Rhein ameliorates MASH via EGFR/AKT/PPARα-mediated coordinated regulation of metabolism and inflammation.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) presents a growing global health challenge with limited therapeutic options. Rhein, an active anthraquinone derived from the traditional medicine rhubarb (Rheum palmatum L.), has demonstrated potential in alleviating metabolic liver disorders, yet its precise mechanism of action against MASH remains unclear. METHODS: An integrated strategy combining network pharmacology, molecular docking, and experimental validation was employed. A "Rhein-MASH" target network was constructed to identify core targets. The in vivo therapeutic

Genistein ameliorates lupus nephritis via enhancing ERβ-mediated inhibition of STAT3-driven inflammation.

BACKGROUND: Systemic lupus erythematosus (SLE) is an autoimmune disease with marked female predominance. Lupus nephritis (LN), one of its most severe manifestations, is strongly associated with aberrant activation of signal transducer and activator of transcription 3 (STAT3). Estrogen receptor β (ERβ) has been reported to exert immunoregulatory effects, but its role in restraining STAT3 signaling in LN remains unclear. PURPOSE: This study aimed to investigate whether genistein, a phytoestrogen with selective affinity for ERβ, can modulate the ERβ-STAT3 axis to attenuate lupus nephritis. METH

Rapamycin treatment ameliorates HLA-B27-mediated gut inflammation and alters the microbiome in experimental spondyloarthritis.

OBJECTIVE: To determine whether rapamycin affects HLA-B27-mediated gut inflammation in experimental spondyloarthritis (SpA). METHODS: HLA-B27/human β2-microglobulin transgenic (B27-Tg) rats with gut inflammation were treated with rapamycin (1.5 mg/kg intraperitoneally, 3 times a week) or vehicle for 5 weeks. Healthy age-matched wild type (WT) rats were treated in parallel. Gut inflammation was evaluated via stool scoring and histological assessment. Transcriptome and microbiome analyses were performed on colon tissue and cecal luminal contents, respectively. Bulk immune cells were isolated fr

Piezo2 Mediates a Vicious Cycle of "Mechanical Homeostasis Imbalance-Inflammation" in Sensory Nerves and the Cartilage Endplate.

Low back pain is a global health problem. Discogenic low back pain, the most common type of low back pain, is closely related to cartilage endplate (CEP) degeneration and inflammation. In this study, by constructing a rat model of lumbar spine instability (LSI) and combining biomechanical analysis with molecular biology techniques, we revealed the central role of the mechanosensitive channel Piezo2 in the vicious cycle of discogenic low back pain. The results revealed that abnormal mechanical stress triggers calcium influx and promotes CGRP release by activating Piezo2 in the dorsal root gangl

Exosome-mediated immune modulation in rheumatoid arthritis and its role in synovial inflammation and autoimmune amplification.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent articular inflammation, cartilage destruction and aberrant synovial angiogenesis. While current cytokine-centric therapies have revolutionized RA management, their limitations highlight the need for a deeper understanding of the integrated synovial microenvironment. This review explores the critical role of exosomes as central regulatory nodes orchestrating intercellular communication within the RA joint. Under conditions of hypoxia and metabolic stress, exosomes secreted by fibroblast-like synoviocytes (FLS)

Apoptotic body-encapsulated nanoparticles regulate inflammation through macrophage polarization mediated by lactic acid.

BACKGROUND: Inflammation has been implicated in numerous diseases. The treatment of inflammation-related diseases is a significant global burden. Recent studies have found that lactic acid and its signalling pathway can inhibit inflammatory responses through macrophage polarization without any toxic side effects. OBJECTIVE: This study aimed to develop a rational drug delivery system that can release lactic acid into the desired area. METHODS: We extracted the macrophage-derived apoptotic bodies (ABs) to prepare the AB-encapsulated nanoparticles (ABNs) and evaluated the amount of lactic acid

Inflammation mediates the association of heavy metal exposures with kidney function: Evidence from a longitudinal study.

The kidney is a primary organ of heavy metal accumulation, and metal toxicity poses a great challenge to kidney health. We conducted a repeated-measure study to examine the longitudinal associations between blood heavy metal concentrations and kidney function biomarkers, including the urinary albumin-to-creatinine ratio (UACR), estimated glomerular filtration rate (eGFR), and N-acetyl-β-d-glucosaminidase (NAG). Using linear mixed models, we found that chromium (Cr) and lead (Pb) were associated with impaired kidney function across multiple biomarkers. Manganese (Mn) levels were linked to lower

Association of inflammatory bowel disease with cardiovascular disease, and the mediating role of inflammation: a matched-cohort study.

BACKGROUND: The elevated risk of cardiovascular disease (CVD) in inflammatory bowel disease (IBD) has been increasingly recognized. Although inflammation is implicated in both IBD and CVD, the IBD-CVD association through the mediation of systemic inflammation remains underexplored. OBJECTIVE: To evaluate the associations between IBD and incident CVD and examine the mediating role of inflammatory biomarkers. DESIGN: A sex- and age-matched cohort study. METHODS: Using data from the UK Biobank, the study included 1494 participants with a first-ever diagnosis of IBD from 1999 to the recruitment

Flow-mediated endothelial remodeling and inflammation drive developmental vascular susceptibility in ldlr loss of function.

Atherosclerosis, the leading cause of cardiovascular disease, is associated with aberrant lipid metabolism, endothelial dysfunction, and chronic inflammation, yet its early manifestations and mechanisms remain incompletely understood. As low-density lipoprotein receptor loss of function is the most common monogenic cause of atherosclerosis, we employed low-density lipoprotein receptor knockout (ldlr-/-) zebrafish to investigate the developmental origins of atherosclerotic cardiovascular disease. Single-cell RNA-sequencing under differential flow conditions in embryonic ldlr-/- zebrafish identi

Mast cell-expressed Mrgprb2/MRGPRX2 mediates gout pain and inflammation via a neuroimmune axis.

Acute severe joint pain is a major symptom in gouty arthritis (GA), and its adequate treatment represents an unmet medical need. Mrgprb2, a specific mast cell receptor, has been implicated in the generation of chronic pain by mobilizing mast cell degranulation, yet its significance in GA pain and joint inflammation is still not well defined. Here, we found that Mrgprb2 was expressed in mouse synovial mast cells. In a murine model of GA, acute blockade or genetic deletion of Mrgprb2 significantly attenuated arthritis pain and hyperexcitability of joint nociceptors with significant reductions in

Electroacupuncture alleviates synovial inflammation in rheumatoid arthritis rats by suppressing caspase-1-mediated pyroptosis.

Rheumatoid arthritis (RA) causes inflammatory bone and cartilage damage. While electroacupuncture (EA) alleviates joint inflammation, its anti-inflammatory mechanisms remain unclear. Sprague-Dawley rats were randomly assigned to control, collagen-induced arthritis (CIA), EA (EA+CIA), and sham-EA (sham-EA+CIA) groups. Rats in the EA group received EA at unilateral ST36 (Zusanli) every other day from day 1. Paw swelling and paw withdrawal latency (PWL) were measured during the treatment. The levels of caspase-1, GSDMD, IL-1β, and IL-18 in the synovium were assessed after 28 days of EA treatment.

scRNA sequencing revealed HIV-associated inflammation-mediated lung epithelial dysregulation and fibroblast remodeling.

BACKGROUND: HIV infection is risk factor for a wide spectrum of pulmonary diseases, with incidence rates significantly higher in people living with HIV (PLWH). Despite antiretroviral therapy, persistent immune activation and recurrent injury continue to compromise lung integrity in this population. However, the mechanism by which HIV disrupts alveolar homeostasis and affects epithelial, immune, and stromal compartments in the lungs remain undefined. OBJECTIVE: To characterize the cellular and molecular landscape of HIV infection within the lungs, the focus of the study was to assess epithelia

FABP4 Mediates Inflammation to Regulate Endometrial Epithelial Cell Function During Early Gestation in Sheep.

The objective of this experiment was to understand the impact of fatty acid binding protein 4 (FABP4)-mediated inflammation on sheep endometrial epithelial cells (sEECs). To model the hormonal environment of sheep during pregnancy, estradiol (E2), progesterone (P4) and interferon-tau (IFN-τ) were used. The sEECs were then stimulated with different concentrations of lipopolysaccharide (LPS) to determine suitable concentrations for inducing inflammation, while FABP4 expression was inhibited using a FABP4-specific inhibitor. The expression of genes related to inflammation, cell apoptosis, cell pr

Mediating Effects of Systemic Inflammation on the Association Between Body Roundness Index and Periodontitis in US Adults.

INTRODUCTION AND AIM: Periodontitis is a highly prevalent chronic inflammatory disease affecting periodontal tissues. While the Body Roundness Index (BRI), has emerged as a novel anthropometric measure for evaluating obesity-related health risks, its relationship with periodontal health remains unexplored. Although systemic inflammation is recognized as a key role in both obesity and periodontitis. However, whether BRI affects periodontitis, the mediating role of systemic inflammation in BRI-related periodontitis has not been elucidated. MATERIALS AND METHODS: Data were derived from the Natio

Systemic inflammation mediates the association between docosahexaenoic acid/docosapentaenoic acid ratio and the risk of periodontitis.

Docosapentaenoic acid (DPA_ω6) and docosahexaenoic acid (DHA) are ω6 and ω3 polyunsaturated fatty acids (PUFAs), but their association with periodontitis is controversial. We hypothesize that a novel ratio incorporating these 2 PUFAs could provide a more reliable indicator of the relationship between serum PUFAs and the risk of periodontitis. To test this hypothesis, we examined the association between the DHA-to-DPA_ω6 ratio and the risk of periodontitis and explored the potential mechanism in human subjects. Logistic regression was employed to investigate the relationship between the DHA-to-

ETS2 aggravate allergic airway inflammation by regulating ANT2-mediated cytosolic mitochondrial DsRNA levels.

BACKGROUND: ETS2 has been identified as a pivotal regulator in the development of human inflammatory diseases. Nevertheless, the functional aspects of ETS2 in asthma remain inadequately characterized. The release of mitochondrial dsRNA is recognized as an initiator of innate immune responses and implicated in intensifying inflammation triggered by alternative immunogens. The interplay between these mechanisms remains poorly understood, and only a limited number of direct targets that underpin the pro-inflammatory role of ETS2 have been identified. METHODS: The expression of ETS2 in epithelial

Cardiac Tissue Damage in a Female Animal Post-COVID Model: Relevance of Chemokine-Mediated Inflammation.

Post-COVID cardiac complications have emerged as a significant and persistent clinical concern, yet their underlying mechanisms remain poorly understood. Animal models can act as proxies to investigate the pathophysiology of the human, post-acute sequelae of SARS-CoV-2 infection (PASC). The aim of this experimental study was to evaluate the expression of inflammatory biomarkers in cardiac tissue 28 days after SARS-CoV-2 infection in a female hACE2 mouse model, with a focus on chemokine-mediated immune activation. Twelve female C57BL/6 hACE2 mice were infected with the Omicron variant (BA.1.17
