously generated from the metabolism of L-cysteine by action of two main enzymes called cystathionine β-synthase (CBS) and cystathionine γ-lyase (CSE). This gas has been involved in the pain processing and insulin resistance produced during diabetes development. However, there is no evidence about its participation in the peripheral neuropathy induced by this metabolic disorder. Experimental diabetes was induced by streptozotocin (50mg/kg, i.p.) in female Wistar rats. Streptozotocin injection increased formalin-evoked flinching in diabetic rats

Strategies for the analysis of polar solvents in liquid matrixes.

Various approaches to the analysis of polar compounds in different matrixes by solid-phase microextraction (SPME) were studied. The analysis of polar analytes in nonpolar matrixes was performed with custom-made SPME fibers coated with Nafion perfluorinated resin. The sensitivity of this fiber in this type of analysis was better by 1 order of magnitude on average as compared to those of any of the commercially available fibers. The fiber was the most sensitive for the most polar of the compounds studied, i.e., methanol. Determination of methanol, ethanol, and 2-propanol in unleaded gasoline was
