Rotavirus-specific intestinal resident memory T cells induced by oral vaccination in early-life mice may contribute to protective immunity.

Resident memory T cells (Trm) are key mediators of protective immunity at barrier sites, yet their development during early life remains poorly defined. Here, we show that early-life oral vaccination with a heterologous rhesus rotavirus (RV) induces short-lived intestinal Trm that are negative by intravascular staining and express a CD44+CD69+CD103+/-P2X7R+ phenotype. Blocking lymphocyte homing with an anti-α4β7 antibody during vaccination did not alter the magnitude of RV-specific intestinal T cells or affect protection following murine RV challenge. In contrast, reduction of RV-specific intr

Enumeration, Phenotyping, and Clinical Associations of Tissue-Resident T Cells in the Ecto- and Endocervix of Women Attending a Colposcopy Clinic.

PROBLEM: Tissue-resident memory (TRM) cells represent important immune sentinels that mount rapid recall responses to pathogens and cancers. However, there are limited data in humans on genital tract TRM collected by clinically feasible sampling methods, limiting a full understanding of their role in immunity and clinical disease. METHOD OF STUDY: We used flow cytometry and single cell RNA sequencing (scRNAseq) to characterize T cells isolated from ectocervical biopsies and endocervical cytobrushes collected from women attending a colposcopy clinic in Winnipeg, Canada. RESULTS: The ectocervi

IL-7/IL-15/IL-21 cytokine-fusion scaffold generates highly functional CAR T cells enriched in long-lived T memory stem cells.

Functional persistence of chimeric antigen receptor T cells (CAR T cells) is limited by conventional CAR T cell manufacturing using anti-CD3/CD28 (αCD3/28) stimulation, which generates terminally differentiated and shorter-lived CAR T cells. We demonstrated that HCW9206, a unique protein scaffold linking interleukin-7 (IL-7), an IL-15/IL-15 receptor α (IL-15Rα) complex, and IL-21, generates CAR T cells without requiring αCD3/28 activation, which are highly enriched in long-lived T memory stem cells (TSCM cells) (>50%) and display potent activity across distinct disease models, HIV-1 or B cell

Impact of foot-and-mouth disease virus on memory T and B cell populations in swine.

Foot-and-mouth disease virus (FMDV) is a highly contagious picornavirus that poses a serious threat to the global livestock industry. This study aimed to investigate the impact of FMDV infection on the memory immune response in pigs and to analyze the role of type II interferon (IFN-γ) in this process. By comparing pigs artificially infected with FMDV and those vaccinated with inactivated FMDV vaccine, we found that FMDV infection significantly suppressed the development of memory T helper (Th) and B cell populations, affecting the memory immune response. Further experiments showed that pretre

Autoimmune and Lymphoproliferative Complications of Common Variable Immunodeficiency.

Common variable immunodeficiency (CVID) is frequently complicated by the development of autoimmune and lymphoproliferative diseases. With widespread use of immunoglobulin replacement therapy, autoimmune and lymphoproliferative complications have replaced infection as the major cause of morbidity and mortality in CVID patients. Certain CVID complications, such as bronchiectasis, are likely to be the result of immunodeficiency and are associated with infection susceptibility. However, other complications may result from immune dysregulation rather than immunocompromise. CVID patients develop aut

Glioblastoma stem cells as carriers of tumour memory: a strategy for personalised immunotherapy using tumour-infiltrating lymphocytes.

Glioblastoma (GB) remains one of the most aggressive brain tumours, with a median survival of 15 months, largely due to resistance towards available anti-cancer therapies, including cutting-edge immunotherapy. Growing evidence indicates involvement of cancer stem cells (CSCs) in escalating resistance against existing treatment modalities due to their phenotypic plasticity, elevated expression of drug-resistance pumps, and immunomodulatory behaviour. Such oncogenic consequences are regulated by several epigenetic reprogramming events that are pivotal in retaining adaptive traits associated with

Comparative analysis of cellular immune responses to four seasonal inactivated influenza vaccines in younger and older adults.

The efficacy of seasonal influenza vaccines varies across age groups, even in individuals with strong antibody responses. Since protection against influenza virus infection also involves cellular immunity, identifying immune markers beyond neutralizing antibodies is crucial for informing the design of next-generation vaccines. Participants were categorized by age, 28-60 and 65-85 yr, and were vaccinated with 1 of 4 influenza vaccines (Fluzone Standard-Dose [FSD], Flucelvax [FCEL], Fluzone High-Dose [FHD], or Fluad [FAD]) during the 2023-2024 influenza season. Blood samples were collected befor

Clonal relationships of memory B cell subsets in autoimmune mice.

Immunological memory protects our body from re-infection and it is composed of a cellular and a humoral arm. The B-cell branch with its memory B cells (MBCs), plasma cells and antibodies, formed either in a germinal centre (GC) -dependent or -independent manner, ensure that we can rapidly mount a recall immune response. Previous work in immunised wildtype (WT) mice have identified several subsets of MBCs whereas less is known under autoimmune conditions. Here, we have investigated the heterogeneity of the MBC compartment in autoimmune mouse models and examined the clonal relationships between

Identifying a Csmd3+ Microglial Subpopulation that Drives Cold-to-Hot Transition and Immune-Cure in Glioblastoma.

Glioblastoma (GBM) is immunologically cold and responds poorly to immune-based therapies owing to its highly heterogeneous and immunosuppressive tumor microenvironment (TME). However, strategies to achieve a cold-to-hot transition remain elusive, and suitable research models are still lacking. Here, TME profiling classifies our refractory G422TN-GBM model as the TMEMed (heterogeneous immune populations, "cold") subtype of human GBM, which can be shifted toward the TMEHigh (immune-high, "hot") subtype by inhibiting TGF-β signaling. In the multi-drug regimen, only αTGF-β combining temozolomide c

CD4+ tissue-resident memory Th17 cells are a major source of IL-17A in Spondyloarthritis synovial tissue.

OBJECTIVES: Interleukin (IL)-17A is a key driver of spondyloarthritis (SpA) joint pathology. We aimed to identify its cellular source in synovial tissue from patients with 2 forms of SpA, namely axial SpA (AxSpA) and psoriatic arthritis (PsA). METHODS: Synovial tissue from patients with SpA was profiled using single-cell RNA sequencing (scRNA-seq; AxSpA, n = 5; PsA, n = 6) or spatial RNA profiling (PsA, n = 4). CellPhoneDB was used to infer cell-cell communication. Tissue-resident memory Th17 (TRM17)-like cells were generated in vitro using blood memory CD4+ T cells from SpA patients. An epig

Parity and lactation induce T-cell-mediated breast cancer protection.

Parity and breastfeeding reduce the risk of breast cancer, particularly triple-negative breast cancer (TNBC)1,2, yet the immunological mechanisms underlying this protection remain unclear. Here we show that parity is associated with increased numbers of CD8+ T cells, including cells with a tissue-resident-memory-like phenotype within human normal breast tissue. In mouse models, pregnancy followed by lactation and involution drove the accumulation of CD8+ T cells in the mammary gland, coinciding with reduced tumour growth and increased intratumoural immune cell infiltration, effects that were a

Paradoxical psoriasis: The flip side of idiopathic psoriasis or an autocephalous reversible drug reaction?

Psoriasis is a common, chronic skin disease that results mainly from the complex interplay between T cells, dendritic cells, and inflammatory cytokines including TNF-α, IL-17, IL-12, and IL-23. Successful therapy with anti-cytokine antibodies has proved the importance of these key cytokines, especially TNF-α. During the anti-TNF-α treatment of classical idiopathic psoriasis, a small portion of patients develop new psoriasiform lesions. This contradictory phenomenon was named paradoxical psoriasis which resembles idiopathic psoriasis clinically but presents overlapped histological patterns and

The SATvac model of CD8+ T cell expansion and contraction phases considering memory and effector cell differentiation.

Primary immune responses induce CD8+ T cell responses characterized by activation via antigen presenting cells, expansion, differentiation into effector and memory phenotypes, and contraction resulting in long-term memory populations. In this study a mathematical stochastic agent-based model is developed to simulate all phases of the CD8+ T cell response following vaccination. Importantly, the model successfully captures the stochastic nature of T cell dynamics throughout the response. It predicts T cell population with high accuracy while addressing mouse-to-mouse variability, highlighting it

Identification of proliferating CD103+ CD8+ tissue-resident memory-like T-cells in villitis of unknown etiology in human placenta.

Villitis of unknown etiology (VUE) is a destructive inflammatory lesion of unknown cause in the human placenta. It may be caused by semiallogeneic rejection of fetal tissue by the mother. CD8+ tissue-resident memory T (TRM) cells reside in the peripheral tissues and have an important role in the local defense against reinfection. These cells also contribute to immunopathogenesis under some circumstances and cause local tissue damage and autoimmune disease. In this study, we examine the contribution of CD103+ CD8+ TRM-like cells to the development of VUE. The study included 23 cases of human pl

Single-cell analysis indicating CCR8 modulates CD8+ tissue-resident memory T cells to attenuates rejection in transplantation.

Immune rejection poses a major challenge in organ transplantation, with tissue-resident memory T (TRM) cells playing a critical role in graft rejection. This study investigated the impact of CCR8 on TRM cells using single-cell RNA sequencing (scRNA-seq), flow cytometry, and immunohistochemistry. The results show that CCR8 expression was upregulated on CD8⁺ TRM cells after transplantation, peaking on day 7. Blocking or knocking out CCR8, as well as neutralizing CCL1 and CCL8, significantly reduced CD8⁺ TRM cell accumulation in the graft and their cytokine production. These treatments prolonged

Epidermal resident memory T cell fitness requires antigen encounter in the skin.

CD8+ tissue-resident memory T cells (TRM) develop from effectors that seed peripheral tissues where they persist providing defense against subsequent challenges. TRM persistence requires autocrine TGFβ transactivated by integrins expressed on keratinocytes. TRM precursors that encounter antigen in the epidermis during development outcompete bystander TRM for TGFβ resulting in enhanced persistence. ScRNA-seq analysis of epidermal TRM revealed that local antigen experience in the skin resulted in an enhanced differentiation signature in comparison with bystanders. Upon recall, TRM displayed grea

Decoding Lupus Diagnosis, Pathogenesis and Therapy: From Systemic Autoimmunity to Renal Damage.

Systemic lupus erythematosus (SLE) is a chronic autoimmune disorder characterized by the loss of self-tolerance to nuclear and cytoplasmic antigens, triggering immune activation and tissue inflammation. Lupus nephritis (LN) is a major determinant of disease-related morbidity, disability, chronic kidney disease progression, kidney failure, and mortality in SLE, affecting approximately 30% of patients at diagnosis and up to 50-60% within the first decade. This review examines the disease's pathogenic mechanisms, emphasizing the innate immune system's role in the loss of self-tolerance and subseq

CD8+ effector memory and regulatory T cell dynamics predict subclinical atherosclerotic plaque formation in healthy aging.

BACKGROUND: Subclinical atherosclerotic plaque (SAP) develops silently in older adults. Biological changes that predict plaque formation are not well defined. We examined whether circulating immunological, metabolic, and functional measures identify vascular vulnerability before clinical disease. METHODS: In a three-year longitudinal study, 49 healthy older adults (63.8 ± 3.8 years) underwent carotid ultrasound, T cell phenotyping, serum protein profiling, body composition assessment, and fitness testing at baseline and follow-up. At follow-up, participants were classified as no SAP, new SAP
